A specimen of an unidentified parasite was taken out of the eye ball of an 8-year-old boy hospitalized in April 1964. A detailed analysis of the parasite showed that in was the Ist larval stage of Hypoderma bovis. The conservative treatment was unsuccessful and the eye ball, largely domaged by the parasite, had to be removed.

Three acidic glycosidases: β-galactosidase (β-GAL, EC 3.2.1.23), α-neuraminidase (NEUR, sialidase, EC 3.2.1.18), N-acetylaminogalacto-6-sulfate sulfatase (GALNS, EC 3.1.6.4) and serine carboxypepidase cathepsin A (EC 3.4.16.1) form a functional high molecular weight complex in the lysosomes. The major constituent of this complex is cathepsin A, the so-called “lysosomal protective protein” (PPCA). By forming a multienzyme complex, it protects the glycosidases from rapid intralysosomal proteolysis, and it is also required for the intracellular sorting and proteolytic processing of their precursors. In man, a deficiency of cathepsin A leads to a combined deficiency of β-GAL and NEUR activities, called “galactosialidosis”. Multiple mutations identified in the cathepsin A gene are the molecular basis of this lysosomal storage disease. This review describes the structural organization of the lysosomal high molecular weight multienzyme complex and the importance of the protective protein/cathepsin A in physiology and pathology.

Proline-rich protein (PRP), isolated from ovine colostrum, possesses strong immunotropic activity. The nonapeptide (Val-Glu-Ser-Tyr-Val-Pro-Leu-Phe-Pro) and the hexapeptide (Tyr-Val-Pro-Leu-Phe-Pro) PRP fragments reveled biological activity similar to that of the native protein. Seeking for analogues of PRP fragments with costrained structure, two cyclic peptides were synthesized by the solid phase method: Cys-Val-Glu-Ser-Tyr-Val-Pro-Leu-Phe-Pro-Cys and Ac-Glu-Tyr-Val-Pro-Leu-Phe-Pro-Lys-NH2. Immunotropic activity of both peptides in murine system was the same as for linear nonapeptide, whereas all three peptides were practically inactive in human system, where resistance to hydrocortisone and induction of two cytokinins IFN and TNF were used as indicators, respectively.
