The aim of the study was to investigate the effects of sodium fluoride (NaF) and nitrate (NO₃⁻) either administered orally alone or in a combination, on the biochemical parameters of rats. Tests on general toxicity were conducted on the basis of methodical recommendations 408 of the Organisation for Economic Co-operation and Development Guideline for Testing of Chemicals: "Subchronic Oral Toxicity - Rodent: 90-d Study".The following combined effects of the tested chemicals were predominantly observed: additive, synergistic, and antagonistic. Methaemoglobinaemia was noted in groups given NO₃⁻ or NaF + NO₃⁻ complex.

Sodium fluoride (NaF) is an agent promoting bone formation around the hip implant and it can be used as an additive in poly(methyl methacrylate) (PMMA) bone cement. The influence of such an addition on the fatigue endurance of the material as used in biomedical applications was tested. Fatigue tests were performed to simulate the mode of loading that the cement mantle withstands in vivo. Tensile tests were conducted according to current standards and based on protocols being previously validated. The results obtained prove that the addition of NaF (12% in weight) to PMMA bone cement does not have any detrimental effect on the fatigue resistance of the material. This percentage is higher than that normally used for BaS04 that is added to PMMA as a radiopacifier. This latter addition was proved to reduce the fatigue resistance of the material.

Three groups of 25 female rats each were dosed ad libitum with a sodium fluoride solution containing 0 mg F/dm³, 20 mg F/dm³, and 40 mg F/dm³ in the drinking water 3 to 5 weeks before pregnancy and 3 weeks after insemination. The doses used did not produce any teratogenic effect except for the highest dose (40 mg/dm³) producing foetus abnormalities in a few cases.

The influence of four inhibitors: boric acid, thioglycolic acid, sodium fluoride and acetohydroxamic acid on the activity of urease, both in the native form and immo­bilized covalently on glutaraldehyde-pretTeated chitosan membrane, was studied. Urea hydrolysis was carried out in phosphate buffer, pH 7, at 25°C at urea concen­tration of 50 mM. The immobilized urease was more resistant than the native one to the action of all the investigated inhibitors, except boric acid. This property of the enzyme offers a possibility of its practical application.

Badano odczyn histochemiczny na aktywność dehydrogenazy mleczanowej [LDH 1.1.1.27] w wątrobach szczurów poddanych działaniu fluorku sodu (NaF). W wyniku przeprowadzonych badań stwierdzono zmiany w aktywności LDH w wątrobach szczurów po podaniu NaF z wodą do picia.

The aim of the study was to evaluate the effect of fluorine on the activity of lactate dehydrogenase (LDH) in rat hepatocytes. Wistar rats (n = 90) were used in the experiment. The animals were divided into 3 groups; the control group was drinking tap water with trace amounts of fluoride, while two other groups received fluoride at 10.6 mg NaF/dm3 and 32.0 mg NaF/dm3, respectively. The rats were exposed to fluoride for 4 months since the intrauterine life to maturity. It has been found that long-term dietary exposure to sodium fluoride leads to changed LDH activity in the rat liver and that the applied fluoride dose affects reversibility of the changes in lactate dehydrogenase activity.

Przeprowadzono badania nad wpływem fluorku sodu na parametry stresu oksydacyjnego w mitochondriach z łożysk ludzkich. Zaobserwowano, że fluorek sodu powoduje zwiększenie stężenia dialdehydu malonowego i zmniejszenie zawartości grup tiolowych białek, natomiast nie wpływa na generację rodnika hydroksylowego (•OH).

Increasing presence of fluorine compounds in the environment and the growing human exposure require continuous monitoring. The aim of the study was to assess natrium fluoride (NaF) influence on some of the oxidative stress parameters, such as: malonedialdehyde (MDA) concentration, content of protein sulfhydryl groups (SH) and the ability to generate the hydroxyl radical, (•OH). The assays were performed by spectrophotometry on mitochondria from human placenta. It was noted that NaF caused MDA level increase, SH groups content decrease, and had no influence on hydroxyl radical generation. To sum up, the mechanism of lipid peroxidation increase caused by NaF seems to be connected with generation of superoxide radical and reduction of SH groups content rather than with increased hydroxyl radical generation.

Szczurom rasy Wistar FL podawano fluorek sodu dootrzewnowe oraz w wodzie pitnej. Spowodowało to w obu doświadczeniach powstanie hiperglikemii fluorkowej, której towarzyszył spadek aktywności glukozo-6-fosfatazy wątrobowej, co sugeruje, że przyczyną hiperglikemii nie jest stymulacja szlaku glukoneogenezy.

Glucose-6-phosphatase activity in the liver of rats (Wistar FL strain males) receiving a single intraperitoneal injection of NaF at 35 mg/kg body weight, and of rats exposed for 4 months to sodium fluoride in drinking water at 50 mg/dm3 and 100 mg/dm3 (2.63 mmol/dm3 and 5.26 mmol/dm3, respectively). In both experiments, the fluoride hyperglycaemia was accompanied by reduced glucose-6- phosphatase activity. The results of this study do not support the hypothesis that hyperglycaemia developed as a result of fluoride intoxication is associated with stimulation of the gluconeogenesis pathway enzymes.

Celem pracy było zbadanie aktywności amylazy w surowicy krwi i w trzustce szczurów szczepu Wistar FL, którym podano jednorazowo, dootrzewnowo 35 mg NaF/kg masy ciała. W surowicy krwi nie stwierdzono zmian aktywności amylazy, natomiast w trzustce wykazano tendencje wzrostowe (o 23%) aktywności tego enzymu. Ze względu na fakt, że źródłem alfa-amylazy w surowicy szczura są ślinianki i wątroba, nie zaobserwowaliśmy zależności między aktywnością amylazy w surowicy krwi (brak zmian) a aktywnością amylazy oznaczanej w trzustce (tendencje wzrostowe).

The aim of this study was to examine the amylase activity in blood serum and pancreases of rats of the Wistar FL strain which had been administered intraperitoneally a single dose of 35 mg NaF per 1 kg of body mass. No changes of the amylase activity were found in the blood serum, while in the case of pancreases, upward trends (by 23%) of the enzyme activity were seen. With salivary glands and the liver being sources of alpha amylase in the rat blood serum, no relation was observed between the amylase activity in blood serum (no changes) and the amylase activity assayed in the pancreases (upward trends).

Szczurom samcom szczepu Wistar podawano przez trzy miesiące fluorek sodu o stęż. 100 mg F-/dm3 wody. Część szczurów otrzymywała dodatkowo chryzynę p.o. w dawkach 10 mg/kg m.c./24 godz. oraz 20 mg/kg m.c./24 godz. Stwierdzono, te podawanie chryzyny łagodzi zaburzenia aktywności enzymów w surowicy krwi wywołane działaniem fluorku sodu.

Male Wistar rats received sodium fluoride at 100 mg F-/dm3 water for 3 months. Some of these rats received additionally chrysin at 10 mg/kg b.w./24 h or 20 mg/kg b.w./24 h. Chrysin given to the rats was found to alleviate irregularities in the activity of blood serum enzymes resulting from the exposure to sodium fluoride.

Badano stężenie fluorków w surowicy krwi i jego zawartość w zębach (siekaczach) i kościach (udowe) szczurów szczepu Wistar FL. Wykazano wzrost zawartości F w surowicy krwi, zębach i kościach zależny od podanej dawki fluorku sodu. Stwierdzono największą zawartość F- w kościach udowych po dawce 100 ppm F- oraz wzrost stężenia glukozy w surowicy krwi szczurów zależny od podaży NaF.

We determined concentrations of fluoride ion in serum, incisor teeth, and femoral bones of Wistar FL rats. The rats received sodium fluoride at 50 or 100 ppm F- in drinking water for 4 months The concentration of F" in the femoral bones was significantly higher in the F group; a significant, NaF dose-dependent increase of glucose level in rat serum (fluoride hyperglycaemia) was also observed.

Celem pracy była ocena wpływu fluorku sodu (NaF) na obraz ultrastrukturalny wątroby szczura. Zmiany w obrazie dotyczyły w większości jąder komórkowych, mitochondriów, siateczki endoplazmatycznej szorstkiej.

The aim of the study was to evaluate the effect of fluorine on the ultrastructural picture of rat liver hepatocytes. Wistar rats received aqueous NaF solution at two concentrations: 10.6 mg NaF/L and 32.0 mg NaF/L. The animals were exposed to NaF since their intrauterine life till maturity. The rat liver was examined in the 14th, 30th, 60lh and 90th day of the fluorine-exposed life and on the 30th day after the exposure had been ceased. The ultrastructural examination procedure comprised collecting four 1 mm3 sections of the anterior lobe from each of 75 rats. Normal preparation procedures were used. The resultant preparations were examined on a OPTON 900 PC Scanning Electron Microscope. The analysis of the ultrastructural pictures of rat liver hepatocytes indicates that ultrastructural changes in the hepatocytes become intensified with increased fluorine dose. This refers to the nuclei, mitochondria, rough and smooth cytoplasmic reticulum and sinusoids. Most of these changes are reversible.

Stężenie magnezu i fosforu w surowicy krwi szczurów po zatruciu ostrym fluorkiem sodu (35 mg NaF/kg m.c.) nie zmieniało się istotnie w porównaniu z grupą kontrolną. Statystycznie znamienny spadek stężenia wapnia w surowicy krwi szczurów po zatruciu ostrym, świadczy o zaburzeniu gospodarki wapniowej. W doświadczeniu przewlekłym, po 4 mies. podawania szczurom 50 i 100 mg F- /dm3 stwierdzono spadek stężenia wapnia i magnezu po dawce 100 mg mg F- /dm3 i wzrost stężenia fosforu po dawce 50 mg F- /dm3. Uzyskane wyniki świadczą o zaburzeniu gospodarki mineralnej u badanych szczurów.

Biochemical analysis in rat serum after acute poisoning by sodium fluoride (35 mg NaF/kg body wt.) showed unchanged concetration of magnesium and phosphorus vs. control. Statistically significant decrease of calcium concetration in rat serum after acute poisoning points to disturbed calcium metabolism. In the chronic experiment, after 4 months administration of 50 and 100 mgF/dm3 we confirmed decreased calcium and magnesium concentration after exposure to 100 mg mgF/dm3, and increased phosphorus concetration after exposure to 50 mgF/dm3. Our results show disturbed mineral metabolism in studied rats.
