Właściwości nowo opisanej adhezyny P-like Klebsiella badano metodami hemaglutynacyjnymi, mikroskopowo-elektronowymi i biochemicznymi. Stwierdzono amorficzny charakter adhezyny i jej swoistość receptorową zbliżoną do adhezyny PapG E.coli.

Colonisation and remaining of microorganism on mucus membrane of microorganism is tightly connected with adhesion mechanisms and determine the first step of physiological settlement of the organism or the first stage of clinically demonstrated infection. In Klebsiella rods there are known three types of fimbrial adhesins (type 1, 3 and KPF-28) and non-fimbrial adhesin CF29K. It is stated that Klebsiella strains adhesions are responsible for their adherence to the epithelial cells of both respiratory and urinary tracts and to intestine epithelium. The in vitro research affirmed Klebsiella rods adherence to protein matrix. The aim of our work was the establishment of character, receptor specifity and the appearance frequency of P-like called adhesin. The frequency of expression of P-like adhesin was estimated among 380 isolated from the patients strains on the basis of agglutinating methods. The amorphic character of P-like adhesin was proved using electron microscopy method. The isolation and purification of P-like protein with a help of affinity chromatography enabled to estimate the receptor specifity of the adhesin. The receptor specifity was established as similar to E.coli PapG adhesin.

Porównano lekowrażliwość, właściwości adhezyjne oraz wzory chromo- somalnego DNA morfologicznych wariantów kolonii pałeczek Escherichia coli. U 90,0% z nich stwierdzono różnice we wrażliwości na antybiotyki. Zdolnością adhezji do cewników moczowych różniło się 40,0% wariantów. Warianty o morfologii kolonii jasnych w wyższym odsetku niż warianty o morfologii kolonii ciemnych adherowały do wszystkich objętych badaniem cewników. Metodą PFGE u 35% wariantów wykazano 100% podobieństwa genetycznego. Pozostałe warianty były blisko ze sobą spokrewnione.

The aim of this study was comparison of the antimicrobial sensitive, adhesive properties and genetic relatedness of the morphological variants of E. coli strains. This study included 10 of E. coli strains isolated in the Clinical Microbiology Department of dr. A. Jurasz University Hospital nr 1 in 2003-2006, which have grown in two morphological variants - light and dark variant. Ninety percent of morphological variants of E. coli strains differed among themselves in antibiotic sensitivity. Forty percent of morphological variants differed among themselves in adhesion to catheters. The light-colony variants more often adhered to all used catheters. The PFGE analysis of chromosomal DNA showed that 35,0% of morphological variants (two pairs and one triple) had 100,0% relatedness, and the rest of them were closely related.

Stwierdzono, że pałeczki E. coli izolowane z moczu istotnie częściej niż izolowane z krwi wytwarzały beta-laktamazy typu ESBL i były wielolekooporne. Z kolei szczepy E. coli wyosobnione z krwi w wyższym odsetku adherowały do cewników lateksowych i cewników z polichlorku winylu.

The aim of this study was comparison of the antimicrobial sensitivity and adhesive properties of E. coli strains isolated from urine and blood samples. This study included 169 of E. coli strains isolated in the Clinical Microbiology Department of dr. A. Jurasz University Hospital in 2003-2006. E.coli strains isolated from urine samples revealed statistically higher production of Extended Spectrum Beta-Lactamases and multidrug resistance than strains isolated from blood samples. Stains isolated from the blood adhered to latex and PCV catheters more frequently when compared with strains isolated from urine.

Badano lekooporność, aktywność hemaglutynacyjną i adherencję do linii komórkowych szczepów Klebsiella izolowanych z przypadków biegunek od dzieci hospitalizowanych w oddziałach pediatrycznych we Wrocławiu i w Opolu. Stwierdzono występowanie szczepów wieloopornych, których odsetek był szczególnie wysoki w oddziale opolskim. Zdolności adhezyjne szczepów Klebsiella pochodzących z przewodu pokarmowego określono jako bardzo wysokie.

Gram negative Klebsiella bacilli present many pathogenic properties, which determine their ability to survive and rapid spreading in hospital environment. There are many factors responsible for the pathogenicity of Klebsiella strains: capsule, fimbriae, nonfimbrial adhesins, lipopolysaccharide of the cell wall and extracellular secreted exotoxins. Klebsiella strains are etiological agents of different nosocomial infections but also colonized gastrointestinal and respiratory tracts. The aim of our work were adhesive properties and antibiotic resistance of Klebsiella strains isolated from stool of hospitalized children, according to source of potential nosocomial infections - 100 Klebsiella strains from Wroclaw and 76 strains from Opole, isolated in cases of diarrhea. The resistance of this strains to different group of antibiotics, the expression of ESBL enzymes, the activity in hemagglutination and their ability to adherence to different cell lines were tested. The highest resistance of all strains to aminopenicillins was observed. The production of ESBL was highest in strains from Opole (51% strains) then in Wroclaw (9%). In both hospital units, ESBL+ strains were resistant to aminoglicosides and Cotrimoxazol but sensitive to Ciprofloxacine. Using hemagglutination method the types of fimbriae were defined. Above 90 % investigated Klebsiella strains showed the presence of fimbriae (in Wroclaw more strains simultaneously expressed fimbriae type 1 and 3, in Opole mainly fimbriae type 3). Over 70 % strains demonstrated the high level of adherence to cell lines. Only several strains showed the low level or the lack of adhesion. These results suggested that among Klebsiella strains in gastrointestinal tract were presented multiresistant strains with high ability to adherence, which may be potential source of nosocomial infections.
