Przekazywanie bakteriofagów niosących geny dla verotoksyn (Shiga-like toksyn) - stx-1 i stx-2, między pałeczkami jelitowymi odgrywa kluczową rolę w rozprzestrzenianiu się szczepów zdolnych do produkcji verotoksyn. Bakteriofagi te, w przeciwieństwie do dzikich szczepów pałeczek jelitowych, są łatwo akceptowane przez laboratoryjny szczep E. coli C600. Spośród 125 dzikich szczepów pałeczek z rodziny Enterobacteriaceae, głównie E. coli, żaden in vitro nie akceptował bakteriofagów warunkujących wytwarzanie verotoksyn.

Lysogenised verotoxigenic strains are the source of structural genes of verocytotoxins (stx-1 and stx-2) for the others intestinal bacili. The aim of the study was to estimate the ability of transfer of bacteriophages induced with UV irradiation from reference verotoxigenic strains of E. coli O157:H7 (CB571 and EDL933) into 125 wild-strains of bacili of Enterobacteriaceae family. None of tested recipient strains showed the production of cytotoxin on Vero and HeLa cell lines, what was acknowledged as the lack of stx genes. Contrary to the laboratory strain of E. coli C600 none of 125 tested recipient strains accepted the phages. Obtained lysogenised laboratory strains of E. coli C600/CB571 and E. coli C600/EDL933, besides of the ability to produce verotoxins (with the presence of stx-1 and stx-2 genes), did not differ phenotypically and genotypically from parent strain of E. coli C600. The estimation of the ability to transfer of phages carried stx-1 and/or stx-2 genes was impossible because of too small number of tested wild strain of bacili or because of really low frequency of acceptation of phages by wild strains of intestinal bacili.

Pod koniec lat osiemdziesiątych opisano nową grupę enteroagregacyjnych szczepów Escherichia coli, które skupiają potencjał chorobotwórczy wszystkich, dotychczas poznanych patogenów jelitowych i które charakterystycznie związane są z przewlekłymi, trwającymi nawet kilka miesięcy, biegunkami.

Enteroaggregative strains of E. coli (EAEC) are an important agents possessing among many virulence factors, aggregative fimbria AAF hcmagglutinating in the presence of mannose human group A or/and rats erythrocytes. The aim of the study was to determine the correlation between the presence of AAF fimbria and pattern of adherence in vitro. Tested strains of E. coli were obtained from children with diarrhea (133 strains) and healthy children (105 strains). Among strains of E. coli from children with diarrhea 81 (61%) showed the presence of AAF fimbria and 19 (23%) were adhering in aggregative pattern. In the group of strains of E. coli isolated from healthy children 31 (30%) were AAF positive and 8 (25,8%) of them presented aggregative adherence. Examination of AAF fimbria only dose not allow to distinguish EAEC strains. The data showed the participation of EAEC strains in diarrhea of children below 3 years old.

The aim of the study was to determine the expression of mannose-sensitive and mannose-resis- tant adhesins by agglutination of cattle, sheep, goat, rabbit, horse, and chicken red blood cell assay, and curli fimbriae by Congo red binding assay among 341 E. coli strains isolated from 51 milk samples of clinically recognized bovine mastitis. Curli fimbriae expression within biofilms created on an inert surface was also investigated. To determine whether curli fimbriae are expressed both in conditions optimal for their production and in conditions resembling the host organism, the study was conducted in anaerobic atmosphere at 37CC, and at room temperature in aerobic atmosphere. The results demonstrated that although the E. coli isolates examined were deprived of mannose-sensitive and mannose-resistant adhesins they were able to produce curli fimbriae in both aerobic and anaerobic conditions at room and higher temperature, indicating that these adhesins may be involved in the pathogenesis of bovine mastitis.

Podstawowym rezerwuarem szczepowi', coli wytwarzających toksyny shiga jest bydło. W województwie dolnośląskim wśród zdгоwego bydła wykazano nosicielstwo pałeczek E. coli wytwarzających toksyny shiga, które mogą stanowić źródło zakażeń ludzi.

The aim of the study was to determine if cattle from the region of Lower Silesia is the reservoir of shiga toxin-producing E. coli strains (STEC) and the analysis of virulence factors of isolated STEC strains. The ability of tested animal strains to shiga toxin synthesis was analysed in cytotoxicity assay in vitro on Vero cell line and then confirmed by detection of shiga toxin-encoding genes by PCR. STEC strains were isolated from 12 (15,2%) of animals examined, 21,4% of these strains were obtained from 9 of 42 calves, and 8,1% from 3 of 37 cows. Most of STEC isolated (75%) was enterohemolysin- producing. The cattle from the region of Lower Silesia is the reservoir of pathogenic for humans sorbitol-fermenting non-0157 STEC strains.

The association between Escherichia coli virulence factors and chronic intestinal disorders is mostly unknown. The presented study compared the distribution of virulence genes and phylogroups among E. coli isolated from chronic intestinal disorders such as Crohn’s disease and irritable bowel syndrome (IBS) with strains isolated from patients with acute diarrhea as a control group. The presence of 159 virulence genes corresponding to known E. coli pathotypes was determined among 78 E. coli archive strains isolated from IBS, acute diarrhea and Crohn’s disease using CGH microarray. E. coli isolated from IBS demonstrated a mosaic of virulence genes specific to enteropathogenic, enterotoxigenic, enterohemorrhagic E. coli strains and Shigella species. In contrast, virulence factors and phylogroups distribution among E. coli isolated from children with acute diarrhea was similar to extraintestinal E. coli strains that probably acquired some virulence genes. The acquisition of virulence genes might have an impact on diarrheagenic potential of these strains. On the other hand, E. coli isolated from children with Crohn’s disease seem to be similar to adherent-invasive E. coli strains (AIEC), as it lack most known virulence genes. The presented study showed that these analyzed groups of E. coli strains differed from each other with the respect to the distribution of virulence genes. The differences in gene content support the idea that the participation of E. coli in chronic intestinal diseases is mostly related to virulence potential of these strains.
