Exposure to giant hogweed brings about the risk of serious skin damage, usually in the form of phytophotodermatitis. Initially, skin changes are signaled by a burning sensation, followed by a vesiculobullous rash and long-term hyperpigmentation. Usually, skin disorders improve after the application of topical and oral corticosteroids. In extremely rare instances, full skin thickness burns or epidermal necrosis occur. The study presents a case of occupational exposure to hogweed, which resulted in extensive skin lesions leading to the disablement of a 27-year-old man. The principles of procedure to be followed when contact with giant hogweed is an occupational hazard are also outlined.

Objective: Cervico-facial arteriovenous malformations (AVMs) are complex and rare vascular lesions, and present in 0.1% of the population. Of traumatic or congenital origin, they are characterized by variable growth, and their complications can be disfiguring and potentially fatal. The treatment of choice is embolization followed by surgery if necessary. The main complications are recurrence and postoperative bleeding. Case report: We report the rare case of a 59-year-old female patient who underwent embolization of a right upper labial and jugal AVM, followed by complete necrosis of the right hemi-palatal mucosa associated with dental mobility and pain. Follow-up at 6 months showed complete reepithelialisation of the palate.  Conclusions: Soft tissue necrosis after AVM embolization is a rare event and is more commonly described after embolization for epistaxis. The evolution is generally favourable within a few weeks.

Content available remote Cytotoxicity of β-carotene cleavage products and its prevention by antioxidants

When we investigated the genotoxicity of β-carotene cleavage products (CPs) in primary rat hepatocytes stimulated to proliferate, we observed dose-dependent increases of chromosomal aberrations, sister chromatid exchanges and micronuclei. In contrast to other genotoxic substances, however, this increased genotoxicity was not accompanied by increased cytotoxicity. As a consequence we observed metaphases showing massive chromosomal damage, indicating inhibition of apoptosis by CPs enabling these cells to proceed in the cell cycle. Since proliferative stimulation by growth factors may support this effect, the in vitro toxicological effects of CPs were studied on proliferatively quiescent primary rat hepatocytes. A significant increase of both apoptosis and necrosis was found. Supplementation with antioxidants did not significantly lower the level of apoptosis, while the level of necrosis was significantly reduced by Trolox and N-acetylcysteine at all concentrations tested as well as ascorbic acid (50 µM) and a combination of Trolox (50 µM) and ascorbic acid (50 µM). These observations indicate that a) the cytotoxic potential in combination with the genotoxic potential of CPs may promote the initiation of cells due to compensatory cell division in exposed tissues and may aggravate inflammatory processes under chronic exposure, and b) the applied antioxidants may protect from cytotoxicity primarily via the detoxification of aldehydic β-carotene cleavage products.

Known since a long time but relatively neglected, the venous vasa vasorum form a tiny network to irrigate and drainer the wall. Actually it is re-realized that, just like any tissues, the vein is a living tissue and the vasa vasorum play a key role in the maintenance of its homeostasis. Under the effects of intraluminal pressure, the Young's modulus (E) of the vein following an empirical relationship (Log E)2= - ('alpha' Log(p) + 'beta'), can vary in a very large range (a factor of 100) en even in physiological conditions. In this work, we simulated the deformation of venous vasa vasorum by finite element method. Thus it can be suggested that, at first, a permanent venous hypertension provoquer a local modification of the wallís mechanical properties (decrease of E) to favoriser a large deformation which will lead to a decrease of the irrigation, and secondly, with the decreased irrigation, the wall will loose its elasticity, become rigidified and keep its large deformation. A vicious cycle could be thus created.

Urban plants play a significant role in shaping the microclimate of the modern city, in addition to the recreational and aesthetic functions. Climate change and sharp changes in temperature affect the plant growth and development, so the question of studying the adaptive potential of the plant range in cities to temperature variability is relevant and important. Researchers and scientists around the world are studying the impact of biotic and abiotic factors on plants, but the variability of the plant organisms in the urban ecosystem is still unexplored. The data from the analysis of the frequency of occurrence plants in Kyiv green spaces show that Tilia cordata Mill., Aesculus hippocastanum L., Spiraea×vanhouttei (Briot) Zabel. and Carpinus betulus L. are most represented among the researched species. The article presents the results of the assessment of heat resistance by using the method of Matskov (1976) of 13 ornamental woody and shrubby species. Besides, the plants were separated into groups of tolerance to high temperatures. It was established that the T. cordata plants are characterized by the highest indicators of heat tolerance and they can be recommended for the creation of open landscapes. The A. hippocastanum, Catalpa bignonioides Walt., S. vanhouttei and Forsythia europaea Degen & Bald. plants are characterized by ‘moderate tolerance’ indicators to high temperature stress, therefore they can be recommended for the creation of semi-open/open landscape types. Consequently, for the formation of semi-open landscapes, using the species Platanus occidentalis L., Quercus robur L., Q. rubra, Syringa vulgaris L., Berberis thunbergii DC. and Ligustrum vulgare L. which were assessed as ‘moderately sensitive’, is recommended. The leaves of the Carpinus betulus L. and Ribes aureum Pursh. plants were most vulnerable to high temperature stress; therefore, the species are recommended for the formation of closed landscape types. Despite the comparative classification of ornamental plants in terms of heat-resistance, many issues remain unstudied and need to be clarified in terms of ecology, physiology, biochemistry and phytopathology for the plants of urban green spaces.

The effects of inhibitors of plasma membrane NADPH oxidase on menadione-induced cell injury processes were studied using human osteosarcoma 143B cells. The intracellular level of superoxide in the cells treated with menadione for 6 h reached a maximum followed by an abrupt decrease. The population of apoptotic cells detected by Annexin V and propidium iodide double staining also reached its maximum at 6 h of menadione-treatment while that of necrotic cells increased continuously reaching 90% of the total population at 9 h of the treatment. Pretreatment of the cells with inhibitors of NADPH oxidase, including diphenyliodonium chloride, apocynin, N-vanillylnonanamide and staurosporine was effective in lowering the menadione-induced elevations of superoxide, and also in the suppression of the switch of the cell death mode from apoptosis to necrosis in menadione-treated cells except for the case of staurosporine. These results strongly suggest that superoxide generated by NADPH oxidase, besides that generated by the mitochondria, may contribute to the remarkable increase in the intracellular level of superoxide in the cells treated with menadione for 6 h resulting in the switch from apoptosis to necrosis, although a direct evidence of the presence of active and inactive forms of NADPH oxidase in control and menadione-treated 143B cells is lacking at present.

Children born with a low birth weight (below 2500g) exhibit a slower rate of development, and a greater tendency towards morbidity and mortality, together with a deficit of weight and height. One reason could be an increase in the level of cell elimination by apoptosis. The aim of this study was to evaluate and compare the incidence of apoptotic and necrotic (dead) cells in cultures of peripheral blood lymphocytes obtained from children born with a low birth weight and from children with a normal birth weight. Peripheral blood lymphocytes were obtained by venipuncture (10 ml) and isolated using the density gradient centrifugation method. The lymphocytes were cultured for 48 h in a culture medium containing low concentrations of fetal calf serum. A comparison study was performed between low birth weight children and normal birth weight children and the susceptibility of their lymphocytes to apoptosis and to necrosis in serum-deficient feeding culture conditions. The amount of apoptotic cells and the percentage of dead cells were significantly higher in cultures of lymphocytes obtained from low birth weight children than in cultures from normal birth weight children. The two estimated parameters inversely correlated with the concentration of fetal calf serum in the culture medium. Pulsed field gel electrophoresis showed increased DNA degradation patterns in the cultures of lymphocytes obtained from low birth weight children. Our results should be perceived as an indication that, under worse feeding conditions, the elimination of cells by apoptosis and by necrosis is significantly higher for lymphocytes of low birth weight children than for those of normal birth weight children. The enhanced elimination of lymphocytes is related to a greater susceptibility to infections, especially of the respiratory tract, as established in the retrospective analysis of the anamneses of the examined group of low birth weight children.

Multiwalled carbon nanotubes (MWCNTs) have attracted huge attention due to their multifunctionality. Their unique properties allows for covalent and noncovalent modifi cations. The most simple method for functionalization of carbon nanotubes is their decoration with the oxygen containing moieties which can be further simultaneously functionalized for design of new class carriers for targeting and imaging. Here, we present methodology for chopping nanotubes, characterization of MWCNTs, the effect of size on the biocompatibility in culture of L929 mouse fi broblasts using WST-1, LDH and apoptosis assays. The analysis provides the optimal carbon nanotubes length and concentration which can be used for functionalization in order to minimize the effect of the secondary agglomeration when interacting with cells.

Phomopsis diachenii was isolated from caraway cultivars Konczewicki, firstly in 2006 and next in 2007. Single cultures were obtained from the roots and the stem base of eight six-week-old seedlings and from the stems of two plants with symptoms of necrosis, in the second year of planting. This fungus was isolated from the plant parts superficially disinfected on malt agar medium with an addition 0.01% of streptomycin. The identification of the species was made on PDA medium. The biotic interactions between P. diachenii and S. carvi and other species of phyllosphere fungi of caraway were studied. Interactions among the fungi, i.e. between P. diachenii and one of the fungi representing the studied community, were examined using the biotic series method. The biotic effects of the fungi in dual cultures were evaluated after 10 and 20 days of common growth and were expressed as the individual biotic effect (IBE). It was shown that P. diachenii is a weak competitor because its growth was limited by numerous species of phyllosphere fungi. The obtained results indicated the dominance of biotic activity of P. diachenii over that of S. carvi. It is possible that P. diachenii has a greater ability to survive in the phyllosphere fungal community than S. carvi, causing septoriosis of caraway.

Phomopsis diachenii wyizolowano po raz pierwszy w Polsce z roślin kminku zwyczajnego, odmiany Konczewicki w 2006 roku, a następnie w 2007 r. Pojedyncze kultury otrzymano z korzeni i szyjki korzeniowej ośmiu 6-tygodniowych siewek oraz z łodyg dwu roślin w drugim roku uprawy z objawami nekrozy. Grzyb wyizolowano z odkażonych powierzchniowo części roślin na pożywce maltozowej z dodatkiem 0,01% streptomycyny. Identyfikację do gatunku prowadzono na pożywce PDA. Przebadano również biotyczne oddziaływanie pomiędzy P. diachenii i Septoria carvi oraz innymi gatunkami grzybów fylosferowych z kminku zwyczajnego. Na pożywce PDA w szalkach Petriego zakładano hodowle dwuorganizmowe, tj. składające się z P. diachenii i jednego z grzybów reprezentujących badane zbiorowisko. Biotyczne oddziaływanie grzybów w kulturach dwuorganizmowych oceniono po 10 i 20 dniach wspólnego wzrostu i wyrażono jednostkowym efektem biotycznym IBE. Wykazano, że P. diachenii posiada niewielkie właściwości konkurencyjne. Jego wzrost ograniczają liczne gatunki szybko rosnących grzybów fylosferowych. Uzyskane wyniki wskazały na przewagę właściwości biotycznych P. diachenii nad Septoria carvi. Można wnioskować, że P. diachenii ma większe możliwości utrzymania się w zbiorowiskach grzybów fyllosferowych niż S. carvi, powodujący septoriozę kminku.

The effects of thiamine (vitamin B1) on the level of spontaneous or radiation-induced genetic changes in human lymphocytes in vitro were studied. Cultured lymphocytes were exposed to increasing concentrations of thiamine (0-500 μg/ml) and irradiated with X-rays. The DNA damage was estimated as the frequency of micronuclei and apoptotic or necrotic morphological changes in fixed cells. The results show that thiamine alone did not induce genetic changes. A significant decrease in the fraction of apoptotic and necrotic cells was observed in lymphocytes irradiated in the presence of vitamin B1 at concentrations between 1-100 μg/ml compared to those irradiated in the absence of thiamine. Vitamin B1 at 1 and 10 μg/ml decreased also the extent of radiation-induced formation of micronuclei. Vitamin B1 had no effect on radiation-induced cytotoxicity as measured by nuclear division index. The results indicate that vitamin B1 protects human cells from radiation-induced genetic changes.

Two water soluble porphyrins: meso-tetra-4-N-methylpyridyl-porphyrin iodide (P1) and 5,10-di-(4-acetamidophenyl)-15,20-di-(4-N-methylpyridyl) porphyrin (P2) were synthesised and evaluated in respect to their photochemical and photophysical properties as well as biological activity. Cytotoxic and phototoxic effects were evaluated in human malignant melanoma Me45 line using clonogenic assay, cytological study of micronuclei, apoptosis and necrosis frequency and inhibition of growth of megacolonies. Both porphyrins were characterised by high UV and low visible light absorptions. Dark toxicity measured on the basis of the clonogenic assay and inhibition of megacolony growth area indicated that P1 was non-toxic at concentrations up to 50 μg/ml (42.14 μM) and P2 at concentrations up to 20 μg/ml (16.86 μM). The photodynamic effect induced by red light above 630 nm indicated that both porphyrins were able to inhibit growth of melanoma megacolonies at non-toxic concentrations. Cytologic examination showed that the predominant mode of cell death was necrosis.

Microsomal glutathione transferase 1 (MGST1) possesses glutathione transferase and peroxidase activities and is active in biotransformation of xenobiotics and in defense against oxidative stress. To assess MGST1 role in the development and functioning of PC12 cells, we constructed a cell line with reduced MGST1 (PC12_M). Real-time PCR and immunoblot assays showed MGST1 expression lowered to 60 % and immunocytochemical analyses demonstrated an altered concentration and distribution of the enzyme. PC12_M cells revealed a larger tendency to grow in clusters, weaker adhesion, irregular shape of bodies, short neurite outgrowth and higher percentage of necrotic cells (34 %). The total GSTs activity determined with non-specific substrate CDNB (1-chloro-2,4-dinitrobenzene) decreased by 15-20 %, whereas that with DCNB (2,4-dichloro-1-nitrobenzene), a substrate more specific for cytosolic GSTs, was similar to the one in control cells. This suggests that reduction of MGST1 cannot be compensated by other glutathione transferases. In PC12_M cells the total glutathione content was higher by 15-20 %, whereas the GSSG/GSH ratio was lower than in control cells. Moreover, the laminin-dependent migration rate was much faster in control cells than in PC12_M, suggesting some alterations in the metastatic potential of the line with suppressed MGST1. The amount of MAP kinases (p38, JNK, ERK1/2) was elevated in PC12_M cells but their phosphorylation level declined. Microarray analysis showed changed expression of several genes, which may be linked with differentiation and necrosis of PC12_M cells. Our data suggest that MGST1 could be an important regulator of PC12 cells development and might have significant effects on cell growth and proliferation, probably through altered expression of genes with different biological function.
