Thermolysins constitute a family of secreted bacterial metalloproteases expressed, among others, by several pathogens. Strains of Staphylococcus pseudintermedius isolated from diseased dogs and judged as protease-positive, by skim milk agar plate culture, were investigated for protease content. No proteolytic activity was detected when the bacteria were grown in regular liquid media. Unexpectedly, supplementation of the medium with calcium ions resulted in expression of a metalloprotease and profound changes in the profile of extracellular proteins. On the basis of homology to other staphylococcal metalloproteases, the nucleotide sequence of the gene encoding this protease (Pst) and its flanking regions was determined. The full-length pst codes for a protein with an open reading frame of 505 amino acids. The internal region contains the HEXXH catalytic domain that is conserved in members of the thermolysin family. Regardless of the presence of calcium in the medium, the expression of the protease gene was of the same intensity. This suggests that regulation of the metalloprotease production by calcium ions is at a post-transcriptional level. Isolates of S. pseudintermedius exhibit a proteolytic phenotype due to the metalloprotease expression, however only in presence of calcium ions, which most probably stabilize the structure of the protease.

The article is devoted to establishing the influence of multifunctional environmentally friendly growth-regulating preparations on the phytosanitary state of agrocenoses of various types of wheat varieties and their productivity under non-irrigated conditions of the steppe zone of Ukraine. Field studies were conducted over 2016–2020 in two agroclimatic points of the steppe zone: such as in the experimental field of the Kherson State Agrarian and Economic University (GPS: 46.743447, 32.481064 Kherson, Ukraine – point 1) and the land use of the farm "Svetlana" (GPS: 47.635522, 32.099772 Vossiyatskoye Village, Mykolaiv region, Ukraine-point 2) under non-irrigated conditions. The implementation of the scientific research program was carried out by laying three – factor experiments in these agroecological points, where Factor A was the varieties of winter wheat – Khersonska 99, Kirena, Askaniyska, Mudrist, Clarissa, Khutoryanka; variants of Factor B: growth-regulating preparations – VuksalMicroplant, Helafit Combi, Phytomare, without cultivation (control); variants of factor C: terms of sowing winter wheat – 10.09, 20.09, 30.09 and 10.10. Studies established that the use of multifunctional growth-regulating preparations helped to reduce the level of plant damage by pathogenic microflora at all sowing periods and varieties of winter wheat. Phytomare and Helafit Combi were the most effective in this respect, under various growing conditions. In most cases, they reduced the degree of damage to winter wheat plants by pathogenic microflora by 40–50% or more. All applied growth regulators increased the mass of grain from the ear and the mass of 1000 grains at different sowing times. It was found that for five years of field research, the greatest increase in yield under different research conditions and different varieties was shown by the Helafit Combi preparation. When it was applied in the experimental field of the Kherson State Agrarian and Economic University, the yield was additionally formed from 0.22 up to 0.5 t/ha, in the field of FG "Svetlana", respectively, it was formed 0.14 up to 0.36 t/ha.

Plants respond to infection by accumulating many compounds some of which may function in disease resistance. These include: phytoalexins, antifungal proteins, chitinases, glucanases, esterases, proteaes, phospholipases, lipoxygenases, ribonucleases, peroxidases, phenoloxidases, lignin, callose, hydroxyproline and glycine-rich glycoproteins, phenolic cross-linked polysachcarides, melanin-like pigments, salicylic acid, jasmonic acid, ethylene, peptides, oligosaccharides, hydrogen peroxide and active oxygen species. Though specific avirulence genes, elicitors and elicitor receptors have been reported, the production of defense-related compounds is nonspecific and can be elicited by pathogens, pathogen products and many organics and inorganics. The molecular implications of this specificity/nonspecificity and their significance to disease resistance and practical disease control will be discussed.

Reports of gastrointestinal disease outbreaks caused by pathogenic protozoans in rural areas are still small in numbers. This study has evaluated, at global level, the parasitological contamination by Cryptosporidium spp. and Giardia spp., of water resources used as sources for human consumption in rural areas, spanning from 1994 to 2022. For such, a systematic review was conducted, following the PRISMA guidelines, applying a descriptive and statistical analysis to the data. A total of 29 eligible articles reported contamination in 90 different springs spread throughout six continents. It was possible to see a greater tendency of giardiasis outbreak occurrences in water coming from surface springs in countries with higher HDI (above 0.8), justified by the lack of data in countries with other HDIs. The water sources intended for human supply in rural areas are susceptible to parasitological contamination, especially in Asia and Oceania. In spite of the fact that there have been no reports regarding giardiasis outbreaks in the reviewed studies, it is important to notice that the concentration of pathogens may have been greater due to the low recovery of the methods of detection. Therefore, it is advisable to conduct more studies about the contamination of water resources in rural areas, as well as the adoption of prevention measures regarding waterborne diseases, such as boiling water.

W artykule przedstawiono korzyści oraz mankamenty stosowania preparatów srebrowych służących do zabezpieczania systemów wentylacyjnych przed rozwojem szkodliwych dla zdrowia mikroorganizmów. Dodatkowo wskazano sposoby obniżenia ryzyka rozwoju patogenów, zwłaszcza w miejscach gdzie ich obecność przyczynia się do pogorszenia zdrowia lub pogarsza rachunek ekonomiczny użytkownika systemu.

The article presents the advantages and drawbacks of the use of silver preparations for protecting ventilation systems against the development of harmful microorganisms. In addition show ways to reduce risk of pathogens grovth have been indicated, especially in places where their presence contributes to the deterioration of health or worsens the economic calculation of the system user.

The Shiga toxin-producing Escherichia coli (STEC) strains are currently considered important emerging pathogens threatening public health. Among Shiga toxin-producing Escherichia coli, E. coli O157:H7 strains have emerged as important human pathogens. This study was conducted to determine the presence of Escherichia coli O157 and O157:H7 in raw milk samples and Van herby cheese samples. For this purpose, 100 samples of raw milk were collected and 100 samples of herby cheese sold for consumption in Van province in Turkey were obtained from grocers and markets in order to detect the presence of Escherichia coli O157 and O157:H7. The method of E. coli O157 and O157:H7 isolation proposed by the Food and Drug Administration (FDA) was used. E. coli O157 in raw milk and herby cheese samples was found in 11% and 6% of samples respectively, and E. coli O157:H7 was found in 2% of herby cheese samples. No E. coli O157:H7 was detected in raw milk samples. This study showed that raw milk was contaminated with E. coli O157 and herby cheese was contaminated with both E. coli Ol 57 and E. coli Ol57:H7; therefore, herby cheese poses a serious risk to public health.

Similary to other fungal species of the genus Alternaria, Alternaria radicina is a major pathogen that infects both the aboveground and underground parts of carrot plants. Changes in the concentrations of phenolic compounds are observed in infected carrot plants. Carrot seedlings were inoculated with the most pathogenic isolates of Alternaria radicina selected in a laboratory test. A phytopathological analysis was performed to determine the health status of carrot plants. The concentrations of phenolic acids in petioles were determined four weeks after inoculation with A. radicina and at harvest. The results of a greenhouse experiment revealed more severe disease symptoms on carrot plants cv. Koral inoculated with A. radicina isolates, compared with cv. Bolero. The content of the predominant phenolic acid (chlorogenic acid) was found to decrease in the leaf stalks of carrots cv. Koral inoculated with A. radicina. A statistically non-significant increase in chlorogenic acid levels was noted in the leaf stalks of carrots cv. Bolero in the experimental and control groups.

Alternaria radicina, obok innych gatunków grzybów, jest groźnym patogenem części nadziemnej i podziemnej marchwi. Infekcje powodują zmiany, m.in. zawartości związków fenolowych. W teście laboratoryjnym wybrano najbardziej patogeniczne izolaty Alternaria radicina, którymi inokulowano siewki marchwi. Przeprowadzono ocena fitopatologiczną zdrowotności marchwi. Analizowano zawartość fenolokwasów w ogonkach liściowych w terminie 4 tygodni po inokulacji grzybem i podczas zbioru. Wyniki doświadczenia w hali wegetacyjnej wskazują na silniejsze objawy choroby wskutek inokulacji izolatami Alternaria radicina na odmianie Koral niż na odmianie Bolero. W ogonkach marchwi odmiany Koral inokulowanych A. radicina stwierdzono zmniejszenie zawartości głównego fenolokwasu (kwasu chlorogenowego). Wzrost zawartości kwasu chlorogenowego w ogonkach odmiany Bolero w grupie eksperymentalnej i kontrolnej nie był istotny.

Sclerotinia sclerotiorum (Lib.) de Bary, the causal agent of Sclerotinia stem rot, is one of the most important pathogens of Brassica napus L. in northern Iran. In this study, 13 mycelial compatibility groups (MCGs) of the fungus were identified among 31 isolates sampled from four regions of Mazandaran province, Iran. Effective fungicides are useful in the integrated management of the disease. So, the effect of tebuconazole, propiconazole, cyproconazole, and Rovral-TS at five doses (0.0001, 0.001, 0.01, 0.1, and 1 ppm) was studied on the growth inhibition of S. sclerotiorum as in vitro tests. Maximum inhibition (100%) of S. sclerotiorum mycelial growth was obtained by the highest dose (1 ppm) of all tested fungicides, as well as by the doses of 0.1 and 0.01 ppm of propiconazole, cyproconazole, and tebuconazole. In this investigation, the reaction of S. sclerotiorum isolates belonging to different MCGs was evaluated against tebuconazole, propiconazole, cyproconazole, and Rovral-TS at their EC50 ranges. The results revealed that there was high variation of S. sclerotiorum MCGs against different fungicides. The inhibition percentage varied between 4.29% and 71.72%.

Apple scar skin viroid (ASSVd) is a serious pathogen of apple fruits that results in severe loss in apple production. Up to nowadays, many ASSVd management options are applied to resist the disease, but the diserable resulits are not achieved. Therefore, this study was conducted in 2010–2012 at experimental field of Penglai City, Shandong Province, China (E 120°57’22’’, N 370°38’33’’) to investigate whether arbuscular mycorrhizal (AM) Glomus versiforme protects Red Fuji apple trees (Malus × domestica Borkh) against apple scar skin viroid. Red Fuji apple trees were inoculated with Glomus versiforme and then potential protection mechanism was explored and compared to noninoculated plants. The results showed that inoculation with Glomus versiforme significantly increased root length colonization rate and clearly decreased the percentage of disease severity of apple scar skin disease. Compared to non-inoculated plants, Glomus versiforme obviously enhanced total nitrogen and phosphorus concentrations in leaves. Root colonization by Glomus versiforme induced an increase in defense-related enzyme activities in fruits, such as the enhanced activities of catalase, ascorbate peroxidase, chitinase and glucanase. Significant differences in acid content of fruit and fruit yield were observed as apple roots were colonized by Glomus versiforme. It is therefore concluded that Glomus versiforme can be regarded as a biocontrol agent to protect apple trees against the infection with ASSVd.

Wiroid bliznowatości skórki jabłek (ASSVd) jest poważnym patogenem jabłek powodującym znaczne straty w ich produkcji. Obecnie stosuje się wiele opcji kontrolowania ASSVd w celu uzyskania odporności na chorobę, ale nie osiągnięto jeszcze pożądanych rezultatów. Niniejsze badanie przeprowadzono w latach 2010–2012 na polu doświadczalnym miasta Penglai, prowincja Shandong w Chinach (E 120°57’22”, N 370°38’33”) w celu zbadania, czy mikoryza abuskularna (AM) Glomus versiforme chroni jabłonie Red Fuji (Malus × domestica Borkh) przed wiroidem bliznowatości skórki jabłek. Jabłonie Red Fuji zaszczepiono Glomus versiforme a następnie badano mechanizm potencjalnej ochrony oraz porównano go z roślinami nieszczepionymi. Wykazano, że inokulacja Glomus versiforme istotnie zwiększyła szybkość kolonizacji korzeni oraz wyrażnie zmniejszyła procent ostrej choroby bliznowatości skórki jabłek. W porównaniu z roślinami nieszczepionymi, Glomus versiforme wyrażnie wzmagał stężenia całkowitego azotu i fosforu w liściach. Kolonizacja korzeni przez Glomus versiforme powodowała wzrost aktywności enzymatycznej związanej z mechanizmem obronnym, np. wzmożoną aktywność katalazy, peroksydazy askorbinianowej, chitynazy oraz glukanazy. Zaobserwowano istotne różnice w zawartości kwasów oraz plonie owoców w miarę kolonizowania korzeni jabłoni przez Glomus versiforme. Można więc wyciągnąć wniosek, że Glomus versiforme może być uważany za czynnik bio-kontroli chroniący jabłonie przed zakażeniem ASSVd.

Since C. pseudotuberculosis is a facultative intracellular pathogen the aim of this study was focused on evaluating mechanisms that allowed these bacteria to survive in macrophages and determining their influence on induction of cell death. The influence of Corynebacteria on the programmed cell death of macrophages was determined on the basis of induction the autophagy and apoptosis in the cultures of murine macrophage cell lines J774 infected with bacteria. Corynebacterium pseudotuberculosis strains could survive within macrophages more than 48 hours. During that time bacteria were released as a result of the process that lead to death of phagocytes. This property varied among studied strains. There was no increase of micro- tubule-associated protein I light chain 3 (MAP I LC3) activity in macrophages infected with examined strains comparing with uninfected cultures and cultures treated with autophagy inducer (rapamycin) that served as negative and positive controls, respectively. The study with confocal microscopy did not show the increasing of caspase-3 activity in the infected macrophages and their nucleus did not reveal the fragmentation.

Powszechnie występujące w przyrodzie grzyby z rodzaju Trichoderma wykazują silne działanie antagonistyczne wobec grzybów powodujących choroby roślin. Mogą hamować a nawet zwalczać, powodowane przez grzyby, infekcje roślin. Grzyby rodzaju Fusarium stanowią poważny problem w rolnictwie, szczególnie dotkliwy dla upraw zbożowych. Fusarium spp. Są sprawcami wielu chorób powodując m.in. fuzaryjną zgorzel podstawy źdźbła i korzeni, fuzariozę liści i kłosów. W celu porównania skuteczności antagonistycznego oddziaływania Trichoderma viride na patogeniczne grzyby rodzaju Fusarium przeprowadzono badania in vitro wykorzystując pięć gatunków Fusarium (F. avenaceum, F. culmorum, F. graminearum, F. oxysporum, F. poae) wyizolowanych z pszenicy. Dodatkowo wprowadzono do doświadczenia trzy biopreparaty: Biochikol 020 PC, Bioczos BR i Biosept 33 SL oraz fungicyd Amistar 250 SC. Testowane grzyby wyszczepiano parami (Trichoderma viride/Fusarium spp.) na sterylną pożywkę PDA, na płytkach Petriego. W trakcie trwania doświadczenia stwierdzono antagonistyczny wpływ T. viride na kultury Fusarium spp. oraz zróżnicowane oddziaływanie biopreparatów na testowane grzyby.

Commonly occurring in nature fungi of the genus Trichoderma have strong antagonistic activity against fungi that cause plant diseases. They can inhibit or even fight fungal infections of plants. However fungi of the genus Fusarium are a serious problem in agriculture, especially difficult for cereal crops. Fusarium spp. causes many diseases of stem base, leaves and ears. In order to compare the effectiveness of the antagonistic effects of fungus Trichoderma viride on Fusarium pathogens studied in vitro using five species of Fusarium (F. avenaceum, F. culmorum, F. graminearum, F. oxysporum, F. poae) isolated from wheat and Trichoderma viride isolate. In addition, introduced to experiment three biopreparations: Biochikol 020 PC, Bioczos BR and Biosept 33 SL and the fungicide Amistar 250 SC. Tested fungi were inoculated in pairs (T. viride/Fusarium spp.) on sterile PDA medium, in Petri dishes. During the experiment antagonistic effect of T. viride on cultures of Fusarium spp. was observed. Also was found different impact of biopreparations on tested fungi.

Between 2000 and 2006 the sum of ciprofloxacin and folic acid antagonists prescriptions to Bavarian (South-eastern Germany) outpatients stayed constant. However, prescription numbers of ciprofloxacin increased while those of folic acid antagonists decreased suggesting an apparent shift in the treatment of urinary infections toward ciprofloxacin. During the observation period the proportion of E. cbti resistant against ciprofloxacin increased from 5% to 10% while that against co-trimoxazole increased from 21 % to 27%. The proportion of E. coli simultaneously exhibiting resistance to ciprofloxacin and co-trimoxazole increased from 3.9% to 8.5%. A leading influence of ciprofloxacin application for these developments is discussed.
