Extremophilic and modified aminotransferases as a versatile tool for the synthesis of optically pure building blocks for pharmaceutical industry

Considerable progress has been made in the past few years with industrial use of essential key intermediates for chemical and pharmaceutical industry. The increasing demand for obtaining chiral drugs in enantiomerically pure form makes it necessary to search for novel biocatalysts useful in the synthesis of amino acids, chiral amines, amino sugars and alcohols. According to the reaction mechanism, aminotransferases (ATs) have useful applications because of their capability of transfer of an amino group from a donor substrate to an acceptor, thus resulting in the synthesis of a wide variety of building blocks. This article reviews current biocatalytic approaches using microbial ATs in the synthesis of optically active products. Focus is also put on the engineering of ATs and their limitations in the industrial applications. Moreover this review covers biocatalytic approaches using ATs isolated from extreme environments.

Soil samples collected in 2006 from 4 different sites (Puchalski Hill and a moraine below this hill, an elephant seal colony and a penguin rookery) in the vicinity of Arctowski Polish Antarctic Station at King George Island were characterized in terms of physicochemical properties (pH, humidity, concentration of selected inorganic and organic substances), microbial colonization and the overall activity of selected enzymes (differentglycosidases and esterases). Activity of the latter enzymes was assayed by p-nitrophenyl and 4-methylumbelliferone derivatives of fatty acids (by spectrophotometric and fluorimetric method, respectively). The highest lipolytic activity was found in soils from the elephant seal colony and penguin rookery.

Dominującą pozycję wśród biokatalizatorów przemysłowych zdobyły enzymy zoptymalizowane pod kątem określonych zastosowań, wytwarzane przez drobnoustroje modyfikowane genetycznie (GMO), służące do ekspresji natywnych i zmutowanych genów. Produkcja tych enzymów jest prowadzona w ściśle kontrolowanych warunkach, zapewniających czystość mikrobiologiczną finalnego produktu oraz uniemożliwiających wydostanie się rekombinowanego organizmu na zewnątrz. Biokatalizatory wytwarzane z zastosowaniem GMO charakteryzują się wyższą aktywnością, stabilnością i precyzją działania w porównaniu z preparatami tradycyjnymi.

Tailormade enzymes, optimized for individual applications have gained the dominating position on the market of commercial biocatalysts. They are synthesized by genetically modified microorganisms (GMOs), used in order to express native and mu-tated genes. Production of these enzymes is run under conditions providing microbial safety of final enzymatic preparations and protecting from any leakage of transformants outside. Biocatalysts synthesized by GMOs display higher activity, stability and precision of function, in comparison to traditionally produced enzyme preparations.

W drugiej połowie lat dziewięćdziesiątych bardzo popularnym zagadnieniem rozpatrywanym przez Podkomisje UIC jest rozwój platform do przewozów kontenerów w tzw ruchu kombinowanym szynowo-drogowym. Zespół takiego pociągu towarowego (tzw pociągu zwartego) składa się z dwóch wagonów skrajnych oraz z wagonów środkowych. Masa każdej platformy nie może być większa aniżeli 16 000 kg. Bardzo poważne ograniczenia ciężaru każdego wagonu wywierają znaczny wpływ na konstrukcję układu biegowego.C

In the second half of ninetieth years very important question considered by UIC Subcommittee was the development of platforms for the transport of the containers in the combined rail-route movement. That goods-train (so called compact train) is composed of two extreme wagons and middle wagons. The mass of each platform should be no greater as 16000 kg. That very great limitation of the weight of each wagon exerts a considerable influence on construction of the running system.I
