Background: Oncogenic Notch1 is known to activate the NF-κB pathway in T cell acute lymphoblastic leukemia (T-ALL) and to up-regulate the transcription of Asb2α, a specificity factor for an E3 ubiquitin ligase complex that plays an important role in hematopoietic differentiation. Therefore, we hypothesize that Notch1 might regulate the NF-κB pathway through Asb2α. Methods: The study involved down-regulation of Notch1 in T-ALL cell lines (CCRFCEM cells and MOLT-4 cells) through treatment with gamma-secretase inhibitor (GSI) as well as the modulation of Asb2 in CCRF-CEM cells and MOLT-4 cells through transduction with lentivirus carrying Asb2 or Asb2-shRNA. Experiments using real-time PCR, western blot and co-immunoprecipitation were performed to evaluate the expression levels of related genes. Cell proliferation and apoptosis were measured while the expression of Asb2 was enhanced or inhibited. Results: Here, we demonstrated for the first time that Notch1 can activate the transcription of Asb2α, which then stimulates activation of NF-κB in T-ALL cells. Asb2α exerts its effects by inducing degradation and dissociation of IκBα from NF-κB in T-ALL cells. Moreover, specific suppression of Asb2α expression can promote apoptosis and inhibit proliferation of T-ALL cells. Conclusion: Notch1 modulates the NF-κB pathway through Asb2α, indicating that Asb2α inhibition is a promising option for targeted therapy against T-ALL.

An experimental study was carried out on the fatigue behaviour of multi-axial warp-knitted fabric composites. Composite samples reinforced with multi-axial warp-knitted fabric/matrix were manufactured by the vacuum-assisted resin transfer moulding method. Tensile-tensile fatigue cycling was carried out at different load levels, and S-N curves, tensile stress-strain curve and stiffness degradation of the multi-axial composite samples were obtained. Finally post-fatigue tensile tests were done at a stress level of 75%, at the stages of 1/3N and 2/3N, and the equivalent residual strength and stiffness degradation were obtained.

Dokonano oceny właściwości mechanicznych i zmęczeniowych kompozytowych dzianin wieloosiowych. Kompozyty wzmocnione dzianiną wieloosiową wytworzono przy zastosowaniu próżniowej metody formowania. Dokonano oceny zmęczenia materiału i właściwości wytrzymałościowych przy różnych poziomach obciążenia. Przedstawiono krzywe S-N, krzywą naprężenie-odkształcenie i degradację sztywności wieloosiowych próbek kompozytowych. Wykonano również testy zmęczenia materiału przy poziomie naprężenia 75%, w etapach 1/3N i 2/3N. Omówiono równoważną wytrzymałość szczątkową i degradację sztywności kompozytowych dzianin wieloosiowych.

We used the “space for time substitution” method to test the effects of reclamation history on evolution processes of soil properties and heavy metal concentrations. A total of 129 surface soil samples were collected in a reclamation zone of the Liaohe River Delta, which was divided into three reclaimed zones (1930s, 1960s, and 1990s), marsh, and intertidal flats. Soil metals (Fe, Mn, Cd, Cr, Cu, Ni, Pb, and Zn), soil particle size, pH, salinity, soil organic carbon (SOC), nitrate nitrogen (NO₃⁻-N), and available phosphorus (AP) were analyzed. Results obtained from correlation analysis and factor analysis showed that there were significant positive correlations (p<0.01) between the fine particle fractions (clay and silt) and metal concentrations. Using ANOVA, we found that soil erosion and leaching desalination had significant effects on the distributions of soil properties and heavy metal contents in short-term reclamation (1990s zone), and that sustained human activities have played an important role in long-term reclamation (1960s zone). Results of the enrichment factor (EF) and inverse distance weighting (IDW) indicated that Cd pollution had reached high pollution levels, followed by Cr pollution. Taking necessary measures is significant for sustainable utilization and management of coastal resources.

A full-length cDNA of a new serine/threonine (Ser/Thr) protein kinase gene, designated as BnSOS2 (GenBank Acc. No.AY310413), was cloned from Brassica napus by rapid amplification of cDNA ends (RACE). The fulllength cDNA of BnSOS2 was 1779 bp and contained a 1539-bp open reading frame encoding a protein of 512 amino acids. Homology analysis shows that BnSOS2 strongly resembles other Ser/Thr protein kinase genes, and that its putative protein belongs to a typical Ser/Thr kinase family. Northern blot analysis reveals that BnSOS2 is salt-inducible. Our results indicate that BnSOS2 is a new member of the plant SOS2 gene family, which may play an important role in salt tolerance of plants.
