The objective of the study was to perform a comparative analysis of phenotypic and genetic similarity, determination of resistance profiles, detection of toxin-encoding genes and molecular typing of Clostridium perfringens isolates originating from patients with gas gangrene. The study encompassed three patients with a clinical and microbiological diagnosis of gas gangrene who were hospitalized in one of the hospitals of the Kujawsko-Pomorskie province in the same period of time between 8th April 2015 and 20th April 2015. The three C. perfringens isolates studied had identical biochemical profiles. Two isolates had identical resistance patterns, while the third presented a different profile. Using the multiplex PCR method, all isolates showed the presence of cpa gene encoding α-toxin; furthermore, the presence of the cpb2 gene encoding β2-toxin was confirmed in two isolates. Genotyping with the use of pulsed field gel electrophoresis (PFGE) indicated that the isolates originating from the three studied patients represent three genetically different restrictive patterns which correspondedto three different clones – clone A, clone B and clone C. As a result of the study, it is possible to conclude that the studied patients simultaneously hospitalized in a single Department of Orthopedics and Traumatology developed three different endogenous infections.

Bakterie z rodzaju Lactobacillus wytwarzające nadtlenek wodoru dominują w populacjach bakterii zdrowej pochwy. W pracy podjęto próbę określenia, które z gatunków Lactobacillus wykazują taką właściwość, a ponadto wyznaczono stężenia bójcze zarówno czystego chemicznie nadtlenku wodoru jak i supernatantów pochodzących z hodowli Lactobacillus wobec bakterii i grzybów kolonizujących drogi rodne kobiet.

A total of 33 strains of Lactobacillus belonging to 9 species, isolated from vagina, were tested for production of hydrogen peroxide. We observed that the following species: L. delbrueckii, L. acidophilus, L. crispatus, L. johnsonii and L. gasseri dominated over other species in secretion of hydrogen peroxide to the growth medium. Concentration of this substance amounted from 0,05 to 1,06 mM (in case of strong aeration the concentration increased up to 1,8 mM). Moreover, killing properties of the pure hydrogen peroxide exerted toward Escherichia coli and Candida albicans were less prominent than these of the supernatants of cultures of Lactobacillus strains producing H2O2

The aim of the study was a retrospective analysis of the frequency of group B streptococci (Streptococcus agalactaie; GBS) carriage in pregnant women from the region of Krakow, together with an analysis of their drug resistance, carried out between 2008-2012. The study included 3363 pregnant women between 35 and 37 weeks of gestation, studied in accordance with the guidelines of the Polish Gynecological Society (2008). A high percentage of pregnant women who are carriers of group B streptococci was demonstrated. Each year covered by the study, it was in the range of 25-30%, with an average value equal to 28%. The results confirm the need for taking swabs from both the vagina and anus, since 15% of GBS-positive patients showed only rectal carriage. High percentage of isolates resistant to erythromycin was detected, which ranged from 22% to 29%, with an average value equal to 25%, as well as a high proportion of isolates resistant to clindamycin being 17-25%, with an average of 20%. The results indicate the need to standardize the methodology of collecting samples for GBS testing and introduce microbiological diagnostic standards in all gynecological and obstetric centers in Poland, in order to carry out a detailed epidemiological analysis in our country.

Bakterie Lactobacillus delbrueckii, występujące w mikroflorze pochwy wspólnie z gatunkami z grupy Lactobacillus acidophilus, hamują wzrost Candida albicans w czasie 90 minut, zaś bakterie z gatunku Lactobacillus plantarum - po 24 godzinach. Wczesne działanie Lactobacillus można przypisać nadtlenkowi wodoru, zaś późne - innym metabolitom. W warunkach naturalnych bakterie z rodzaju Lactobacillus prawdopodobnie współdziałają ze sobą przeciwko C. albicans, wykorzystując kilka metabolitów.

Growing frequency of therapeutical failures of vulvovaginal candidiasis, resulting from resistance of certain species of Candida to imidazole agents, raises interest in the use of probiotics from Lactobacillus genera as prophylaxis. Unfortunately, little is known about inhibitory mechanisms of Lactobacillus on Candida. The aim of this study was to compare the activity of selected Lactobacillus species, representing the physiological vaginal flora, against Candida as well as investigation whether their inhibitory activity against Candida is related strictly to hydrogen peroxide and lactic acid production. 125 strains from vaginal smears of healthy women were classified by making use of phenotypic and genotypie methods. The majority of strains belonged toL. acidophilus: L. acidophilus sensu stricto, L. crispatus, L. gasseri and L. johnsonii as well as L. fermentům and L. plantarum species. Culture supernatants of selected 25 strains representing the isolated species were examined for their inhibitory activity against the growth of Candida albicans and C. glabrata. The results showed that the strongest and the fastest activity against C. albicans was demonstrated by L. delbrueckii strains, producing the largest quantities of hydrogen peroxide. On the other hand, extended activity, demonstrable after 24 hours, was shown by non-H2O2 producing L. plantarum supernatants. Growth of C. glabrata was not inhibited by any of the examined strains of Lactobacillus. Comparison of activity of live active cultures of Lactobacillus strains and their mixtures with this of pure H2O2 and lactic acid has shown that pure chemical compounds were less active than the cultures. This suggests that mixtures of Lactobacillus strains are in cooperation with each other using many different metabolites.

The study aim was evaluation of the usefulness of several thermostable DNA polymerases in real time PCR conducted in the presence of the heme. Our study had the advantage of testing several different polymerases, one of which proved to be the least sensitive to heme activity. We also found that there is no need of supplementing the reaction mixture with protective substances like BSA. Selection of the appropriate polymerase can increase the efficiency of the PCR reaction which is very important for diagnosis of sepsis and for other analyses performed on DNA template isolated from the blood.

Streptococcus agalactiae (Group B Streptococci, GBS) constitutes a risk factor for infections of the newborns born by colonized mothers. The adherence of GBS to epithelial cells has been proved to be an important factor in the colonization of mucus membranes of both human rectum and vagina. The objective of the study was to assess the adhesion of the selected GBS strains to the human colon adenocarcinoma cell line (HT-29) and human epidermoid vulvo-vaginal cells (A-431) in relation to the capsular polysaccharides and alpha-like protein genes. GBS strains from the human sources belonging to Ia, Ib, II, III and V serotypes possessing different surface alpha-like protein genes such as the alp 2, alp 3, bca, epsilon and rib in the conventional adherence assay were examined. The adherence of GBS strains to the HT-29 cell line was considerably higher than to the A-431 cell line. For GBS serotype Ia and III, a significant difference between the adhesion to the HT-29 and A-431 cell lines was presented. The adhesion of GBS strains to the HT-29 cell line depended on alpha-like protein genes. The most adhesive ones were the GBS strains containing the rib and alp 2 genes. The adherence of GBS strains to the A-431 cell line depended on both their serotype and alpha-like protein genes. Serotype III adhered to the A-431 cells most tightly, particularly the strains containing the rib and alp 2 genes. GBS strains containing the rib gene adhered to the HT-29 and A-431 cell lines more firmly than GBS strains containing other alpha-like protein genes.

Podjęto próbę oceny antybakteryjnego potencjału bakterii z rodzaju Lactobacillus, reprezentujących gatunki najczęściej izolowane z dróg rodnych kobiet, wobec szczepów Streptococcus agalactiae, z uwzględnieniem serotypu oraz miejsca ich izolacji. Badania przeprowadzono metodą hodowli w zawiesinie, oznaczając liczbę drobnoustrojów metodą seryjnych rozcieńczeń. Wykazano dużą zdolność pałeczek kwasu mlekowego do zahamowania wzrostu Streptococcus agalactiae. Wrażliwość badanych izolatów paciorkowców z grupy В (GBS) na oddziaływanie metabolitów Lactobacillus była cechą charakterystyczną danego szczepu, niezależną od jego serotypu, czy pochodzenia. Natomiast, wykazano różnicę istotną statystycznie pomiędzy badanymi gatunkami Lactobacillus w zahamowaniu wzrostu paciorkowców, z których największą aktywność wykazywał gatunek L. plantarum.

Streptococcus agalactiae (Group В streptococci - GBS) is one of the coexistent components of vaginal microflora. Lactic Acid Bacteria (LAB) are the predominant bacteria in the physiological flora of vagina. The production of antagonistic substances like short chain fatty acids, hydrogen peroxide, bacteriocin-like substances protects against multiplication of pathogenic microorganisms. However, during pregnancy there is a disturbance of vaginal microflora and there are optimal conditions for bacterial or candidal vaginitis. Despite sufficient protection of LAB during pregnancy, Streptococcus agalactiae is one of the microorganisms, which can multiply and it is dangerous for newborns. The aim of the study was the assessment of antagonistic activity of chosen species of LAB to GBS strains. Antagonism between LAB and GBS was tested in a mixture of fluid 24 hrs cultures and the results were determined quantitatively by serial dilutions. Susceptibility of GBS strains to LAB activity was analyzed according to serotypes of GBS strains (la, Ib-V) and to place of isolation of GBS strains (there are three groups: GBS strains isolated from vagina of carriers; vaginal GBS strains isolated from women, who had clinical symptoms of vaginitis and GBS strains isolated from blood of children, who were ill with sepsis). The obtained results indicated strong inhibitory activity of Lactobacillus species to GBS strains after 2 hrs duration. Statistically significant dependence was found in the susceptibility of GBS strains to Lactobacillus. L. plantarum has shown the strongest activity against GBS strains. There were no statistically significant relationship found in the susceptibility of GBS to LAB activity between serotypes of GBS and between investigated groups of GBS.
