Problem of Post-operative infections of implant materials caused by bacterial adhesion to their surfaces is very serious. Enhancement of antibacterial properties is potentially beneficial for biomaterials value. Therefore, the metallic and metallic oxide nanoparticles attract particular attention as antimicrobial factors. The aim of this work was to create nanotubular (NT) oxide layers on Ti with the addition of ZnO nanoparticles, designed for antibacterial biomedical coatings. Antimicrobial activities of titanium, TiO2 NT and ZnO/TiO2 NT surfaces were evaluated against bacterial strain typical for orthopaedic infections: S. epidermidis. TiO2 NT alone killed the free bacterial cells significantly but promoted their adhesion to the surfaces. The presence of moderate amount of ZnO nanoparticles significantly reduced the S. epidermidis cells adhesion and viability of bacterial cells in contact with modified surfaces. However, higher amount of loaded nanoZnO showed the reduced antimicrobial properties than the medium amount, suggesting the overdose effect.

L-forms of S. epidermidis were induced at 35°C with the use of an L-form medium with penicillin. The aim of this study was to evaluate the frequency of L-form induction and demonstrate whether the origin of the clinical strains affects the frequency of L-forms induction, as well as to study whether the time of action of the antibiotic has an influence on frequency of L-form induction.

Oceniono zdolność wytwarzania śluzu przez 32 szczepy Staphylococcus aureus і 32 szczepy Staphylococcus epidermidis wykorzystując metody: ocenę morfologii kolonii na podłożu z czerwienią Kongo (CRA), z zastosowaniem roztworu safraniny (CT) i z użyciem metody spektrofotometrycznej z zastosowaniem czerwienieni Kongo (SC). Nie wykazano istotnych różnic w ocenie wytwarzania śluzu przez S. aureus i S. epidermidis przy wykorzystaniu metody CT i SC (p > 0,05), w przeciwieństwie do metody CRA (p < 0,0004 i p < 0,00001).

In this article, slime production of Staphylococcus aureus and Staphylococcus epidermidis strains from infective skin lesions was evaluated by three different methods: Congo red agar method (CRA), Christensen tube method (CT) and spectrophotometric method (SC). All strains by CT method interpreted as negative (dark-claret or red colonies of the surface). 12 (37,5%) strains of S. aureus, 16 (50,0%) strains of S. epidermidis produced slime as shown by CT method, 6 (18,7%) strains of S. aureus, 8 ( 25,0%) strains of S. epidermidis by SC method. They also found a correlation of slime production by CT and SC method (p > 0,05).

Quaternary ammonium compounds are broad-spectrum bacteriocides widely used as antiseptics, disinfection and preservation agents. The aim of this study was to examine the activity of two quaternary ammonium salts, cetylpyridinum bromide and a newly synthesized quaternary bis ammonium salt, against S. epidermidis biofilm. The average values of killing efficiency for cetylpyridinum bromide ranged from 26.6% to 64.1% for all tested concentrations (0.125 to 8.0 μg×mL⁻¹) and for quaternary bis ammonium salt the percentage of killing efficiency ranged from 59.7% to 88.4% for tested concentrations (from 2.0 to 128.0 μg×mL⁻¹). Both tested compounds significantly affect staphylococcal biofilms, but any of used concentrations caused a total eradication of bacterial biofilm.

Scharakteryzowano szczepy Staphylococcus aureus i Staphylococcus epidermidis, wyizolowane z ran i tkanek głębokich oraz z powierzchni skóry, pobranych w celu poszukiwania obecności genów dla czynników wirulencji. Określono genetyczne zróżnicowanie badanych szczepów uzyskane metodą reakcji PCR ze specyficznymi starterami.

The aim of study was the molecular characteristic of S. aureus and S. epidermidis isolates obtained from skin surface, wounds, deep tissues of hospitalized patients and from skin surface of non-hospitalized patients. Genes encoding virulence factors were examined using PCR reaction and specific primers. Genes encoding adhesins fnbA and cna and gene eta for epidermolytic toxin were mostly present in S. aureus isolates coming from wounds and deep tissues compared to these from skin surface. Gene atlE encoding autolysin of S. epidermidis was detected in all studied isolates, whereas gene icaAB was present in almost all isolates. Comparison of results obtained by PCR and conventional method of the resistance to methicillin estimation showed discrepances suggesting the need for using of both methods in some clinically difficult cases of S. aureus infection.

Zatrucia pokarmowe u ludzi mogą być związane z obecnością enterotoksyn gronkowcowych w żywności. W 2011 r. w krajach Unii Europejskiej stwierdzono wzrost zachorowań wywołanych spożyciem żywności zanieczyszczonej enterotoksynami gronkowcowymi. Najwięcej zatruć pokarmowych wywołanych przez enterotoksyny gronkowcowe związanych było z mieszaną żywnością (40%) oraz produktami piekarskimi (11,4%). Obecnie opisano 21 typów serologicznych enterotoksyn, oznaczonych literami od A doV.

The occurrence of food-borne outbreaks in humans may be associated with a presence of staphylococcal enteroto- xins in food. Most food-borne infections are caused by mixed food (40%).The highest number of food-borne outbreaks, caused by staphylococcal entero- toxins was recorded in the EU in 2011. The second most freguently single food category reported was bakery products (11.4%). Currently, there are described 21 staphylococcal enterotoxins, marked with letters A-V.

Scharakteryzowano 33 szczepy gronkowców koagulazo-ujemnych izolowane z wydzieliny zalegającej w drenach chorych poddanych resekcji anatomicznej miąższu płuca z powodu raka. Najczęściej identyfikowano Staphylococcus epidermidis oraz S. warneri. Większość szczepów wykazywała zdolność do produkcji śluzu oraz właściwości hydrofobowe. Przeważały szczepy oporne na penicylinę i oksacylinę, ale wrażliwe na amoksycylinę z kwasem klawulanowym. Dwa szczepy S. haemolyticus wykazywały metycylinooporność.

Coagulase-negative staphylococci (CNS) represent an important group of etiologie agents of infections associated with plastic biomateriale, e.g. drains. In the present paper 33 strains of CNS were characterized. All of them were isolated from fluid of pleural drains in patients with lung cancer after pulmonary resection under conditions of antimicrobial prophylaxis. The most frequently isolated species were Staphylococcus epidermidis and S. warneri. The majority of CNS strains showed ability to produce slime and possessed hydrophobic properties of cell surface. Strains of CNS resistant to penicillin and oxacillin, but sensitive to amoxicillin/clavulanate were isolated most frequently. Only two methicillin-resistant strains, belonging to S. haemolyticus, were found. The obtained data indicate that CNS strains colonizing pleural drains had potential ability to adhere to smooth surfaces. Most of isolated strains were susceptible to antibiotics and chemioterapeutics routinely used in staphylococcal infections.

Since there are more and more cases of multiresistance among microorganisms, rational use of antibiotics (especially their systemic vs. local application) is of great importance. Here we propose polymeric nanoparticles as locally applied gentamicin delivery system useful in osteomyelitis therapy. Gentamicin sulphate (GS) was encapsulated in the poly(lactide-co-glycolide) (PLGA 85:15) nanoparticles by double emulsification (water/oil/water, W1/O/W2). The nanoparticles were characterized by dynamic light scattering, laser electrophoresis and atomic force microscopy. UV-vis spectroscopy (O-phthaldialdehyde assay, OPA) and Kirby-Bauer tests were used to evaluate drug release and antimicrobial activity, respectively. Physicochemical characterization showed that size, shape and drug solubilization of the nanoparticles mainly depended on GS content and concentration of surface stabilizer (polyvinyl alcohol, PVA). Laser electrophoresis demonstrated negative value of zeta potential of the nanoparticles attributed to PLGA carboxyl end group presence. Drug release studies showed initial burst release followed by prolonged 35-day sustained gentamicin delivery. Agar-diffusion tests performed with pathogens causing osteomyelitis (Staphylococcus aureus and Staphylococcus epidermidis, both reference strains and clinical isolates) showed antibacterial activity of GS loaded nanoparticles (GS-NPs). It can be concluded that GS-NPs are a promising form of biomaterials useful in osteomyelitis therapy.

The aim was to study the activity of lysostaphin in monotherapy or in combination with oxacillin, towards biofilms built by clinical and reference S. aureus and S. epidermidis strains in the wells of microplate, in the chambers of a LabTekII chamber slide or on the polyethylene catheter. MICs of oxacillin and lysostaphin for planktonie bacteria were determined according to the standards of NCCLS. BIC (Biofilm Inhibitory Concentration) was estimated by the MTT assay. The integrity of biofilm treated with antimicrobials was also examined: by staining with FITC and laser scanning fluorescence confocal microscopy and visually by TTC reduction assay. Despite the fact that susceptibility of planktonie cultures of 25 staphylococcal strains to lysostaphin action was various, we have demonstrated the effectiveness of lysostaphin in the treatment of biofilm, built not only on the flat surface of the microplates but also on catheter's surface. The synergistic effect of subBIC lysostaphin+oxacillin was observed forMSSA and MRSA biofilms but not for 1474/01 hVISA strain. Also BICOXA for S. epidermidis RP12 and A4c strains, but not for 6756/99 MRSE biofilms was reduced when lysostaphin was simultaneously used.

This paper presents the degree of contamination of table eggs with bacteria of the genus Staphylococcus, taking into account the source of the eggs. The results of the study indicate a relatively high degree of contamination of table eggs with Staphylococcus bacteria. In 1125 bacteriological tests conducted on whites, yolks and shells of eggs from three sources, staphylococci were found in 514 cases. Thirteen strains were isolated from the whites, but Staphylococcus bacteria were found more often in yolks – 199 strains. The highest percentage of staphylococci were found on the surface of the egg shell – 302 strains. Twelve species of staphylococci were isolated from the eggs tested, including both coagulase-positive strains (Staphylococcus aureus, S. hyicus) and coagulase-negative strains, particularly Staphylococcus lentus, S. warneri, S. epidermidis and S. xylosus. This study determined that regardless of the source of the eggs, egg yolks were more often contaminated with Staphylococcus aureus than with coagulase-negative Stapphylococci. It was also demonstrated that S. aureus dominated in the yolks and on the shells of eggs from the small-scale poultry farm.

The aim of this study was to evaluate the effect of ciprofloxacin (CIP), N-acetylcysteine (NAC) alone and in combination on biofilm production and pre-formed mature biofilms on ureteral stent surfaces. Two strains each of Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Klehseilla pneumoniae, Pseudomonas aeruginosa and Proteus vulgaris, recently isolated from patients undergoing ureteral stent removal and shown to be capable of biofilm production, were used in this study. The inhibitory effects of ciprofloxacin, N-acetylcysteine and ciprofloxacin/N-acetylcysteine combination were determined by static adherence assay. Ciprofloxacin (MIC and 2 MIC) and N-acetylcysteine (2 and 4 mg/ml) inhibited biofilm production by ≥60% in all tested microorganisms. Disruption of pre-formed biofilms of all tested microorganisms was found to be ≥78% in the presence of ciprofloxacin (MIC and 2 MIC) and ≥62% in the presence of N-acetylcysteine (2 and 4 mg/ml), compared to controls. Ciprofloxacin/N-acetylcysteine showed the highest inhibitory effect on biofilm production (94-100%) and the highest disruptive effect on the pre-formed biofilms (86-100%) in comparison to controls. N-acetylcysteine was found to increase the therapeutic efficacy of ciprofloxacin by degrading the extracellular polysaccharide matrix of biofilms. These data are statistically significant. The inhibitory effects of ciprofloxacin and N-acetylcysteine on biofilm production were also verified by scanning electron microscope (SEM). In conclusion, Ciprofloxacin/N-acetylcysteine combinations have the highest inhibitory effect on biofilm production and the highest ability to eradicate pre-formed mature biofilms.
