Badania przeprowadzono na ponad 8 tysiącach ejakulatów pobranych od 112 knurów. Badaniami objęto wszystkie ejakulaty pobrane i zakwalifikowane do inseminacji w czteroletnim okresie działania jednej ze stacji unasienniania loch. Nasienie od knurów pobierano dwa razy w tygodniu, metodą manualną, rano po pierwszym karmieniu. Pobrane ejakulaty poddano ocenie według metodyki stosowanej w polskich stacjach unasienniania loch. Zebrane dane podzielono na 12 podgrup obejmujących ejakulaty pobierane w poszczególnych miesiącach roku. Wykazano, że cechy ejakulatów knurów inseminacyjnych zależą od pory roku pobrania. Ejakulaty pozyskiwane w okresie jesienno-zimowym cechują się dużą objętością i zawierają najwięcej plemników o ruchu postępowym. Ejakulaty te cechują się także relatywnie dużą ruchliwością plemników. Ma to związek z dużą liczbą dawek inseminacyjnych wytwarzanych z ejakulatów pobieranych w tym okresie. Ejakulaty pobierane w okresie lata (czerwiec–sierpień) zawierają mniej plemników wykazujących prawidłowy ruch. Uzyskuje się z nich o około 3 dawki inseminacyjne mniej niż z ejakulatów pobieranych w okresie zimowym.

We investigated more than 8 thousand ejaculates collected from 112 boars. The investigation involved all the ejaculates collected and qualified for insemination during a four- -year period at a sow insemination centre. Semen was manually collected from the boars twice weekly, in the morning, after the first feeding. The collected ejaculates were evaluated according to a methodology used at Polish sow insemination centres. The data we gathered were divided into 12 subgroups comprising ejaculates collected in particular seasons of the year. We found the characteristics of insemination boar ejaculates to be dependant on the season in which the ejaculates were collected. Ejaculates obtained in the autumn-winter period are characterised with a high volume and contain the greatest number of progressively motile spermatozoa. These ejaculates also contain relatively highly mobile sperms. This results in obtaining a high number of insemination doses prepared from ejaculates collected at the time. Ejaculates gathered in the summer (June-August) contain fewer correctly motile spermatozoa. Roughly, they produce 3 insemination doses fewer than the ejaculates collected in the autumnwinter period.

A study was carried out of 30, 160 ejaculate samples collected from 81 boars of the wbp (Polish Large White) breed and 182 boars of the pbz (Polish Landrace) breed. The ejaculate was collected manually with the sampling frequency of 4 - 5 days. An assessment of the ejaculate has been done with regard to the following physical properties: ejaculate volume, sperm cells concentration, percentage of sperm cells exhibiting proper motility, total number of sperm cells in the ejaculate and number of insemination doses obtained from one ejaculate. The collected data have been analyzed according to the season criterion with indication of the ejaculate collected in the individual months. It has been revealed that the most suitable ejaculate was collected from the domestic pig boars in November and December whereas in March, April and May the parameters lowered. The ejaculate collected in the springtime is characterized by a lower volume and a lower number of the motile sperm cells than in the other seasons. From the ejaculate collected in the autumn and winter period more insemination doses can be prepared than from those collected in spring and summer. No difference has been found between the boars of the wbp and pbz breeds with regard to their susceptibility to the seasonal factors’ influence.

The experimental material consisted of 75 ejaculates collected form 8 Duroc boars. The ejaculates were divided into three groups according to sperm concentration in an ejaculate. An ejaculate was obtained from each boar monthly and it was used to make microscopic preparations to examine spermatozoa morphology. In each preparation morphometric measurements were taken of fifteen randomly selected spermatozoa characterized by normal morphology. The following measurements of spermatozoa were taken: length and width of the spermatozoa head, head area, length of the flagellum, perimeter of the spermatozoon head and total spermatozoon length. The results were used to calculate indicators of spermatozoa morphology. Moreover, assessments were made of frequency of morphological defects to isolate spermatozoa with primary and secondary abnormalities following the Blom classification system. It was found that the concentration of spermatozoa in the ejaculate influenced the morphometric characteristics of spermatozoa. Ejaculates with low sperm concentrations are characterized by larger spermatozoa as compared to ejaculates with high sperm concentrations. However, sperm concentration in the ejaculate does not much influence the shape of spermatozoa.

The aim of the study was to characterize the enzymatic and non-enzymatic components comprising the antioxidant system in spermatozoa and individual fractions of dog ejaculate. Ejaculates were collected from six dogs of mixed-breeds. Total protein content, activity of antioxidant enzymes and the content of low-molecular antioxidants, such as L-glutathione (GSH), L-ergothioneine (ERG), L-ascorbic acid and total SH-group, were analyzed in the ejaculated spermatozoa and seminal plasma of the pre-spermatic, spermatic and post-spermatic fractions. The total antioxidant status (TAS) and antiperoxidant activity of the seminal plasma were also determined. The enzymatic antioxidant system of canine spermatozoa is mainly represented by superoxide dismutase (SOD) activity and, to a lesser extent, by glutathione peroxidase (GPx) and phospholipid hydroperoxide glutathione peroxidase (PHGPx) activity. Catalase activity was not detected either in the spermatozoa or in the different ejaculate fractions. GSH and ERG were detected in each fraction. Furthermore, a high level of L-ascorbic acid was observed in fractions of the ejaculate. Proteins and low-molecular weight antioxidants could influence the total antioxidant status and antiperoxidant activity of the seminal plasma. Increased suppressive activity against lipid peroxidation was shown only in the pre-spermatic and post-spermatic fractions. The different fractions of dog ejaculate, which are the main source of enzymatic antioxidants and low-molecular antioxidants, play an important role in protecting spermatozoa against reactive oxygen species.

Relations between the concentration of glucose and urea in blood plasma and the qualitative ejaculate parameters were investigated in 24 breeding bulls (13 Holstein and 11 Czech Spotted) over a 12-month period. The average concentrations of plasma glucose and urea were 3.32 mmol ∙ 1⁻¹ and 3.57 mmol ∙ 1⁻¹ , respectively. No statistically significant differences in the concentrations of glucose and urea (i.e. 3.34 vs. 3.30 mmol ∙ 1⁻¹ and 3.53 vs. 3.61 mmol ∙ 1⁻¹, respectively) were observed between the Holstein bulls and Czech Spotted bulls. A statistically significant (P ≤ 0.05) positive correlation was found between the concentration of plasma urea and the ejaculate volume for the whole investigation period (rp = 0.19). Similarly, the correlation between the concentration of plasma urea and sperm motility was positive (rp = 0.11), whereas the increase in plasma urea concentration negatively influenced sperm concentration ( rp = -0.10). Correlations between plasma glucose concentrations and the respective parameters of ejaculate quality were similar in magnitude, i.e. ejaculate volume ( rp = 0.12), sperm motility ( rp = 0.12) and sperm concentration ( rp = -0.08). During the investigations the coefficients of correlation between plasma glucose and urea concentrations as well as qualitative parameters of the ejaculate were found to be negative in months when concentrations of glucose and urea in the blood plasma of the bulls markedly changed.

Zależności pomiędzy koncentracją glukozy i mocznika w surowicy krwi i jakościowymi cechami ejakulatów badano w okresie 12 miesięcy u 24 (13 rasy holsztyńskiej i 11 czeskiej pstrej) buhajów rozpłodowych. Średni poziom glukozy i mocznika w surowicy krwi wynosił odpowiednio: 3,32 mmol ∙ 1⁻¹ i 3,57 mmol ∙ 1⁻¹. Nie stwierdzono istotnych statystycznie różnic w obu grupach buhajów w poziomach glukozy i mocznika (tj. 3,34 i 3,30 mmol.1⁻¹ oraz 3,53 i 3,61 mmol ∙ 1⁻¹). Dla całego okresu badań wykazano istotne na poziomie P ≤ 0,05, dodatnie zależności pomiędzy poziomem mocznika a objętością ejakulatu ( rp = 0,19). Również dodatnia okazała się zależność pomiędzy poziomem mocznika a ruchliwością plemników ( rp = 0,11). Zwiększanie się poziomu mocznika wpływało ujemnie na koncentrację plemników ( rp = - 0,10). Obliczone współczynniki korelacji pomiędzy poziomem glukozy a cechami nasienia były podobnego rzędu, tj. z objętością ejakulatu ( rp = 0,12), z ruchliwością plemników ( rp = 0,12) i koncentracją plemników ( rp = -0,08). Stwierdzono występowanie ujemnych wartości wskaźników korelacji pomiędzy poziomami glukozy i mocznika a cechami jakościowymi ejakulatu w czasie (miesiące), gdy poziom ich w surowicy krwi buhajów ulegał nagłym zmianom.

The present study was undertaken to investigate the effect of various concentrations of cobalamin (vitamin B₁₂) in Bioexcell® extender on the spermatozoa quality of Simmental bulls. The semen was collected from 12 bulls by means of an artificial vagina. Vitamin B₁₂ at concentrations of 1.50, 2.50, 3.50, and 5.00 mg/mL was added to bovine Bioexcell® extender. The semen aliquots were cooled and preserved at 4°C. Their quality was evaluated during pre-freezing, and then the cooled semen samples were packaged into 0.25 ml straws. The straws were frozen in the vapor of liquid nitrogen, and stored at -196ºC in a container. The straws were thawed one day later, and the characteristics of spermatozoa were examined. The results showed that the effect of vitamin B₁₂ on the viability, DNA fragmentation and motility of spermatozoa was significant under both pre- and post-freezing conditions (p <0.05), and revealed that supplementation of the extender with B₁₂ improved the post-thaw spermatozoa quality in bulls.

The aim of this study was to determine the activity of proacrosin and acrosin in spermatozoa originating from the sperm-rich fractions (SRF) and whole ejaculates (WE) of dog semen. In addition, experiments were conducted to determine the activity of antitrypsin inhibitors in the fluids of different ejaculate fractions and whole seminal plasma. Ejaculates were collected from five dogs of mixed breed and one Beagle dog (aged from 2 to 9 years). In the SRF, it was confirmed that the activity of the free acrosin form was predominant (acrosin / proacrosin; 2.38 ± 0.22 / 1.05 ± 0.08 mIU / 106 spermatozoa). On the other hand, spermatozoa originating from the WE exhibited significantly higher (p<0.05) proacrosin activity (proacrosin / acrosin; 2.19 ± 0.19 /1.30 ± 0.11 mIU / 106 spermatozoa). Furthermore, acrosin inhibitor activity was lower in the fluids of the pre-sperm fraction (0.09 ± 0.006 IU / cm3), whereas it was higher in the fluids of the post-sperm fraction (0.11 ± 0.007 IU / cm3). Using PAGE analysis, the antitrypsin activity of the enzyme was represented by the presence of one electrophoretic band in the fluids of the pre-sperm and post-sperm fractions and whole seminal plasma. Furthermore, two electrophoretic bands were detected in the fluids of the SRF. The findings of this study indicate that specific proteinase inhibitors present in the individual ejaculate fractions of dog semen may act by stabilizing the sperm acrosin system.

Целью труда было определение связи между активностью энзимов в плазме семени 6-месячных хрячков и его качеством выраженным количественными параметрами и показателями микроскопной оценки в рамках трех породных групп: крупной белой польской породы (32 головы), породы дюрок (29 голов) и многопородных помесей (22 головы) содержимых в Гибридизационном центре Павливице. Биохимическая оценка семени была выражена активностью энзимов в его плазме (AspAT, ALAT, LDH и АР). Среди анализируемых биохимических тастов плазмы семени, свидетельствующих о степени повреждения сперматозоидов, не был найден универсальный тест, который смог бы касаться хотя бы одного параметра количественной и микроскопной оценки для всех исследуемых породных групп. Характерными и специфическими представляются высокие коэффициенты корреляции для связи между активностью AspAT и ALAT в плазме семени с одной стороны и движением массы сперматозоидов (r = -0,57** , -0,67**) и общим числом сперматозоидов в эякуляте (r = -0,45* и -0,75**) и между активностью LDH и концентрацией и процентом живых сперматовоидов (r = -0,67** и -0,61**) с другой стороны для хрячков породы дюрок, семя которых характеризовалось самыми низкими значениями в проведенной оценке. Была получена прямая эависимость между активностью LDH и процентов живых сперматозоидов (r = 54*) в рамках помесных храчков. Рост процента сперматозоидов с основании изменениями связан с пониженной активностью LDH в плазме семени (r = -0,40*).

The aim of the work was to find the relationship between the activity of enzymes in the semen plasma of the 6-month boars and its value expressed in terms of quantitative parameters and microscopic estimation indices within the framework of three breed groups: Polish Large White (32 heads), Duroc (29 heads) and polybred hybrids (22 heads) at the Hybridization Centre of Pawłowice. The biochemical assessment of the semen was expressed in terms of activity of enzymes in its plasma (AspAT, AlAT, LDH and Ap). In analyzed biochemical tests of the semen plasma concerning the spermatozoid injury degree no universal test, which could concern at least one parameter of the quantitative and microscopic estimation for all breed groups, has been found. Characteristic and specific seem to be the obtained high coefficients of correlation for the relationship between the AspAT and AlAT activity in the semen plasma, spermatouoa mass movement (r = -0.57**, -0.67**) and total number of spermatozoa in the ejaculate (r = -0.45* and -0.75**) on the one hand and the AlAT activity and concentration and per cent of living spermatozoa (r = -0.67** and -0.61**) for the Duroc boars, the semen of which appeared to be the worst in the ejaculate, on the other. A simple relationship between the LDK activity and the per cent of living spermatozoa (r = 54*) within the group of hybrid boars has been obtained. Increase of the per cent of spermatozoa with basic changes is connected with a lowered LDH activity in the semen plasma (r = -0.40*).

Celem badań była analiza wpływu rasy/wariantu krzyżowania młodych knurów, w wieku od rozpoczęcia użytkowania do ukończenia pierwszego roku życia, na wybrane cechy biologiczne ich nasienia i liczbę dawek, jaka może być sporządzona z jednego ejakulatu. Podjęto również próbę określenia wpływu wieku na cechy nasienia w obrębie rasy wielkiej białej polskiej (wbp) i mieszańców – hampshire x duroc (hamp x dur).Materiał badawczy stanowiło łącznie 7719 ejakulatów pobranych od osobników rasy polskiej białej zwisłouchej (pbz) i wbp, oraz mieszańców hamp x dur, użytkowanych w stacji unasieniania loch w Kraśniku. Badane osobniki podzielono na dwie grupy, w zależności od wieku: I gr. – młode knury w wieku od rozpoczęcia użytkowania do ukończenia pierwszego roku życia, II gr. – knury w drugim roku życia. Ejakulaty oceniano na podstawie: objętości, koncentracji plemników, odsetka plemników o ruchu postępowym i liczby dawek inseminacyjnych uzyskanych z ejakulatu. W badaniach stwierdzono, że parametry ejakulatów młodych knurów ocenianych ras były istotnie zróżnicowane. Największą objętością ejakulatu, odsetkiem plemników o ruchu postępowym i liczbą sporządzonych dawek inseminacyjnych z jednego ejakulatu charakteryzowały się młode knury pbz. Knury mieszańce hamp x dur charakteryzowały się najwyższą koncentracją plemnikow (422,76 tys. na mm3). Stwierdzono istotny wpływ wieku na analizowane cechy u knurów rasy wbp oraz mieszańców (z wyjątkiem koncentracji plemników).Wysoki potencjał reprodukcyjny, wyrażony liczbą dawek inseminacyjnych (powyżej 20 szt.) prezentowały knury rasy wbp i mieszańce hamp x dur w drugim roku życia.

The aim of the study was to analyse the influence of a breed/crossbreed of young boars, at the age from starting the exploitation till the end of their first year of life on selected biological features of their semen and on the number of doses which can be made from one ejaculate. An attempt has been made to determine the influence of age on features of semen in Polish LargeWhite (PLW) breed and crossbreed boars – Hampshire x Duroc (Hamp x Dur). The material under investigation consisted of the total of 7 719 ejaculates taken from boars belonging to Polish Landrace (PL) breed, PLWbreed and from crossbreed boars Hamp x Dur, used at the insemination station in Kraśnik. The investigated boars were divided into two groups, depending on their age: the first group comprised boars at the age from starting the exploitation till the end of their first year of life, and the second group consisted of boars at the second year of life. Ejaculates were compared with respect to their volume, concentration of spermatozoa, percent of progressive motility spermatozoa and the number of insemination doses obtained from one ejaculate. The results of the study have shown that the parameters of ejaculates of young boars of the investigated breeds varied considerably.Young boars belonging to PL breed were characterized by the largest volume of ejaculate and the largest percent of progressive motility spermatozoa; also the largest number of insemination doses which could be made from one ejaculate in the case of these boars. Crossbreed boars Hamp x Dur were characterized by the highest concentration of spermatozoa (422.76 thousand per mm3). Age of young boars belonging to PLWbreed had a significant impact on all the analysed features; in the case of crossbreed boars Hamp x Dur no distinct changes in ejaculates taken from boars which were up to up to 24 months old were observed only in the concentration of spermatozoa. High breeding potential, which manifested itself in the number of insemination doses (above 20), was also observed in the case of boars belonging to PLWbreed and crossbreed boars Hamp x Dur at the second year of life.

Badania przeprowadzono na 48 knurkach mieszańcach (♀ polska biała zwisłoucha x ♂ wielka biała polska) o początkowej masie ciała ok. 42-45 kg. Zwierzęta podzielono na 3 grupy otrzymujące: 1) kontrolną mieszankę zbożowo-sojową, 2) mieszankę z udziałem 10% poekstrakcyjnej śruty rzepakowej i 3) mieszankę z udziałem 12% śruty z nasion rzepaku dwuzerowego. 24 knurki ubito przy średniej masie ciała 115 kg, a pozostałe jako zwierzęta doświadczalne przyuczono do oddawania ejakulatów i kontynuowano badania nad wpływem długotrwałego podawania mieszanek z udziałem produktów rzepakowych na przebieg wzrostu i jakość nasienia knurów użytkowanych do wieku 24 miesięcy. Ocena jakości ejakulatów obejmowała następujące parametry: całkowita objętość ejakulatu, objętość ejakulatu po przesączeniu, ruchliwość plemników, koncentracja plemników, ogólna liczba plemników w ejakulacie, aktywność fosfatazy alkalicznej, aktywność fosfatazy kwaśnej oraz zawartość białka ogólnego w plazmie nasienia. Ogółem oceniono 378 ejakulatów. Podawanie mieszanek doświadczalnych nie wpłynęło istotnie różnicująco na tempo wzrostu oraz spożycie i wykorzystanie paszy przez knury. Średnia masa ciała knurów w wieku 2 lat wynosiła w kolejnych grupach: 287.7; 298.0 i 299.5 kg. Zróżnicowane żywie­nie knurów nie wpłynęło w sposób konsekwentny i jednoznaczny na wszystkie analizowane cechy ejakulatów.

The research was carried out on 48 crossbreed boars (♀ Polish Landrace x ♂ Polish Large White), with initial body weight equal to ca. 42-45 kg. They were divided into 3 groups and fed on: 1) control cereal-soyabean diet, 2) diet containing 10% of rapeseed meal, and 3) diet containing 12% of rapeseed '00' meal. 24 animals were slaughtered when their average body weight amounted to 115 kg. The rest of the animals were taught to give ejaculates, which enabled to continue studies concerning the effect of long-term feeding on diet containing rape products on growth and semen quality of boars aged up to 24 months. Evaluation of ejaculate quality included the following parametres: total volume of ejaculate, ejaculate volume after filtration, spermatozoa mobility and concentration, total number of spermatozoa in ejaculate, activity of AP and AcP, and total protein content in semen plasma. 378 ejaculates were evaluated altogether. The experimental diets had no significant effect on growth rate and feed intake and feed conversion. Average body weight of boars aged 2 years was, in respective groups, equal to 287.7; 298.0 and 299.5 kg. Differentiated feeding did not affect the analysed ejaculate parametres in a definite way.
