Cimicifuga racemosa, is a plant with a diverse and long history of medicinal use. Caffeic acid, bioactive compound, which often occurs with other polyphenols can influence the biological activity of this plant. The aim of our work was quantitative analysis of caffeic acid in roots and rhizomes of two varieties of C. racemosa. Analysis was performed by HPLC method. The extracts were separated on C18 reversed-phase column using mixture of methanol, water and formic acid (25:75:0.5 v/v/v) as a mobile phase. The flow rate of eluent was 1.0 ml·min-1. The obtained validation parameters such as linearity, linear regression equation and precision expressed as a relative standard deviation were adequate for quantitative determination. Caffeic acid was found in all tested extracts. The highest total amount of caffeic acid was determined in C. racemosa var. racemosa (255.3 μg·g-1) while its concentration in C. racemosa var. cordifolia was significantly lower (213.0 μg·g-1).

The aim of this work was the isolation and identification of phenolic acids from fruit and leaves of Ailanthus glandulosa Desf. The methods used in the isolation and identification of the compounds were: isolation of phenolic acids modified by Ibrahim and Towers, acidic and alkaline hydrolysis by Schmidtlein and Herrmann and identification of phenolic acids in the isolated fraction of two-dimensional thin layer chromatography (TLC) on cellulose plates. In the course of the study, the presence of phenolic acids in leaves and fruit of Ailanthus glandulosa Desf was confirmed. Overall, proportions of 15 phenolic acids were found and identified in the analyzed material. These are: gallic acid, ellagic acid, caffeic acid, gentisic acid, protocatechuic acid, p-hydroxybenzoic acid, m-hydroxybenzoic acid, ferulic acid, p-coumaric acid, syryngic acid, vanillic acid, salicylic acid, 2-hydroxy-4- methoxybenzoic acid, 2,5-dimethoxycinnamic, p-hydroxyphenylacetic acid, and 3 other compounds named A, B, C, whose identification was not possible due to the lack of suitable reference patterns. Studies have shown that leaves and fruit of the plant Ailanthus glandulosa Desf. contain a large number of phenolic acids which possess many important pharmacological activities.

An analysis of the fractions of free acids and phenolic acids liberated by way of hydrolysis in the rhizomes and herbaceous tissues of Sanguisorba officinalis L. was conducted through utilizing the 2D-TLC method. Fifteen phenolic acids were identified. Our work showed that ellagic, protocatechuic, gentisic, p-hydroxybenzoic, syringic, vannilic and ferulic acids were common in all tested fractions, and gallic and protocatechuic acids were dominant in fractions obtained from the rhizomes, while caffeic, p-coumaric, syringic, vannilic and ferulic acids were abundant in the herbaceous tissues. However, α and β - resorcylic acids were detected only in rhizome phenolic acid fractions liberated from their conjunction with sugars and alcohols. Furthermore, chlorogenic acid was present only in a free form in the herbaceous tissues, while p-hydroxyphenylacetic acid was liberated from conjunction from a herbaceous extract.

Densitometric HPTLC and HPLC have been used for quantification of p -coumaric and protocatechuic acids in an ethereal fraction from a methanolic extract of Aquilegia vulgaris L. HPLC analysis was performed on an RP-18 column with methanol-water-formic acid 25:75:0.5 ( υ/υ ) as mobile phase. Thin layer chromatography was performed on Si60 F 254 HPTLC plates with mixtures of heptane, dichloromethane, diisopropyl ether, formic acid, and water as mobile phases. Satisfactory separation of the phenolic acids was achieved by use of the multiple gradient development technique. The quantities of p -coumaric and protocatechuic acids determined by HPLC were 0.374 and 2.283 mg g -1 dry plant material, respectively; HPTLC results were somewhat higher - 0.396 and 2.584 mg g -1 , respectively. The precision of both methods, expressed as relative standard deviation, was satisfactory. The methods are useful for quality control of Aquilegia vulgaris extracts.

Red amaranth (Amaranthus cruentus) and quinoa (Chenopodium quinoa ) are pseudocereals with particularly highly regarded nutritional value. Because of the high biological significance of the flavonoids and phenolic acids in these plants, qualitative and quantitative analysis has been performed by HPLC. Extracts from the seeds of two amaranth varieties (A. cruentus v. Rawa and v. Aztek) and quinoa seeds, and their sprouts grown in natural conditions and in the dark were analyzed. The main phenolic acid found both in seeds and sprouts was gallic acid. p -Hydroxybenzoic acid, vanillic acid, p -coumaric acid, caffeic acid, and cinnamic acid were also found in the seeds and p -coumaric acid, syringic acid, and ferulic acid in the sprouts. The main flavonoid found in the sprouts was rutin. Vitexin, isovitexin, and morin were also detected in the sprouts, and orientin, vitexin, isovitexin, morin, and traces of hesperidin and neohesperidin in the seeds. Although sprouting conditions (daylight or darkness) had no effect on gallic acid content, light caused an increase in the amount of rutin and darkness resulted in increased amounts of isovitexin and vitexin.

Grzyby z gromady Basidiomycota od dawna budzą zainteresowanie ze względu na występowanie w ich owocnikach szeregu związków o uznanych właściwościach leczniczych. Obiektem przeprowadzonych badań były kultury mycelialne dwóch gatunków grzybów afylloforoidalnych występujących na terenie Polski Hydnum repandum L. oraz Sparassis crispa (Wulf.) Fr. Celem podjętych badań była jakościowa i ilościowa analiza ekstraktów otrzymanych z kultur mycelialnych pod kątem występowania związków o udokumentowanej aktywności biologicznej: kwasów fenolowych, niehalucynogennych związków indolowych oraz steroli. Do oznaczeń wykorzystano wysokosprawną chromatografię cieczową faz odwróconych (RP-HPLC). Na podstawie analizy stwierdzono w ekstraktach z otrzymanej biomasy obecność ośmiu kwasów fenolowych: galusowego, gentyzynowego, p-hydroksybenzoesowego, kawowego, kumarowego, protokatechowego, syryngowego, wanilinowego oraz kwasu cynamonowego. Ilościowo dominującym związkiem był kwas protokatechowy w ilości 6,23 μg/g s.m. (H. repandum) oraz kwas hydroksybenzoesowy w ilości 4,52 μg/g s.m. (S. crispa). Spośród związków pochodnych indolu ilościowo oznaczono: indol, serotoninę, tryptaminę i tryptofan. Całkowita ich zawartość wynosiła 1,28 μg/g s. m. (ekstrakty z H. repandum) oraz 3,07 μg/g s.m. (ekstrakty z S.crispa). Ilościowo dominującym metabolitem był tryptofan. Spośród steroli oznaczono ergosterol w biomasie z kultur in vitro S. crispa (700,87 μg/g s.m). Uzyskane wyniki wskazują, że przebadane kultury mycelialne krajowych gatunków grzybów afylloforoidalnych są zdolne do akumulacji metabolitów aktywnych biologicznie.

For a long time fungi belonging to Basidiomycota phylum have been in the center of attention because of the presence in their fruiting bodies of compounds with known therapeutic activity. Mycelial cultures of two aphyllophorales species occurring in Poland, Hydnum repandum L., and Sparassis crispa (Wulf.) Fr., were analyzed in our study. The main aim of the study was qualitative and quantitative analysis of extracts obtained from the mycelial cultures for the presence of known biologically active compounds, including phenolic acids, non-hallucinogenic indole compounds and sterols. For analyses a reversed-phase chromatography (RP-HPLC) method was used. The presence of eight phenolic acids including gallic, gentisic, p-hydroxybenzoic, caffeic, p-coumaric protocatechuic, syringic, vanillic and cinnamic acids was confirmed in the extracts obtained from the biomass. The quantitatively predominant metabolites in biomass from in vitro cultures of H. repandum and S. crispa were protocatechuic acid (6.23 μg/g DW) and p-hydroxybenzoic acid (4.52 μg/g DW). Derivatives of indole such as indole, serotonin, tryptamine and tryptophan were measured quantitatively. Their total content was estimated as 1.28 μg/g DW and 3.07 μg/g DW in H. repandum and S. crispa extracts, respectively. The major metabolite found was tryptophan. In addition, ergosterol, one of the sterols present in the biomass of in vitro cultures of S. crispa was analyzed (700.87 μg/g DW). The obtained results confirm the hypothesis that mycelial cultures of domestic species of aphyllophorales are able to accumulate biologically active metabolites.

The purpose of this study was to show relationships among the levels of essential metals (zinc, iron, sodium, magnesium, calcium and potassium) and phenolic acids (caffeic, chlorogenic, ferulic, gallic, rosmarinic and syringic) in commercial herbs (lemon balm, thyme, rosemary, mint, sage and angelica) and spices (caraway, lovage, hyssop and oregano). In the herbs higher quantities of metals and phenolic compounds were found than in spices. All plants contained high levels of calcium, potassium and rosmarinic acid, but low levels of zinc and gallic acid. By using principal component and hierarchical cluster analyses several clusters were identified grouping samples originating from a plant of a particular botanical species. Multivariate analysis has also shown that the contents of phenolic acids had a stronger impact on the scattering of herbs and spices than the metals levels. Furthermore, statistically significant correlations were found between calcium and ferulic, gallic, rosmarinic and syringic acids as well as between zinc and sodium and caffeic acid. This suggests co-operate between these biologically active constituents in metabolic processes occurring in plants.

A magistral preparation based on the sage leaf decoction or infusion is one of the most popular herbal prescription prepared in pharmacies in Lower Silesia (Poland) for gargling in the pharyngitis or mouth infections. Other components of this preparation are boric acid, aluminum(III) acetotartrate or Burow's solution, and glycerol. The study aimed to investigate the polyphenolic composition and relations between herbal and chemical ingredients that are present in this mixture in comparison with the traditional aqueous galenic forms – infusion and decoction. The analysis was performed using HPLC-DAD and ESI-MS methods. The main polyphenolic ingredients of sage leaf were rosmarinic acid and luteolin 7-O-β-glucuronide. The content of analyzed polyphenols was higher in the decoction than in the infusion. The pharmacological activity of the examined mixture results from the presence of boric acid, aluminum(III) salts and polyphenolic components of sage leaf. The study showed that in the presence of excess salts of aluminum(III) and boric acid, sage polyphenols are partially soluble in an aqueous medium (mainly phenolic acids) and complexed as precipitated sediment (especially flavonoids). The aqueous solutions of this preparation are cloudy, which suggests limited solubility of the formed complexes. Therefore, the therapeutic activity of the sage leaf magistral preparation should be associated with the presence of water soluble caffeic acid esters like rosmarinic acid. The influence of luteolin glycosides coordination complexes is not evident. The obtained results also confirm the stability of the examined formula in the conditions of cold storage (4-6 °C) within 7 days from its production.

Everlasting interest at herbal preparations as potential supporting drugs in treatment is the reason for determination of type and concentration of phenolic acids in them, as well as of their elemental contents. Due to this the objective of the work was to determine the contents of phenolic acids (ferulic, gallic and caff eic) and macroelements (K, Mg, Ca, Na) and microelements (B, Fe, Zn) in 14 plant materials used popularly in medicine. Analysis of the obtained data showed that the concentrations of ferulic, gallic and caff eic acids were in the ranges: 1.39–145.23 μg/g d. m.; 0.33–85.95 μg/g d. m. and 0.84–270.54 μg/g d. m., respectively. However, in some plant materials the level of ferulic and gallic acids was so low, that it was not possible to determine them. Macroelements were found in the ranges of concentrations: 10.60 – 38.69 mg K/g d. m.; 1.67–8.65 mg Mg/g d. m.; 5.31–39.71 mg Ca/g d. m. and 31.95–995.58 mg Na/kg d. m., whereas microelements in amounts of 24.04–67.01 mg B/kg d. m.; 31.27–260.74 mg Fe/kg d. m. and 13.93–83.35 mg Zn/kg d. m. Correlation analysis indicated the strict inter-relation between Fe level in the studied samples and the concentration of Ca, B and caff eic acid. Principal component analysis (PCA) and cluster analysis (CA) enabled identifi cation of plant materials characterized by high contents of phenolic acids and elements, as well as of such materials, in which the studied analytes were on the lowest level. Cluster analysis confi rmed also the results of correlation analysis, indicated by clusters created on the lowest bonding level, which describe the pairs of analytes with high values of the correlation coeffi cients.

CEL PRACY Nieustające zainteresowanie preparatami roślinnymi jako potencjalnymi lekami wspomagającymi przyczynia się do określenia w nich rodzaju i zawartości substancji czynnych oraz ich składu pierwiastkowego. Celem pracy było oznaczenie zawartości kwasów fenolowych (ferulowego, galusowego, kawowego) oraz makroelementów (K, Mg, Ca, Na) i mikroelementów (B, Fe, Zn) w 14 surowcach roślinnych powszechnie stosowanych w lecznictwie. WYNIKI Stwierdzono że zawartość kwasów: ferulowego, galusowego i kawowego mieściła się w zakresach, odpowiednio: 1,39–145,23 μg/g s. m.; 0,33–85,95 μg/g s.m. i 0,84–270,54 μg/g s.m., ale w przypadku niektórych surowców zawartość kwasów ferulowego i galusowego była tak niska, że nie udało się ich oznaczyć. Makroelementy oznaczono w zakresie stężeń 10,60–38,69 mg K/g s.m.; 1,67–8,65 mg Mg/g s.m.; 5,31–39,71 mg Ca/g s. m. i 31,95–995,58 mg Na/kg s.m., natomiast mikroelementy w ilości 24,04–67,01 mg B/kg s.m.; 31,27–260,74 mg Fe/kg s.m. i 13,93–83,35 mg Zn/kg s. m. WNIOSKI Analiza korelacji wskazała na ścisłą współzależność między stężeniem Fe w badanych próbkach a zawartością Ca, B i kwasu kawowego. Analiza głównych składowych (PCA) i analiza skupień (CA) umożliwiły identyfikację surowców roślinnych charakteryzujących się wysoką zawartością fenolokwasów i pierwiastków oraz surowców, w których ilości oznaczanych analitów były na najniższym poziomie. Analiza skupień potwierdziła również wyniki analizy korelacji, na co wskazują skupienia utworzone na najniższym poziomie wiązania, opisujące pary analitów, które charakteryzują wysokie wartości współczynników korelacji.
