Background & Aims: Infrared spectroscopy is an increasingly common method for bacterial strains' testing. For the analysis of bacterial IR spectra, advanced mathematical methods such as artificial neural networks must be used. The combination of these two methods has been used previously to analyze taxonomic affiliation of bacteria. The aim of this study was the classification of Escherichia coli strains in terms of susceptibility/resistance to cephalothin on the basis of their infrared spectra. The infrared spectra of 109 uropathogenic E. coli strains were measured. These data are used for classification of E. coli strains by using designed artificial neural networks. Results: The most efficient artificial neural networks classify the E. coli sensitive/resistant strains with an error of 5%. Conclusions: Bacteria can be classified in terms of their antibiotic susceptibility by using infrared spectroscopy and artificial neural networks.

This study aimed to assess the prevalence of enterococci (including vancomycin-resistant, VRE) strains in meat plants and evaluate their biofilm-forming potential. In two Polish meat plants, 75 samples of raw pork meat, swabs from work surfaces (floors, tables, machine parts and tools) and employees’ hands were collected. The analyses indicated that enterococci were present in more than 72% of the tested samples. In addition, VRE isolates were found in more than 25% of the tested samples (especially in cutting and processing rooms). VRE strains of Enterococcus faecium and E. faecalis were resistant to penicillin, ampicillin, erythromycin, and rifampicin. Moreover, 77% of E. faecium and 43% of E. faecalis isolates showed biofilm-forming ability. The observed high biofilm-forming potential among the analyzed VRE strains indicates that these agents may play an essential role in spreading drug resistance in the food chain through contaminated surfaces, meat, and workers’ hands.

This study deals with the antibiotic resistance of heterotrophic bacteria isolated from the surface and subsurface water of the Gdansk Deep. The level of resistance of bacteria to various antibiotics differed considerably. As a rule, there were no significant differences in antibiotic resistance between neustonic and planktonic bacteria. Considerable diel fluctuations in bacterial antibiotic resistance were found. There were significant differences between pigmented and non-pigmented bacteria in their resistance to the tested antibiotics. Bacterial resistance to antibiotics was dependent on their chemical structure.

Background: Bacterial biofilms have been linked to recurrent adenotonsillar diseases, with special concern regarding therapeutic management hindered by antibiotic resistance. Aims: we aimed to find if there was a relationship between the presence of a bacterial biofilm and the development of cervical lymphadenopthy, tonsillar hypertrophy and adenoid hypertrophy in patients with chronic tonsillitis. Patients and Methods: Tissue samples from tonsillar biopsies of 30 children who underwent tonsillectomy were examined by scanning electron microscopy (SEM). Settings and Design: This cross-sectional study was conducted between August 2012 and July 2015 in Al-Azhar University Hospitals. Results: It was found that 23.3% of children had a fully formed bacterial biofilm (Grade III), 6.7% had grade I and 6.7% had grade II biofilms as demonstrated by SEM. Staphylococcus aureus was the most common identified isolate (26%) followed by Coagulase-negative Staphylococci (17.4%) and Klebsiella pneumoniae (10.4%). No statistically significant difference regarding the presence of a bacterial biofilm and the development of cervical lymphadenopthy, tonsillar hypertrophy or adenoid hypertrophy was found. Conclusion: Bacterial biofilm is a possible cause of the chronicity of tonsillar diseases in children. There is no relationship between the presence of a bacterial biofilm and the development of of cervical lymphadenopthy, tonsillar hypertrophy or adenoid hypertrophy.

The scope of this study was to apply the Biolog system to identify and characterize a Serratia strain isolated from the surface of black plastic pieces which constitute the fluidized bed filter (onsite wastewater technology, OSWT). The preliminary isolation of the strain was done in the medium with tetracycline at a 16 mg/l concentration. To characterize the isolated strain, the following Biolog methods were applied: (1) EcoPlates microplates for evaluation of physiological profiling, (2) GEN III OmniLog® ID System for identification of the isolate, and (3) phenotypic microarrays (PM) technology for evaluation of sensitivity to antibiotics (PM11 and PM12). Results were recorded using the original OmniLog® software. The Serratia strain was identified as Serratia marcescens ss marcescens with similarity index 0.569. The same identification was obtained by the 16S rDNA analysis. PM analysis showed an enhancement of phenotype (resistance or growth) of this strain to 35 antibiotics. The loss of phenotype (sensitivity or non-growth) was observed only for 5 antibiotics: lomefloxacin (0.4 µg/ml), enoxacin (0.9 µg/ml), nalidixic acid (18.0 µg/ml), paromomycin (25.0 µg/ml) and novobiocin (1100 µg/ml). This study acknowledges that the methods proposed by the Biolog system allow correct and complete identification and characterization of the microbes isolated from different environments. Phenotypic microarrays could be successfully used as a new tool for identification of the multi-antibiotic resistance of bacteria and for determination of the minimal inhibition concentrations (MIC).

Antibiotic resistance of Staphylococcus-like organisms (STLO) isolated from a recreational sea beach located on the southern coast of the Baltic Sea was studied. The results of the present study showed that STLO inhabiting sand and seawater of the beach strongly differed in the resistance level to tested antibiotics. These microorganisms were most resistant to ampicillin, penicillin, oxytetracycline and susceptible to gentamicin, neomycin and streptomycin. Moreover, the level of antibiotic resistance among bacteria isolated from different parts of the beach also differed. Bacteria inhabiting the seawater, shoreline and the middle part of the beach were more antibiotic-resistant than bacteria isolated from the dune. The majority of bacteria inhabiting the seawater and sand were resistant to 3–8 antibiotics out of 12 tested in this study. Generally, there was no difference in antibiotic resistance between Staphylococcus-like organisms isolated from the surface and subsurface sand layers. STLO strains isolated from Ustka Beach were most resistant to β-lactam and tetracycline antibiotics, and most susceptible to aminoglycosides.

Antibiotic resistance of 114 strains of the Enterobacteriaceae family bacteria isolated from vegetables, originating from retail, was investigated in the study. The highest number of the strains isolated were resistant to ampicilin (81.9%), whereas a lower number of the strains exhibited resistance to the following antibiotics: neomycin (29.3%), streptomycin (28.4%), rifampicin (21.5%), chloramphenicol (19.8%), colistin (12.9%), and nitrofurantoin (11.%). All the isolated strains appeared to be susceptible to vancomycin, kanamycin, doxycyclin, nalidixic acid and gentamycin.

Określono oporność na antybiotyki 114 szczepów pałeczek z rodziny Enterobacteriaceae wyizolowanych z warzyw, pochodzących z handlu detalicznego. Wśród wyizolowanych szczepów najwięcej było opornych na ampicylinę 81,9%, natomiast mniej szczepów wykazywało oporność na następujące antybiotyki: neomycynę 29,3%, streptomycynę 28,4%, rifampicynę 21,5%, chloramfenikol 19,8%, kolistynę 12,9%, nitrofurantoinę w 11,2%. Wszystkie wyizolowne szczepy były wrażliwe na wankomycynę, kanamycynę, doksycyklinę, kwas nalidyksowy oraz gentamycynę (tab. 3).

Staphylococcus aureus is a very important human pathogen that generates a number of human infections. Isolation of vancomycin-resistant Staphylococcus aureus (VRSA) was carried out from wound samples of patients attending University of Uyo Teaching Hospital, Akwa Ibom state, Nigeria. A total of 45 wound samples from 15 patients were collected aseptically in triplicate, using sterile cotton swabs moistened in sterile normal saline. Microbiological analysis and susceptibility to vancomycin, including minimum inhibitory and minimum bactericidal concentrations (MIC and MBC), were carried out using standard methodologies. A total of 15 isolates were obtained and these showed varying MIC and MBC patterns. Out of the 15 S. aureus isolated, only one isolate had an MIC of approximately 4 µg/ml, while twelve isolates gave MIC values that ranged from 15.62 µg/ml to 250 µg/ml. The remaining 2 isolates gave MIC values that were ≥ 500 µg/ml. These 2 isolates exhibited alpha haemolysis on blood agar, unlike the others that were beta haemolytic. The results of the MBC also showed variations amongst the isolates. A total of 10 isolates gave MBC values that ranged from 62.5 µg/ml to 500 µg/ml, while the remaining 5 isolates gave MBC values that were ≥ 500 µg/ml. The high MIC and MBC values obtained showed that vancomycin-resistant Staphylococcus aureus is increasing at an alarming rate, and this accounts for the gradual decline in the effectiveness associated with the use of vancomycin. Given the widespread prevalence of VRSA, there is a need for newer therapeutics that can reverse this surge.

OmpF plays very important roles in the influx of antibiotics and bacterial survival in the presence of antibiotics. However, high-grade mutant OmpF and its function in decreasing bacterial survival rate have not been reported to date. In the present study, we cloned a high-grade mutant OmpF (mOmpF) and sequence analysis suggested that over 45 percent of the DNA sequence was significantly mutated, leading to dramatic changes in over 55 percent of the amino acid sequence. mOmpF protein was successfully expressed. When grown in the presence of antibiotic, the bacterial survival rate decreased and the antibiotic inhibition zone became larger with the increase of the mOmpF. It was concluded that concentration of high-grade mutant mOmpF dramatically influenced the bacterial survival rate. The study presented here may provide insights into better understanding of the relationships between structure and function of OmpF.

The article reviews the issue of antibiotic resistance of microorganisms to meropenem in intensive care units in Ukraine. An increase in meropenem inefficiency against microorganisms in intensive care units has been observed in the last years. The data analysis suggests a significant predominance of gram-negative flora: A. baumannii, Р. aeruginosa, К. pneumoniae, E. cloacae, E.coli. On average 30% of microorganisms are resistant to 5 and more basic antibiotics including meropenem. 40-80 % of the gram-negative flora in intensive care units in Ukraine are resistant to meropenem. This can be attributed to the free sale of antibiotics without prescription, patients’ self-treatment, inadequate antibiotic therapy, and failure to comply with sanitary norms on the part of intensive care staff. Microbiological diagnostics of infectious pathogens also needs improvement. Unless proper measures are taken within a few years, meropenem as an antibiotic is likely to disappear in Ukraine.

Artykuł przedstawia kwestię antybiotykooporności drobnoustrojów na lek meropenem u pacjentów szpitalnych oddziałów intensywnej opieki medycznej na Ukrainie. W ostatnich latach na ukraińskich oddziałach intensywnej opieki medycznej zaobserwowano znaczny wzrost oporności drobnoustrojów na meropenem, a tym samym coraz mniejszą skuteczność tego leku. Przeprowadzona analiza danych wskazuje na znaczną przewagę gram-ujemnej flory bakteryjnej oddziałów, w tym szczepów takich jak A. baumannii, Р. aeruginosa, К. pneumoniae, E. cloacae czy E.coli. Odsetek szczepów opornych na działanie pięciu lub więcej podstawowych antybiotyków, w tym leku meropenem, wynosił średio 30%, a odsetek gram-ujemnych szczepów flory bakteryjnej ukraińskich oddziałów intensywnej opieki medycznej opornych na działanie samego meropenemu to aż 40 – 80%. Głównymi czynnikami powstawania antybiotykooporności w tym przypadku mogą być m.in. sprzedaż antybiotyków bez recepty, samoleczenie, niewłaściwa antybiotykoterapia oraz niespełnianie wymogów sanitarnych przez personel medyczny szpitali. Zdecydowanie należałoby również popracować nad diagnostyką mikrobiologiczną drobnoustrojów patogennych. Jeżeli w ciągu kilku kolejnych lat nie zostaną podjęte odpowiednie działania, meropenem prawdopodobnie zniknie z listy antybiotyków stosowanych na Ukrainie.

With regard to antibiotic resistance studies in various model animals in the urban environment, the presented study focused on the rook, many behavioural and ecological aspects of which are important from an epidemiological point of view. A total of 130 Escherichia coli strains isolated from rook faeces during a two-year period (2011–2012) were investigated for antibiotic resistance and virulence. Resistance to ampicillin (60%) and streptomycin (40%) were the most frequent, followed by resistance to fluoroquinolones (ciprofloxacin-22% and enrofloxacin-24%), tetracycline (18%), cotrimoxazol (17%) and florfenicol (14%). Ceftiofur resistance occured in 10.7 % of strains and cefquinom resistance in 1.5 % of strains. Twenty-five E.coli strains with a higher level of MICs of cephalosporins (over 2mg/L of ceftazidime and ceftriaxon) and fluoroquinolones were selected for detection of betalactamase genes (CTX-M, CMY), plasmid-mediated quinolone resistance qnrS, integrase 1, and for APEC (avian pathogenic E.coli) virulence factors (iutA, cvaC, iss, tsh, ibeA, papC, kpsII). Genes of CTX-M1, CMY-2, integrase 1, papC, cvaC, iutA were detected in one strain of E.coli, and qnrS, integrase 1, iss, cvaC, tsh were detected in another E.coli. DNA microarray revealed the absence of verotoxin and enterotoxin genes and pathogenicity islands. The results show that rooks can serve as a reservoir of antibiotic-resistant E. coli with avian pathogenic virulence factors for the human population, and potentially transmit such E.coli over long distances.

W środowisku coraz powszechnej stwierdzana jest obecność lekoopornych szczepów Escherichia coli. Alternatywną drogę ich eliminacji mogą stanowić biodegradowalne i nietoksyczne substancje, między innymi olejki eteryczne. Celem pracy była ocena działania olejków eterycznych na środowiskowe lekooporne szczepy Escherichia coli. Materiał badawczy stanowiło 10 wyizolowanych ze środowiska naturalnego lekooopornych szczepów E. coli, w tym 8 glukuronidazo-dodatnich. Wrażliwość szczepów na takie antybiotyki, jak: ampicylina, amoksycylina, cefotaksym, chloramfenikol, cyprofloksacyna, ceftazydym, doksycyklina, gentamycyna, kanamycyna, trimetoprim, tetracyklina, streptomycyna, kwas nalidyksowy oraz na sulfonamidy, oznaczono na podłożu Mueller-Hinton. Metodą dyfuzyjną płytkowo-cylinderkową oceniono wrażliwość szczepów na olejki: tymiankowy, oregano, herbaciany, lemongrasowy, cytrynowy i kminkowy oraz tymol w stężeniach 1,5%; 1,0%; 0,5%; 0,25%. Na podstawie uzyskanych wyników stwierdzono, że testowane szczepy E. coli wykazywały oporność na co najmniej połowę testowanych antybiotyków, a 90% z nich było oporne na ampicylinę i amoksycylinę. Także wrażliwość szczepów na testowane olejki eteryczne była zróżnicowana. Tylko olejek tymiankowy w stężeniach 1 i 1,5% zahamował rozwój 90% badanych szczepów E. coli. Olejki herbaciany, lemongrasowy, cytrynowy, oregano i kminkowy w testowanych stężeniach nie hamowały rozwoju testowanych szczepów Escherichia coli.

In an environment increasingly common finding is the presence of antibiotic-resistant strains of Escherichia coli. An alternative way to eliminate them can provide biodegradable and non-toxic substances, including essential oils. The aim of this study was to evaluate the effect of essential oils on environmental, antibiotic-resistant strains of Escherichia coli. The research material consisted of 10 isolated from the environment, antibiotic-resistant strains of E. coli, including 8 glucuronidase-positive. Using the Mueller-Hinton medium was evaluated the sensitivity to antibiotics such as ampicillin, amoxicillin, cefotaxime, chloramphenicol, ciprofloxacin, ceftazidime, doxycycline, gentamycin, kanamycin, trimethoprim, tetracycline, streptomycin, nalidixic acid and sulfonamides. Using the platelet-cylinders diffusion method was assessed the sensitivity of the strains on essential oils: thyme, oregano, tea tree, lemongrass, lemon, and caraway, at concentrations of 1.5%; 1.0%; 0.5%, 0.25%. Based on the results, it was found that the tested strains of E. coli were resistant to at least half of the used antibiotics and 90% of them showed resistance to ampicillin, and amoxicillin. Also, the sensitivity of the strains to tested essential oils has been diverse. Only thyme oil in a concentration of 1.5% inhibited growth of 90% of the tested strains of E. coli. On the other hand, oregano oil in a concentration of 1.5% inhibited the development of 3 out of 10 tested strains. Oils of tea, lemongrass, lemon and caraway, at the mentioned concentrations have been not bactericidal against strains of Escherichia coli.
