Badano wpływ dawki cynku i miedzi zastosowanej w nawożeniu na zawartość metali w liściach, łodygach i korzeniach rzepaku. Podstawowe nawożenie wykonano na wiosnę przed siewem roślin. Metale ciężkie Zn i Cu dodano w postaci wodnych roztworów soli Zn(CH3COO)2 o 2 H2O i CuSO4-5H2O, które dokładnie wymieszano z glebą. Dodano do gleby odpowiednio: Zn - 2,0, 10,0, 60,0 mg-kg-1 i Cu - 2,0, 7,0, 20,0 mg-kg-1. Doświadczenie wazonowe przeprowadzono w czasie jednego sezonu wegetacyjnego. Wyraźny wzrost zawartości cynku i miedzi w roślinach zaobserwowano wraz ze wzrastającymi dawkami metali zastosowanymi w nawożeniu. Najwyższe zawartości Zn i Cu stwierdzono w korzeniach roślin niezależnie od zastosowanej dawki nawożenia.

The experimental material was the plant organs of Brassica napus from a pot experiment, carried out during one vegetative season. The effect of increase level of zinc and copper fertilization in the content of metals in leafs, steams, and roots of rape were tested. Basic fertilization was applied once before the growing period, in the spring. Heavy metals (Zn and Cu) were added of the aqueous solution salts: Zn(CH3COO)2- 2 H2O, and CuSO4- 5H2O mixed with the soil. The doses of metals added to the soil were: 2.0, 10.0, 60.0 mg Zn- kg-1 and 2.0, 7.0, 20.0 mg Cu- kg-1. Significant increase of zinc and copper content in plants were observed with increasing doses of metals. The highest content of zinc and copper were observed in roots.

Eight bacterial strains from areas contaminated with trace metals were selected for the experiments: Bacillus sp., Bacteroidetes bacterium, Flavobacterium sp., Luteibactor rhizovicina, Pseudomonas fluorescens, Pseudomonas sp., Serratia entomophila and Variovorax sp. Bacteria were transformed with pET21a expression plasmid encoding Brassica napus L. metallothionein 2 (BnMT2). Transformant colonies were examined for growth on solid culture medium containing trace metal ions (Cd and Pb). It was found that most of the analyzed bacterial strains were resistant to ampicillin, though the antibiotic inhibited their growth. It was demonstrated that growth of bacterial strains on a medium containing Pb ions was not inhibited, while Cd ion content limited or inhibited growth of some bacterial strains. No significant differences were noted in growth between transformed and untransformed bacteria of a given strain on the medium containing Pb ions. The BnMT2 transgene presence abolished adverse effects of Cd ions on Flavobacterium sp., Variovorax sp. and L.rhizovicina. Modified bacterial strains were demonstrated for potential use in treatment of soil contaminated with trace metals, especially Cd ions.

W przeprowadzonych badaniach wykorzystano osiem szczepów bakterii: Bacillus sp., Bacteroidetes bacterium, Flavobacterium sp., Luteibactor rhizovicina, Pseudomonas fluorescens, Pseudomonas sp., Serratia entomophila, Variovorax sp. wyizolowanych z terenów zanieczyszczonych jonami metali śladowych. Bakterie stransformowano plazmidem ekspresyjnym pET21a zawierającym fragment genu kodujący metalotioneinę typu 2 Brassica napus L. (BnMT2). Po uzyskaniu kolonii bakteryjnych (transformantów) sprawdzono ich wzrost na podłożu stałym zawierających jony metali śladowych (Cd i Pb). Stwierdzono, że większość analizowanych szczepów bakterii wykazuje oporność na ampicylinę, chociaż antybiotyk ogranicza ich wzrost. Wykazano, iż wzrost badanych szczepów na podłożu zawierającym jony ołowiu nie był hamowany, natomiast jony kadmu ograniczały lub hamowały wzrost niektórych szczepów bakterii. Nie stwierdzono znacznych różnic we wzroście pomiędzy bakteriami transformowanymi i nietransformowanymi danego szczepu na podłożu zawierającym jony ołowiu. Obecność transgenu BnMT2 niwelowała szkodliwe działanie jonów kadmu w przypadku Flavobacterium sp., Variovorax sp. i L. rhizovicina. Wykazano, że zmodyfikowane szczepy bakterii mogą znaleźć zastosowanie w procesie oczyszczania gleby zawierającej jony metali śladowych, a zwłaszcza kadmu.

Arbuscular mycorrhizal fungi are the most widespread root fungal symbionts, forming associations with the vast majority of plant species. Ectomycorrhizal development alters gene expression in plant symbionts. In this work we examined the effect of arbuscular mycorrhizal fungi spores on the growth and development of Brassica and on the expression of BnMT2 in winter rape. In a pot experiment, rape seedlings growing on different types of sterile and nonsterile soils were inoculated simultaneously with mycorrhizal fungi spores of Acaulospora longula, Glomus geosporum, Glomus mosseae and Scutellospora calospora. As compared with control plants growing in the absence of spores, ten-week-old seedlings of Brassica napus L. in sterile soil inoculated with arbuscular spores had longer shoots and higher fresh biomass of above-ground plant parts. In other types of substrates enriched with mycorrhizal fungi spores, the plants were smaller than non-inoculated plants. The presence of AMF spores stimulated the elongation growth of hypocotyls in both analyzed substrates. BnMT2 expression was highest in plants growing on the sterile substrate. Generally, the presence of mycorrhizal fungi spores appeared to have an adverse effect on the growth of rape plants.

The epicuticular wax covering on plant surface plays important roles in protecting plants against UV radiation. However, the role of epicuticular wax in affecting leaf gas exchange under enhanced ultraviolet-B (UV-B) radiation remains obscure. In the present study, different aged leaves of Brassica napus were used to analyze the responses of crystal structure and chemical constituents of epicuticular wax to UV-B radiation and the effects of such responses on gas exchange indices. Enhanced UV-B radiation significantly decreased the amount of esters in all leaves except the first leaf, amount of secondary alcohols in the second, third and fourth leaves, and amount of primary alcohols in the second and third leaves, while increased the amounts of ketones and aldehydes in the first leaf. Enhanced UV-B level had no significant effect on the amounts of alkanes and total wax in all leaves. Exposure to UV-B radiation resulted in wax fusion on adaxial leaf and stomata opening on abaxial leaf. Fusions of plates and rods on adaxial leaf surface covered most of the stomata, thereby influencing the photosynthesis in the upper mesophyll of leaves. Enhanced UV-B level significantly reduced the net photosynthesis rate (PN) but increased the stomata conductance (gₛ), concentrations of intercellular CO₂ (Cᵢ), and transpiration rate (E) in all leaves. Both UV-B radiation and the wax fusion induced by enhanced UV-B radiation resulted in different stomata status on abaxial and adaxial leaf surface, causing decrease of PN, and increase of gₛ, Cᵢ and E in leaves.

A life table can be used as an important and appropriate tool to evaluate the susceptibility or resistance level of different host plant cultivars to insect pests. In the current study, we determined the suitability or inferiority of five different canola cultivars (Hayula420, Hayula401, Hayula50, Hayula60, RGS) to Nysius cymoides, under laboratory conditions. Data were analysed based on the age-stage, two-sex life table theory. Nysius cymoides which fed on Hayula420 had the longest nymphal period, while those which fed on Hayula50 had the shortest nymphal period. Developmental times (sum of incubation and nymphal periods) was longest for those which fe d on Hayula420 and the shortest for those which fed on Hayula50. The adult pre-oviposition period (APOP), total pre-oviposition period (TPOP), mean fecundity, and adult longevity of adults reared on different canola cultivars showed significant differences. The highest and lowest net reproductive rates (R0) were obtained for those which fed on Hayula420 (11.40 offspring per individual) and Hayula401 (5.47 offspring per individual), respectively. The highest value (0.0395 d–1) for the intrinsic rate of increase (r) was obtained for those which fed on Hayula 60 cultivar and the lowest value (0.0261 d–1) for those which fed on Hayula401 cultivar. The shortest and longest mean generation times (T) were obtained for those which fed on RGS and H401 cultivars, respectively. The lowest and highest values of life expectancy (exj) were obtained for those which fed on RGS and Hayula420 cultivars, respectively. The results showed that Hayula401 and RGS were not susceptible cultivars to N. cymoides. These cultivars showed higher resistance to N. cymoides, while Hayula60, Hayula420, and Hayula50 were found to be suitable cultivars but with lower resistance to N. cymoides, respectively.
