The analysis of UV-VIS spectrum was used for testing changes through 36 days and the impact of external conditions (reduced temperature (R), microwave radiation (M) and normal conditions (NC) for comparisons) on the stability of: BSA and BSA-Fe2+ complexes after different treating procedure. The increase of BSA absorption with increasing concentration of Fe2+ was observed. Increased absorption of BSA, and BSA-Fe2+ with time, related probably to conformational changes present in the protein and changes in electrostatic reactions within the BSA-Fe2+ complexes. Examination of the impact of an external factor on stabilization of the protein and complexes showed a tendency to keep the stability under reduced temperature and a trend with accelerated protein and complex aging under microwave radiation. The observed tendencies to changes under the influence of external factors became more significant in time. These effects were most probably related to changes of the protein structure and time, while the observed trend of accelerating the changes was impacted by the applied external factors.

Przebadano charakterystykę albumin surowicy bydlęcej BSA (bovine serum albumin) poddanych działaniu trzech dienoaldehydów (trans, trans-2,4-heptadienal, trans,trans-2,4-nonadienal i trans,trans-2,4-dekadienal). Utrata grupy aminowej i zwiększenie liczby karbonylowej wskazywały na uszkodzenie łańcucha bocznego BSA. Wyniki elektroforezy przeprowadzonej z wykorzystaniem żelu dodecylosiarczan sodu-poliakryloamid wskazały, że wszystkie stosowane aldehydy doprowadziły do powstania agregatów BSA, przy czym najbardziej znaczący efekt zaobserwowano dla heptadienalu. Zmiany wewnętrznej fluorescencji i hydrofobowości powierzchni BSA wskazywały, że aldehydy zmodyfikowały białkową strukturę BSA. Spowodowały ponadto, że albuminy BSA utworzyły żółtobrązowe addukty i fluorescencyjną lipofuscynę. Addukty heptadienal-BSA wykazywały zwiększoną absorbancję w zakresie UV-Vis 270-280 nm oraz 300-400 nm, podobnie jak oba pozostałe addukty aldehyd-BSA. Zaobserwowano silne korelacje między tworzeniem się grup karbonylowych połączonych z białkami, współczynnikiem retencji zawartości wolnych grup aminowych oraz maksymalną wartością absorpcji UV-Vis a zawartością aldehydów. Przeprowadzono także analizę składu podstawowych produktów utleniania i dokonano oceny wpływu parametrów utleniania na modyfikację BSA. Zaobserwowano większe uszkodzenie BSA podczas działania krótkołańcuchowych aldehydów zastosowanych w większych stężeniach.

Bovine serum albumin (BSA) was incubated with trans, trans-2,4-heptadienal, trans,trans-2,4-nonadienal, and trans, trans-2,4-decadienal (concns. 1-50 mM) at 37°C for 24 h to produce yellowish-brown adducts and fluorescent lipofuscin. Loss of the amino groups and increased carbonyl value of the product were indicative for BSA side chain damage. The Na dodecyl sulfate-polyacrylamide gel electrophoresis showed the aggregation of BSA. Changes in intrinsic fluorescence and surface hydrophobicity value of BSA indicated the modification of protein structure. The heptadienal-BSA adducts exhibited an increased UV-Vis absorbance at 270-280 nm and 300-400 nm, similarly to the 2 other aldehyde-BSA adducts. Strong correlations were obsd. between formation of protein-bound carbonyls, the retention ratio of free amino content, max. UV-Vis absorption value, and concn. of aldehydes. Principal component anal. was also performed. In general, the BSA damage increased with increasing the aldehyde concn. and decreasing their chain length.

This paper presents ultrafiltration results of model BSA (bovine serum albumin) and MB (myoglobin) solutions prepared with or without NaCl addition. The protein concentrations in the solutions were equal to 0.05 g dm 3 for MB and 0.5 g dm 3 for BSA. The ultrafiltration tests were performed using a laboratory scale unit equipped with 90 mm ceramic disc membranes with a filtration area of 5:6-10-3 m2 and cut-off of 50 or 150 kDa. The tests were run under constant process conditions, i.e. a cross flow volume (CFV) of 5 ms-1, transmembrane pressure (TMP) of 0.2 MPa, temperature of 20 ◦C and NaCl concentration of 0 or 10 wt%. The installation worked in a semi-open mode with a continuous permeate discharge and retentate recycle. The performance of the membranes was measured with the permeate volumetric flow rate, JV (m3m 2s 1) while their selectivity was determined by the protein rejection, R. The paper evaluates and discusses the protein rejection mechanisms as well as the influence of the membrane cut-off and sodium chloride concentration in the feed on the flux decline during the ultrafiltration of BSA and MB. Moreover, it provides an analysis of the first fouling phase by applying usual filtration laws.

In membrane treatment of wastewater containing proteins, humic acid influences the rate and extent of fouling. Cations may increase the tendency of foulants to precipitate due to the changes in intermolecular electrostatic interactions. In this paper, we investigated the effect of cations, including ionic charge and size of hydrated ion, on ultrafiltration of the solutions containing the mixture of HA and bovine serum albumin (BSA). All experiments were carried out in an Amicon batch stirred cell using Amicon XM50, XM300, YM100 ultrafiltration membranes with molecular weight cut-offs of 50 kDa, 300 kDa and 100 kDa, respectively, and Sartorius microfiltration membranes with a nominal pore size of 0.2 mm. Humic substances were extracted from a commercial sample of sphagnum peat from Northern Ireland (Bulrush). In the presence of Ba2+ cations, the HA rejection in the solutions containing HA and BSA was higher compared to the solutions containing Na+ cations. Unlike monovalent cations, divalent cations interact specifically with natural organic matter (NOM) and form metal humic complexes. The binding of divalent cations with natural organic matter is responsible for the development of a dense layer on the membrane surface. Metal cations may also act as a bridge between humic acid and protein. These phenomena result in a higher deposition of substances on the membrane surface leading to lower flux and higher rejection in the presence of divalent metal cations. In addition to the charge, the size of cation plays a role in the deposition process. The solutions containing MgCl2×6H2O are characterizing by higher flux and lower rejection compared to the solution containing BaCl2×2H2O at all ultrafiltration membranes. This phenomenon may be attributed to the size of Mg2+and Ba2+ ions.

Larger size or novel structure molecules are always appreciated by all fields of experimental and computational science. Conversely, molecules with smaller size and simple structures are usually ignored with no explanation as to why. However, the vast majority of more diminutive molecules behave as a cornerstone in the synthesis of a bigger structural framework. Subsequently, we planned to uncover the interactions of small molecules towards macromolecules, and successfully presented the binding results of 2-aminopyridine and Isovanillin towards BSA through NMR techniques. STD epitope mapping and also the DOSY results provided evidence that Isovanillin remained closer to the binding cavity of protein. Titration experiments afforded 584 µM (0.584mM) and 487 µM (0.487 mM) dissociation constants for isovanillin and 2-aminopyridine respectively. Furthermore, changes in diffusion coefficient (with and without protein addition in DOSY spectra) were found to be 0.081 log (m2 s−1) and 0.096 log (m2 s−1) points for isovanillin and 2-aminopyridine respectively. Docking studies exhibit that these molecules can tie to site 1 (sub-area IIA) through the pi-pi interaction and hydrogen bonding with Trp213. Our results demonstrated that both compounds could be utilized as part of a transporter in the circulatory system and their extension-inspired compounds may be utilized in new drug design.

In the paper influence of electromagnetic field (200 MHz) on biosensor matrix component was shown. Specially designed test bench containing the scanner 3D with the robot was used for electromagnetic field monitoring. The scans of electromagnetic field distribution, before and behind sample were collected. Conformation change of studied protein was monitored by UV spectra. It indicates that fragmentation and aggregation of studied protein might occur with time. The stabilization effect caused by electromagnetic 200 MHz in protein solution was found. The effect was dose dependent (cumulative effect of electromagnetic field) and the differences in the absorption intensity between the control sample and samples after exposition were more visible with time.

The aim of the article is to present indicators of the social pillar of sustainable development used in databases that come from large nationwide social science surveys conducted consistently over decades. For this purpose, the following research projects were selected: the American General Social Survey (GSS), the British Social Attitudes (BSA), the German General Social Survey (ALLBUS), and the Polish General Social Survey (PGSS). A Human Wellbeing Index (HWI) developed by the World Conservation Union (IUCN) was adopted as a conceptual basis, in its version put forward by Robert Prescott-Allen. The article includes a compilation of indicators which describe selected aspects of the social pillar of sustainable development and which are used in all of the research projects mentioned.

Celem artykułu jest ukazanie wskaźników społecznego filaru zrównoważonego rozwoju, które stosowane są w bazach danych będących efektem prowadzenia dużych, ogólnokrajowych sondaży społecznych, realizowanych od dziesięcioleci. Dla ilustracji wybrano następujące projekty badawcze: amerykański General Social Survey (GSS), brytyjski British Social Attitudes (BSA), niemiecki German General Social Survey (ALLBUS) i polski Polish General Social Survey (PGSS), Za podstawę koncepcyjną przyjęto Human Wellbeing Index (HWI) wypracowany przez The World Conservation Union (IUCN), w wersji zaproponowanej przez Roberta Prescott-Allena. Artykuł zawiera zestawienie wskaźników niektórych aspektów społecznego filaru zrównoważonego rozwoju dostępnych w każdym z wymienionych projektów badawczych.

The results of ultrafiltration tests carried out with model BSA and myoglobin solutions using ceramic 50 and 150 kDa membranes have been presented. Membrane permeability and selectivity were investigated in function of pH, transmembrane pressure (TMP) as well as molecular modelling data for proteins such as size, geometrical parameters and pH of minimal free energy of folding. The study has shown that the permeate flux J(v) depends on TMP, whereas the protein rejection is mainly influenced by pH. The results demonstrated that molecular modelling data are not sufficient to explain the membrane behaviour and the membrane–protein charge interactions and fouling effects must be also considered to explain the rejection mechanisms.

W pracy za pomocą zmian przewodności zbadano zmiany konformacyjne proteiny BSA oraz jej kompleksów (BSA-Au oraz BSA-NAA) pod wpływem promieniowania UV oraz MF aplikowanego frakcyjnie w dawkach (3 × 10 min oraz 6 × 10 min) oraz w sposób ciągły (30 min, 60 min). Wykonano analizę porównawczą wpływu promieniowania UV i mikrofalowego w dawkach ciągłych i frakcyjnych na BSA, BSA-Au, BSA-NAA oraz przeanalizowano czułość na pole elektromagnetyczne z zakresu UV i MF dla badanych substancji. Udowodniono efekt przyspieszonych zmian konformacyjnych pod wpływem pola elektromagnetycznego z zakresu UV i MF. Wykazano wpływ czynników na stabilność BSA i jej kompleksów zależny od: rodzaju czynnika, sposobu jego aplikacji oraz czasu ekspozycji. Najbardziej czuły na pole elektromagnetyczne okazał się kompleks BSA-Au.

In this study, the conformational changes of BSA protein and its complexes (BSA-Au and BSA-NAA) under UV and MF radiation applied in fractional doses (3 × 10 min and 6 × 10 min) and continuously (30 min, 60 min) were investigated by means of conductivity changes. A comparative analysis of the effect of UV and microwave radiation in continuous and fractional doses on BSA, BSA-Au, BSA-NAA was performed, and the sensitivity to UV and MF electromagnetic fields of the tested substances was analysed. The effect of accelerated conformational changes under the influence of UV and MF electromagnetic field was proved. The influence of the factors on the stability of BSA and its complexes was shown to depend on: the type of agent, the method of its application and the exposure time. The most sensitive to the electromagnetic field turned out to be the complex BSA-Au.

Purpose: This study focuses on increasing production of biogas as an alternative energy from biodegradable wastes (BWs) using BSA coated iron oxides nanoparticles, in view of solving waste management at household level. Many attempts have been performed in order to increase biogas production, including thermal pre-treatment of organic waste, but all of them present limited industrial applications. Iron has been shown to enhance anaerobic digestion, but there are severe drawbacks for introducing the metal ion in an anaerobic closed reactor. Design/methodology/approach: Process for the production of biogas from biodegradable material which comprises the steps of: (a) adding the biodegradable material to the Bio- reactor,(b) inoculating the microorganisms in the digester,(c) synthesis iron oxides and BSA powder coated on the particles (d) adding a colloidal solution of surface-modified BSA-iron oxide nanoparticles to the reactor; (e) providing anaerobic conditions; (f) carrying out the anaerobic digestion; and (g) collecting the biogas, wherein the steps (a), (b) and (c) can be carried out in any order. It also comprises the use of BSA-iron oxide nanoparticles capable of supplying Fe ions to the media for biogas production in anaerobic conditions and in the presence of Fe ions in the media.

Wprowadzenie. Tkanka kostna jest tkanką aktywną metabolicznie, która podlega dynamicznym przeciwstawnym procesom kościotworzenia i resorpcji. Coraz większą uwagę zwraca się na znaczenie aktywności fizycznej jako czynnika warunkującego zdrowy układ kostny. Cel badania. Celem badania była ocena zawartości mineralnej (BMC) i gęstości tkanki kostnej (BMD) oraz wskaźników T-Score i Z-Score: całego szkieletu, kręgosłupa lędźwiowego i nasady bliższej kości udowej prawej i lewej młodych osób w zależności od aktywności fizycznej. Materiał i metoda. W badaniu udział wzięło 120 osób w wieku od 20 do 29 lat – (60 kobiet i 60 mężczyzn), w tym po 60 osób (30 kobiet i 30 mężczyzn) w grupie bardziej aktywnych i mniej aktywnych fizycznie, którzy w badaniu zostali określeni jako nieaktywni fizycznie. Oceniane parametry uzyskano przy użyciu densytometru Lunar iDXA (GE Healthcare). Analizie poddane zostały następujące parametry: BMC, BMD, T-Score i Z-Score całego szkieletu, odcinka L1-L4 kręgosłupa lędźwiowego i nasady bliższej kości udowej prawej i lewej, a także BMI i BSA. Wyniki. Uzyskano wyższe wartości ocenianych parametrów w grupie aktywnych fizycznie w porównaniu z grupą nieaktywnych fizycznie. Istotnie statystyczne różnice uzyskano dla: a) BMD i BMC całego szkieletu oraz BMC odcinka L1-L4 kręgosłupa lędźwiowego i nasady bliższej obu kości udowych, porównując obie grupy bez podziału na płeć; b) BMD i BMC oraz T-Score całego szkieletu i Z-Score odcinka L1-L4 kręgosłupa lędźwiowego tylko w grupie kobiet. Analizując korelację między BMI a BMD oraz BSA a BMD całego szkieletu, stwierdzono istotnie statystyczne dodatnie korelacje w obu grupach bez podziału na płeć oraz w grupie mężczyzn. Natomiast istotność statystyczną wykazano dla różnicy współczynników korelacji pomiędzy grupami dla korelacji BMI i BMD total. Wnioski. Uzyskane w prezentowanej pracy wyniki wskazują, że nawet niewielki, regularny wysiłek na poziomie rekreacyjnym wpływa na zwiększenie gęstości kości.

Introduction. Bone tissue is a metabolically active tissue that undergoes dynamic opposing processes of bone formation and resorption. More and more attention is paid to the importance of physical activity as a determinant of a healthy skeletal system. Aim. The aim of the study was to assess the bone mineral content (BMC) and bone tissue density (BMD) as well as the T-Score and Z-Score indicators: of the whole skeleton, lumbar spine and proximal femoral right and left femurs in young people depending on physical activity. Material and methods. 120 people aged 20 to 29 (60 women and 60 men) took part in the study, including 60 people (30 women and 30 men) in the more active group and less physically active people who were described as physically inactive in the study. The evaluated parameters were obtained using a Lunar iDXA densitometer (GE Healthcare). The following parameters were analyzed: BMC, BMD, T-Score and Z-Score of the whole skeleton, the segment L1-L4 of the lumbar spine and the proximal epiphysis of the right femur and left, as well as BMI and BSA. Results. Higher values of the assessed parameters were obtained in the physically active group compared to the group of physically inactive. Significant statistical differences were obtained for: a) BMD and BMC of the whole skeleton and BMC of the L1-L4 segment of the lumbar spine and proximal epiphysis of both femurs comparing the two groups without gender division; b) BMD and BMC as well as T-Score of the whole skeleton and Z-Score of the L1-L4 segment of the lumbar spine only in the group of women. Analyzing the correlation between BMI and BMD as well as BSA and BMD of the whole skeleton, statistically significant positive correlations were found in both groups without division into sex and in the group of men. On the other hand, statistical significance was demonstrated for the difference in correlation coefficients between the groups for the correlation between BMI and BMD total. Conclusions. The results obtained in the presented study indicate that even small, regular exercise at the recreational level increases bone density.

The stability of eight hydroxycinnamic acids (HCAs) during long-term incubation under physiological conditions was studied by UV-VIS absorption spectroscopy and their possibility of binding to a model protein (bovine serum albumin, BSA) under physiological conditions was investigated by tryptophan fluorescence quenching method. The obtained results suggest that the stability of hydroxycinnamic acids is dependent upon its individual structure and duration of incubation. The monosubstituted derivatives (coumaric acids) were stable within the course of long-term incubation, while di- and trisubstituted derivatives decomposed easily. It was found out that all studied compounds changed fluorescence emission spectrum of BSA. The Stern-Volmer analysis was employed in order to explore binding of HCAs to BSA in details. The binding constants (Kb), number of binding sites (n) and the free energy changes (∆G0) were determined. The binding affinity was strongest for rosmarinic acid and ranked in the following order rosmarinic acid > chlorogenic acid > sinapic acid > caffeic acid > ferulic acid > o-coumaric acid > p-coumaric acid > m-coumaric acid. All free energy changes (∆G0) possessed negative sign indicating the spontaneity of HCAs binding to BSA.

Badano trwałość ośmiu kwasów hydrooksycynamonowych, HCAs, podczas inkubacji długoterminowej w warunkach fizjologicznych i możliwość ich wiązania się z modelowym białkiem (albuminą, BSA). Uzyskane wyniki pokazały, że trwałość tych kwasów zależy od ich indywidualnej budowy oraz czasu inkubacji. Wyznaczono stałe trwałości (Kb), liczbę miejsc wiązania (n) oraz zmianę wartości energii Gibbsa (∆G0) wiązania HCAs do BSA, który to proces przebiegał samorzutnie.
