Objective: The objective of the study was to determine the correct CTV-PTV margin, depending on the method used to verify the PG position. In the study, 3 methods of CBCT image superimposition were assessed as based on the location of the prostate gland (CBCT images), a single gold marker, and pubic symphysis respectively. Materials and methods: The study group consisted of 30 patients undergoing irradiation therapy at the University Hospital in Zielona Góra. The therapy was delivered using the VMAT (Volumetric Modulated Arc Therapy) protocol. CBCT image-based superimposition (prostate-based alignment) was chosen as the reference method. The uncertainty of the PG positioning method was determined and the margin to be used was determined for the CBCT-based reference method. Then, changes in the position of the prostate gland relative to these determined using the single marker and pubic symphysis-based methods were determined. The CTV-PTV margin was calculated at the root of the sum of the squares for the doubled value of method uncertainty for the CBCT image-based alignment method and the value of the difference between the locations of planned and actual isocenters as determined using the method of interest and the CBCT-based alignment method for which the total number of differences accounted for 95% of all differences. Results: The CTV-PTV margins to be used when the prostate gland is positioned using the CBCT imaging, single marker, and pubic symphysis-based methods were determined. For the CBCT-based method, the following values were obtained for the Vrt, Lng, and Lat directions respectively: 0.43 cm, 0.48 cm, 0.29 cm. For the single marker-based method, the respective values were 0.7 cm, 0.88 cm, and 0.44 cm whereas for the pubic symphysis-based method these were 0.65 cm, 0.76 cm, and 0.46 cm. Conclusions: Regardless of the method, the smallest margin values were obtained for the lateral direction, with the CBCT-based method facilitating the smallest margins to be used. The largest margins were obtained using the single marker-based alignment method.

Wykonano badania mineralogiczne i chemiczne fragmentów prostaty usuniętej podczas zabiegów operacyjnych. Stwierdzono w zmienionych i powiększonych gruczołach prostaty występowanie podwyższonych ilości niektórych pierwiastków co związane jest z tzw. ukrytą mineralizacją tkanek. Mineralizacja ta wydaje się być związana z chemizmem moczu przepływającego przez prostatę i prawdopodobnie może sprzyjać powstawania zmian chorobowych prostaty.

Mineralogical and chemical investigation of prostate glands removed after surgical operation were performed. Higher concentration of some elements were determined at changed larger prostate glands. This phenomenon is result of substitution of elements into biological structures of glans because mineral grains or crystals were not observed. Mentioned mineralization is probably the result of higher mineralization of urine flow through prostate.

The aim of this study was to investigate the influence of standardized crude aqueous Epilobium angustifolium L. extract [100 mg/kg/day, p.o.] on the expression level of SRC kinase mRNA - a representatives of non-genomics xenobiotics signaling pathway in prostate ventral lobes of testosterone-induced, castrated rats. We have shown that in all analyzed groups induced by testosterone an elevation of SRC kinase mRNA transcription was observed, in comparison to control animals (not receiving the testosterone), (p<0.05). Finasteride in rats induced by testosterone caused the strongest inhibition of SRC mRNA transcription (p<0.05). In rats receiving testosterone and the plant extract a ca. 90% decrease of mRNA level was observed vs. testosterone-induced animals (p<0.05), while in testosterone-induced animals receiving concomitantly E. angustifolium extract and finasteride the observed reduction reached 87.3% (p<0.05). We did not observed, however, any positive feedback between studied plant extract and finasteride in the inhibitory activity (p<0.05). Further experimental studies should be performed in order to the understanding the molecular basis of interactions, the efficacy and safety of tested plant extract.

Celem pracy było zbadanie wpływu standaryzowanego ekstraktu z ziela Epilobium angustifolium L. [100 mg/kg/dzień, p.o.] na poziom transkrypcji mRNA kinazy SRC - przedstawiciela androgenozależnego, niegenomowego szlaku sygnalizacji komórkowej w brzusznym płacie prostat indukowanych hormonalnie, kastrowanych szczurów. We wszystkich grupach szczurów indukowanych testosteronem zaobserowaliśmy wzrost liczby transkryptów kinazy SRC, w porównaniu do zwierząt kontrolnych (p<0.05). U szczurów indukowanych hormonalnie finasteryd wykazał najsilniejsze właściwości hamujące transkrypcję mRNA (p<0.05). U zwierząt otrzymujących testosteron wraz z wyciągiem z E. angutifolium stwierdziliśmy obniżenie liczby badanych transkryptów o blisko 90%, w porownaniu ze zwierzętami z grupy kontrolnej, indukowanymi testosteronem (p<0.05). U szczurów otrzymujących jednocześnie testosteron i ekstrakt roślinny łącznie z finasterydem redukcja poziomu mRNA kinazy SRC sięgała 87.3% (p<0.05). Nie zaobserwowaliśmy dodatniego sprzężenia w inhibicji transkrypcji badanego mRNA u szczurów otrzymujących badany ekstrakt wraz z finasterydem (p<0.05). Istnieje potrzeba przeprowadzenia dalszych badań zmierzających do wyjaśnienia molekularnego podłoża interakcji, mechanizmu działania oraz skuteczności badanego wciągu roślinnego.

The objective of the present research was to assess the usability of three diagnostic imaging modalities for canine prostate gland evaluation. The studies included 33 male dogs of various breeds. A radiographic diagnostic procedure was employed as well as dual modality ultrasound imaging, i.e. transabdominal (TAUS) and transrectal (TRUS). Radiograms were studied to evaluate the prostate gland size and localisation. Both ultrasound imaging techniques were compared regarding the imaging of various prostate parenchyma areas. It was found that a suitable USG modality should be tailored to the prostate size and localisation. The TRUS technique proved to be most helpful in visualising the gland localised in the pelvic cavity, while the TAUS scanning for the enlarged, displaced cranially prostate.

The results of several epidemiological studies have suggested that a soybean-based diet is associated with a lower risk of prostate cancer. We investigated the effect of the soy isoflavone genistein on the proliferation and contact-stimulated migration of rat prostatic carcinoma MAT-LyLu and AT-2 cell lines. Genistein almost completely inhibited the growth of both MAT-LyLu and AT-2 cells in the concentration range from 25 to 100 μM, but the addition of 1 μM genistein to the medium significantly stimulated the proliferation of both cell lines. Additionally, at concentrations above 25 μM, genistein showed a potent cytotoxic effect. However, the central finding of this study is that at physiologically relevant concentrations (1 μM and 10 μM), genistein inhibits the motility of prostate cancer cells stimulated by homo-and heterotypic contacts. These results show that at physiological concentrations, genistein exerts an inhibitory effect on the migration of prostate cancer cells and suggest that it may be one of the factors responsible for the anti-metastatic activity of plant isoflavonoids.

The aim of this work was to determine the efficacy of sclerotisation of the prostatic retention cysts by giving a 10% solution of povidone-iodine into their lumen. This procedure was considered as an alternative method for diminishing the enlarged prostate size. Diagnosis of prostatic retention cysts in 4 dogs aged from 7 to 13 years was based on physical examination, including rectal and through abdominal integuments palpation, and on endorectal sonography. Subsequently, the puncture of the solitary cysts was performed under sonographic control. After removing their fluid content, a 10% solution of povidone-iodine was introduced into the cyst lumen. During 3 weeks, the cyst size was gradually diminished and after 5 weeks, no cysts were found under sonographic control. No recurrences were noticed during 6 months of observation. Sclerotherapy appeared to be a minimally invasive technique for prostatic cysts treatment, and can be considered as noninvasive technique, and hence can be recommended to clinical practice.

Triterpene saponosides are widely distributed plant secondary metabolites characterized by relatively low systemic cytotoxicity and a range of biological activities. These include anti-inflammatory, antimicrobial, vasoprotective and antitumor properties. In particular, the ability of saponins to enhance the cytotoxicity of chemotherapeutic drugs opened perspectives for their application in combined cancer chemotherapy. Here, we used human prostate cancer DU-145 cells as an in vitro model to elucidate the synergy of the interactions between biological activities of an oleanane type 13β,28-epoxy triterpene saponoside (Lclet 4) and mitoxantrone, which is a cytostatic drug commonly used in prostate cancer therapy. No cytotoxic or pro-apoptotic effect of Lclet 4 and mitoxantrone administered at the concentrations between 0.05 and 0.1 µg/ml could be seen. In contrast, cocktails of these agents exerted synergistic pro-apoptotic effects, accompanied by the activation of the caspase 3/7 system. This effect was paralleled by attenuating effects of Lclet 4/mitoxantrone cocktails on the invasive potential, metalloproteinase expression and motility of DU-145 cells. Multifaceted and additive effects of Lclet 4 and mitoxantrone on basic cellular traits crucial for prostate cancer progression indicate that the combined application of both agents at systemically neutral concentrations may provide the basis for new promising strategies of prostate cancer chemotherapy.

The electrical conductivity and relative permittivity of chosen reproductive organs versus frequency of electromagnetic wave penetrating these organs have been presented in the paper. They have been used for determining a dielectric loss angle tangent, which constitutes the basis for defining the kind of medium in which the wave may be propagated. The change in the wavelength, the propagation speed and the conventional depth of wave's penetration into different organs were calculated.

W artykule przedstawiono przewodności właściwe i względne przenikalności elektryczne wybranych narządów płciowych, w funkcji częstotliwości wnikającej do nich fali elektromagnetycznej. Posłużyły one do wyznaczenia tangensa kąta stratności, który jest podstawą do określenia rodzaju środowiska, w jakim rozprzestrzeniać się będzie fala. Obliczono zmianę długości fali, prędkości jej rozchodzenia się oraz umowną głębokość jej wnikania do poszczególnych narządów.
