2-Deamino-2-methyl-N10-propargyl-5,8-dideazafolic acid (ICI 198583) is a potent inhibitor of thymidylate synthase. Its analogue, Nα-[4-[N-[(3,4-dihydro-2-methyl-4-oxo-6-quinazolinyl)methyl]-N-propargylamino]phenylacetyl]-L-glutamic acid, containing p-aminophenylacetic acid residue substituting p-aminobenzoic acid residue, was synthesized. The new analogue exhibited a moderately potent thymidylate synthase inhibition, of linear mixed type vs. the cofactor, N5,10 -methylenetetrahydrofolate. The Ki value of 0.34 μM, determined with a purified recombinant rat hepatoma enzyme, was about 30-fold higher than that reported for inhibition of thymidylate synthase from mouse leukemia L1210 cells by ICI 198583 (Hughes et al., 1990, J. Med. Chem. 33, 3060). Growth of mouse leukemia L5178Y cells was inhibited by the analogue (IC50 = 1.26 μM) 180-fold weaker than by ICI 198583 (IC50 = 6.9 μM).

Earlier research by the author brought about findings suggesting that people in a special way process words related to demands of a problem they previously solved, even when they do not consciously notice this relationship. The findings concerned interference in the task in which the words appeared, a shift in affective responses to them that depended on sex of the participants, and impaired memory of the words. The aim of this study was to replicate these effects and to find out whether they are related to working memory (WM) span of the participants, taken as a measure of the individual’s ability to control attention. Participants in the experimental group solved a divergent problem, then performed an ostensibly unrelated speeded affective classification task concerning each of a series of nouns, and then performed an unexpected cued recall task for the nouns. Afterwards, a task measuring WM span was administered. In the control group there was no problem-solving phase. Response latencies for words immediately following problem-related words in the classification task were longer in the experimental than in the control group, but there was no relationship between this effect and WM span. Solving the problem, in interaction with sex of the participants and, independently, with their WM span, influenced affective responses to problem-related words. Recall of these words, however, was not impaired in the experimental group.

In masked priming, a briefly presented prime stimulus is followed by a mask, which in turn is followed by the task-relevant target. Under certain conditions, negative compatibility effects (NCEs) occur, with impaired performance on compatible trials (where prime and target indicate the same response) relative to incompatible trials (where they indicate opposite responses). However, the exact boundary conditions of NCEs, and hence the functional significance of this effect, are still under discussion. In particular, it has been argued that the NCE might be a stimulus-specific phenomenon of little general interest. This paper presents new findings indicating that the NCE can be obtained under a wider variety of conditions, suggesting that it reflects more general processes in motor control. In addition, evidence is provided suggesting that prime identification levels in forced choice tasks - usually employed to estimate prime visibility in masked prime tasks - are affected by prior experience with the prime (Exp. 1) as well as by direct motor priming (Exp. 2 & 3).

The masked prime task was used to investigate low-level inhibitory motor control processes in two groups of children (7-8 years and 11-12 years) and in older adolescents/young adults (16-23 years). Masked prime stimuli, presented below the level of conscious awareness, systematically affected reaction times (RTs) to subsequent supraliminal target stimuli: RTs were longer when prime and target were mapped to the same response than when they were mapped to different responses. This negative compatibility effect did not differ significantly between age groups, consistent with the hypothesis that the underlying low-level inhibition processes are already fully developed in children as young as seven years of age. In contrast, performance differences between response repetition and response alternation trials were significantly larger in children, consistent with the hypothesis that higher-level control processes are less effective in children. Results provide converging evidence that whereas the latter processes are mediated by late-maturing (prefrontal cortical) areas, the former processes are mediated by earlier-maturing (possibly subcortical) structures.

Kinetic and thermodynamic studies were made on the effect of caffeine on the activity of adenosine deaminase in 50 mM sodium phosphate buffer, pH 7.5, using UV spectrophotometry and isothermal titration calorimetry (ITC). An uncompetitive inhibition was observed for caffeine. A graphical fitting method was used for determination of binding constant and enthalpy of inhibitor binding by using isothermal titration microcalorimetry data. The dissociation-binding constant is equal to 350 μM by the microcalorimetry method, which agrees well with the value of 342 μM for the inhibition constant that was obtained from the spectroscopy method. Positive dependence of caffeine binding on temperature indicates a hydrophobic interaction.

Protecting the environment requires ensuring such conditions that organisms living in it did not suffer permanent damage to their structures and life functions. This also applies to algae living in the aquatic environment. Some species of water flora especially quickly react to negative stimuli arriving from the environment. Their reactions to experienced stress may be a subtle criterion for assessing the condition of aquatic environment. To this kind of plants belong Chara, which are bioindicators of water quality. Their response to applied environmental stimuli is rapid changes of the electrical parameters of the cell membrane. Our research shows that there is also a change in the intensity of ultraweak luminescence (UWL) generated by these plants. UWL is a phenomenon that continuously accompanies processes occurring in living cells and its intensity is less than the threshold sensitivity of the human eye. The method of measurement of this little known way of Chara's response to environmental stimuli has the advantage that it is completely non-invasive for the test material. Non-invasiveness is due to the fact that the radiation emitted from the samples is measured outside the cell membrane separating the living matter from the surrounding environment. In our study, after exposure to a selected stimulus, three types of responses of plant cells were observed: 1) - after a temporary increase of UWL, there is a return to the level of light corresponding to undisturbed homeostasis (e.g. when ascorbic acid is added to the environment); 2) - after a temporary increase in the UWL, the emission of photons slightly falls and stabilizes at a new higher level of illumination (when the stress stimulus is e.g. lead acetate, the emission intensity is 2.2 times larger than in the beginning); 3) - after a sudden and significant increase of UWL, the high emission level is maintained for many hours (e.g. after adding sucrose follows a 20-fold increase of UWL). UWL curves obtained in our experiments correlate with the changes of entropy corresponding to the disturbance of homeostasis in individual cases, depending on the applied stimulus. Respectively: for 1) - an increase of entropy is temporary and is followed by a return to values before the application of the stimulus, 2) - a lasting change occurs, which consists in an increase of entropy to a new fixed level, 3) - change in entropy is so large that its value exceeds the so-called point of no return, which means that the disorder of homeostasis leads to cell death. The application of luminescence imaging technique Single Photon Counting Imaging (SPCI) to the test cells have revealed that the emission of photons from different parts of the plant has a different intensity. When the plant suffered mechanical damage by cutting pseudoleaves, the emission from the damaged sites increased dramatically. This observation allows the damage to be located. Thanks to SPCI technique, it is also possible to track over time any repair processes. The clear differentiation of UWL kinetics leads to the conclusion that monitoring its intensity can be a good research tool in environmental protection. It makes it possible to determine whether chemicals found in the aquatic environment are harmful to a given species of plants. It can be also assessed whether the degree of the harm threatens the entire population with extinction. Thus, the presented method can be used as an ecotoxicity test.

Polyphenols are a common group of plant based bioactive compounds, that can affect human health because of their antioxidant and antimicrobial properties as well as free-radical scavenging activity. An increasing interest is observed in the interaction between polyphenols and microbiota occurring in food and the human gut. The aim of the work presented here, was to evaluate the effect of some polyphenolic compounds on the growth of two strains of Bifidobacterium: B. adolescentis and B. bifidum. The influence of some flavonoids: naringinin, hesperidin, rutin, quercetin as well as phenolic acids: gallic, caffeic, p-coumaric, ferulic, chlorogenic, vanillic and sinapic was determined by a 96-well microtiter plate assay. In the experiments the effect of three different concentrations of polyphenols: 2, 20 and 100 µg/ml on the growth of Bifidobacterium strains was investigated. All tested compounds influenced the growth of the examined bacteria. Both stimulatory and inhibitory effects were observed in comparison to the positive control. The strongest impact on the growth of bifidobacteria was observed during the first hours of incubation. The constant inhibitory effect was observed for hesperidin and quercetin addition and was dose-dependent. B. bifidum showed a stronger dependence on phenolic acids content in the medium than B. adolescentis during the first hours of incubation.

The activated sludge process of the digestate liquors after chemical separation was conducted using a 10 L lab-scale sequencing batch reactor (SBR) and a 0.50 m3 pilot-scale SBR independently (with pH control). Due to the relatively high concentration of free ammonia (FA), clear inhibitory effects of the digestate liquors on the nitrifying bacteria were observed. The adaptation of the activated sludge to the toxicity was evaluated with the trends of ammonia uptake rate (AUR) and nitrate utilization rate (NUR). The lab-scale AUR values decreased from 5.3 to 2.6 g N/(kg VSS·h) over time after the addition of digestate liquors (5–10% of the reactor working volume), indicating an apparent FA inhibition on the nitrification process in the FA concentration range of 0.3–0.5 mg N/L. The pilot-scale AUR values increased from 1.8 to 3.6 g N/ (kg VSS·h) in the first two weeks and then decreased to 2.4 g N/(kg VSS·h), showing a lag of the inhibition on the nitrifying bacteria at the FA concentration ≈ 0.15 mg N/L. The lab-scale NURs increased from 2.6 to 10.4 g N/(kg VSS·h) over time, and the pilot-scale NURs increased from 1.0 to 4.0 g N/(kg VSS·h) in a similar pattern. The clear dependence of both the lab- and pilot-scale NURs on time indicated the adaptation of the heterotrophic biomass to the digestate liquors. Ethanol – used instead of fusel oil – was found to be a more efficient external carbon source for better adaptation of the activated sludge under unfavorable conditions.

Glutathione reductase (GR, E.C 1.6.4.2) is a flavoprotein that catalyzes NADPH-dependent reduction of oxidized glutathione (GSSG) to reduced glutathione (GSH). The aim of this study was to investigate in vitro effects of phenolic compounds isolated from Sideritis brevibracteata on bovine kidney GR. The Sideritis species are widely found in nature and commonly used as medicinal plants. 7-O-glycosides of 8-OH-flavones (hypolaetin, isoscutellarein and 3'-hydroxy-4'-O-methylisoscutellarein) were isolated from aerial parts of Sideritis brevibracteata. These compounds inhibited bovine kidney cortex GR in a concentration-dependent manner. Kinetic characterization of the inhibition was also performed.

The actual process of the electronic-financial globalisation requires a certain symbolic uniformity, which, in turn, needs languages, even in the most marginal way, although generally this globalised system tends to consider them as a ballast. From the perspective of this symbolic platform, we can talk about the Global Glasshouse, with thin and transparent sides, from/to which one can be expulsed and/or admitted. Languages, thanks to their constitutive malleability, accommodate themselves to this big glasshouse. For that reason, we can say that the intercultural Spanish language as a communicative breath can cushion an unreal-speculative globalisation, which, however, is meant to have ability to (re)convert languages – despite all its initial restrictions – into a permanently exchangeable merchandise within the flow of the symbolic market.

Retrieval Induced Forgetting (RIF) refers to the fi nding that the retrieval of some items from memory (RP+) impairs the retrieval of related items (RP-). The RIF effect is indicated by a comparison of RP- with unrelated but also tobe- remembered items (NRP). Since RIF appears during intentional memorizing of words, therefore we checked whether it depends on attentional control (AC) involved in goal maintenance, and also if implicit evaluations of to-be-remembered (RP) contents moderate this process (causing e.g. inhibition). In three experiments, each including AC as the independent variable, we found AC to be related to the RIF effect. Only high but not low AC subjects showed the presence of RIF. The results of the affective priming procedure showed that implicit evaluations of NRP items moderate the relationship of high AC and the RIF effect. The explanation why temporarily devaluated NRP could enhance the RIF effect and suggestion concerning future research summarize the article.

The list of species of the plant parasitic nematodes presented in this paper (133 species belonging to 14 families) is based on the results of faunistic research conducted in the Wielkopolska region by Polish nematologists up until the year 2010, and the results obtained from the project “Elaboration of Innovative Methods for Rapid Identification of Nematodes Causing Damage to the Economy” managed by the Museum and Institute of Zoology of the Polish Academy of Sciences. During the two years of the project (2010-2011) we found 21 species of nematodes which had not yet been reported in the list of species from the Wielkopolska region. Two of them were reported for the first time in Poland.

 Poly(ADP-ribose) polymerase (PARP) plays a crucial role in DNA repair. Modulation of its activity by stimulation or inhibition is considered as a potentially important strategy in clinical practice, especially to sensitize tumor cells to chemo- and radiotherapy through inhibition of DNA repair. Here we studied the effect of the three PARP inhibitors, 5-iodo-6-amino-benzopyrone (INH2BP), 1,5-isoquinolinediol (1,5-dihydroxyisoquinolinediol (1,5-IQD) and 8-hydroxy-2-methylquinazolin-4-[3H]one (NU1025), and for two of them the efficiency in slowing the rejoining of DNA strand breaks induced by H2O2 was compared. Inhibition of PARP changed its intranuclear localization markedly; cells exposed to the inhibitor NU1025 showed a significant tendency to accumulate PARP in large foci, whereas in untreated cells its distribution was more uniform. The speed and efficiency of rejoining of H2O2-induced DNA strand breaks were lower in cells incubated with a PARP inhibitor, and the kinetics of rejoining were modulated in a different manner by each inhibitor. At a concentration of 100 µM the efficiency of the inhibitors could be ranked in the order NU1025 > IQD > INH2BP. The two first compounds were able to decrease the overall PARP activity below the level detected in control cells, while INH2BP showed up to 40% PARP activity after exposure to H2O2.

Inhibition of jack bean urease by a mixture of boric acid and phosphate buffer pH 6.96 was studied. The inhibition constants of the mixture were determinated in solutions of phosphate buffer (22-155mM) by measuring the initial reaction rates for a series of boric acid concentrations (0-3 mM) at a series of substrate concentrations (2-50 mM urea). It was found that boric acid is a simple competitive inhibitor of urea se with the inhibition constant of the system changing from 0.23 mM in 22 mM phosphate buffer to 0.76 mM in 155 mM phosphate buffer. It was shown, that boric acid and phosphate buffer pH 6.96 are mutually exclusive competitive inhibitors of jack bean urease. The inhibition constants of boric acid and of phosphate buffer pH 6.96 in an inhibitor-free system were determinated to be 0.12 mM and 21 mM, respectively.
