Three-day-old seedlings (t₀ stage) of Vigna radiata (L.) Wilczek obtained from seeds hydroprimed (H) and hydroprimed with proline (HPro) were examined. H and HPro slightly improved mung bean seed germination and seedlings growth at 5℃. The best growth was observed in the seedlings obtain from HPro5 (5 mM) seeds in comparison with the seedlings obtained from the control-non-primed seeds and H seeds. Exposure of mung bean seedlings grown from non-primed seeds to chilling for 4 days induced chilling injury: membrane lipid peroxidation, decrease in endogenous proline level and inhibition of growth of roots and hypocotyls. The seedlings obtain from HPro seeds grew better during the time of chilling and after rewarming at 25℃. The possible role of HPro in chilling injury limitation is discussed.

Melatonin (MEL) was thought to be only a neurotransmitter found in vertebrates until its detection in other organisms including plants. Although the number of publications on MEL function in plants is expanding, the knowledge of this subject is still insufficient. Among many functions which MEL performs in plants, its role as an antioxidant and a growth promoter is most supported by experimental evidence. This compound is an independent plant growth regulator and it may mediate the activities of other plant growth regulators. Due to its antioxidant properties MEL may also stabilize cell redox status and protect tissues against reactive oxygen and nitrogen species which accumulated under stressful environment. Some researchers propose that MEL could be used to improve the phytoremediation efficiency of plants against different pollutants. In this paper we show that exogenous MEL applied into the seeds could be a good biostimulator improving not only seed germination, seedling/plant growth but also crop production especially under stress conditions. We also believe that this compound can increase food quality (the aspect of functional food) and may improve human health. Since MEL is inexpensive and safe for animals and humans its application as a biostimulator could be a good, feasible and cost-effective method useful in agriculture.

A widespread occurrence of melatonin (MEL) in plant kingdom has been reported. MEL is a highly conserved molecule occurring in evolutionary distant organisms. Its role in plants seems to be similar to that in animals. Although MEL function in plants is not well known, yet a hypothesis can be put forward that it probably functions as a night signal, coordinating responses to diurnal and photoperiodic environmental cues. It has also been suggested that MEL is an independent plant growth regulator, probably its action is analogous to IAA and it may mediate the actions of other plant growth regulators. Due to its antioxidant properties MEL may also stabilize cell red-ox status and protect them against reactive oxygen species (ROS) and other harmful environmental influence.

W pracy porównywano wrażliwość na chłód (5°C) dwóch odmian soi (Glycine max (L.) MERR.): Essor i Aldana. Stwierdzono znaczne zahamowanie wzrostu elongacyjnego korzeni w 5°C. Efekt hamowania wzrostu utrzymywał się również po przeniesieniu roślin do warunków optymalnych 25°C i był tym silniejszy im dłużej rośliny eksponowano na działanie chłodu. Odmiana Essor jest bardziej wrażliwa, reaguje gwałtowniejszym zahamowaniem wzrostu korzeni w porównaniu do odm. Aldana. Pod wpływem niskiej temperatury (5°C) wzrasta aktywność amoniakoliazy L-fenyloalaniny (PAL, EC 4.3.1.5) i gromadzą się rozpuszczalne fenole w korzeniach obu odmian soi, w porównaniu z roślinami kontrolnymi rosnącymi w 25°C. W korzeniach odm. Essor różnica zmian aktywności PAL między korzeniami stresowanymi i kontrolnymi jest znacznie mniejsza niż u odm. Aldana. W korzeniach odm. Aldana w odpowiedzi na stres chłodu szybciej gromadzą się związki fenolowe w porównaniu z odm. Essor. Zmiany w poziomie fenoli mogą częściowo wyjaśnić różnicę w odporności na chłód.

In the presented study sensitivity to chilling (5°C) of two soybean (Glycine max (L.) Merr.) cultivars: Essor and Aldana, was compared. A significant inhibition of root elongation was observed at 5°C. This disadvantageous effect persisted even after seedling transfer to optimal (25°C) conditions, and increased with duration of the chilling treatment. Growth inhibition in Essor seedlings was stronger than in Aldana ones and it means that Essor cultivar was more susceptible to chilling injury at 5°C. PAL activity increased and soluble phenolics accumulated in both varieties of soybean at low temperature in comparison to the control grown at 25°C. The difference in PAL activity between stressed roots and control ones was smaller in cv. Essor than in cv. Aldana. In roots of cv. Aldana grown at chilling, phenolics accumulated faster than in cv. Essor. We suppose that changes in phenolic levels can partially explain difference in tolerance to chilling.

Natural phenolic compounds (phenolic acids, flavonoids, tannins, lignans) present in food of plant origin are in the focus of interest due to their prevalence, properties and biological activity. The aim of the presented work was to investigate antioxidant and antigenotoxic effects of the anthocyanin-rich extract from red cabbage leaves (Brassica oleracea rubrum) on the changes induced by toxic Cu²⁺ concentrations. MeOH extract from red cabbage containing anthocyanin (ATH) and phenolic acid derivatives exhibited strong antioxidant properties. Cu²⁺ decreased mitotic index (MI) and inhibited proliferative activity of Vicia faba root meristematic cells. The morphology of mitotic chromosomes was changed; ‘‘erosion’’ and pulverization might result from Cu²⁺ high-cytotoxicity. Numerous micronuclei, chromatid bridges and lagging/lost chromosomes were found in the meristematic cells of V. faba, which indicate the clastogenic effect of Cu²⁺. The application of the ATH-rich extract lowered the number of disturbances induced by Cu²⁺. The positive role of the ATH-rich red cabbage extract will be discussed.

W doświadczeniach prowadzonych z 3-dniowymi, etiolowanymi siewkami soi (Glycine max (L.) Merr. odmiany Aldana) badano wpływ proliny dodanej do hodowli w stresie niskiej temperatury (5°C) i w warunkach optymalnych termicznie (25°C) na wzrost elongacyjny i poziom rozpuszczalnych fenoli. Stwierdzono, że egzogenna prolina obniża hamujący efekt chłodu na wzrost hipokotyli. Chłód stymulował gromadzenie się proliny w tkankach i w pierwszych dniach działania również akumulację związków fenolowych. Podwyższony poziom proliny powoduje zwiększenie zawartości fenoli rozpuszczalnych w hypokotylach niezależnie od temperatury.

The effect of exogenous proline on growth and soluble phenolics level in hypocotyls of 3-days old, etiolated seedlings of soybean (Glycine max (L.) Merr.) cultivated at optimal (25°C) and low (5°C) temperature were investigated. Chilling (5°C) restrained elongation of hypocotyls, whereas proline treatment reduced this unfavourable effect of low temperature. Chilling (5°C) stimulated proline accumulation in tissue and, in the beginning of experiment phenolic compounds accumulation too. Higher level of proline content caused soluble phenolics accumulation in hypocotyls, regardless of temperature.

The mechanisms enabling plants to tolerance high concentrations of copper in their rhizosphere are still poorly understood. Changes of soluble phenolics, anthocyanin and sinapoyl ester contents in red cabbage seedlings which were grown in presence of Cu[to 2+] were investigated. Lipid peroxidation increased together with Cu[to 2+] concentrations, indicating an acceleration of oxidative process. The concentrations of soluble phenolics, anthocyanin and sinapoyl esters were significantly higher in seedlings grown in presence of lower Cu[to 2+] concentrations and they were in the inverse relation to TBARS content. The present study demonstrates that phenolic compounds possibly act by detoxifying reactive oxygen species or/and reduced their production by Cu[to 2+] binding.

Mechanizmy umożliwiające tolerancję roślin na duże stężenia Cu[do 2+] w ryzosferze są słabo poznane. W niniejszej pracy badano zmiany zawartości rozpuszczalnych fenoli, antocyjanów oraz estrów kwasu synapilowego w siewkach kapusty czerwonej rosnącej w obecności dużych stężeń Cu[do 2+]. Wraz ze wzrostem stężenia Cu[do 2+] wzrastał stopień-utlenienia lipidów błon komórkowych, co wskazuje na stres oksydacyjny. Stężenie rozpuszczalnych fenoli, antocyjanów i pochodnych synapilowych wzrastało w siewkach rosnących w obecności małych stężeń Cu[do 2+] i było odwrotne do zmian w zawartości TBARS. Dane przedstawione w tej pracy sugerują, że związki fenolowe (antocyjany, estry synapilowe) działają jako antyoksydanty i biorą udział w detoksyfikacji reaktywnych form tlenu lub/i ograniczają ich powstawanie przez wiązanie jonów miedzi.

Red cabbage seedlings were grown in presence of copper at different concentrations with L-phenylalanine ammonia-lyase inhibitor, 2-amino-2-indanephosphonic acid (AIP) at concentration 5 M. Copper increased anthocyanins accumulation but together with AIP blocked anthocyanin synthesis and significantly decreased the level of soluble phenolic and synapoil esters, in red cabbage. Lipid peroxidation occurred when the plants were treated with AIP and stressed by carrer. In seedlings treated with 2.5 mM CuSO4 solution rapid and significant SOD and APX activity increase were observed whereas catalase activity diminished. When seedlings were grown in presence of the lower Cu[to 2+] concentrations increase in SOD activity was delayed by 3 days. An increased APX activity did not prevent peroxidation in the tested seedlings. These results may suggest an important role of phenolics in protection of red cabbage seedlings against copper stress.

Siewki kapusty czerwonej rosły w obecności inhibitora amoniakoliazy L-fenyloalaniny, kwasu 2-aminoindan-2-fosfoniowego (AIP) 5 M. W siewkach rosnących w obecności jonów miedzi zwiększała się zawartość antocyjanów, lecz poziom tych związków był zahamowany, gdy siewki rosły wobec Cu[do 2+] z dodatkiem AIP. AIP wraz z miedzią hamowały zawartość estrów kwasu synapilowego i rozpuszczalnych fenoli w siewkach. Miedź z AOP powodowały zwiększenie utleniania błon komórkowych. W siewkach rosnących wobec 2,5 mM CU2+ następował wzrost aktywności SOD i APX, podczas gdy aktywność katalazy obniżała się. Natomiast kiedy siewki rosły wobec mniejszych stężeń Cu[do 2+] wzrost aktywności SOD był opóźniony o 3 dni. Wzrost aktywności APX nie przeszkadzał utlenieniu błon komórkowych siewek rosnących wobec miedzi. Wyniki mogą sugerować ważną rolę antocyjnaów i estrów synapilowych w tolerancji siewek kapusty czerwonej na stres miedzi.

The efficiency of pre-sowing conditioning/priming methods used to apply melatonin into seeds was verified: osmopriming in the case of dicot Cucumis sativus and hydropriming of monocot Zea mays seeds. Both priming techniques were selected experimentally as optimal for the studied plant species. Four different seed variants were compared: control non-treated ones, and seeds conditioned with water or with 50 or 500 lM melatonin water solutions. The HPLC–MS quantitative and qualitative analyses were used to determine the content of melatonin and of its potential metabolites in the seeds during 1 year following the conditioning. The control seeds and those conditioned with water contained small amount of endogenous melatonin in both species. However, the level of this indoleamine increased markedly in cucumber and corn seeds primed with exogenous melatonin and it was always correlated with the concentration of melatonin applied. It was noted that melatonin was metabolized during seed storage by its gradual oxidation, thus it protects dry seeds against oxidative stress, prevents potential injuries and significantly increases seeds quality. Interestingly, in the control and water-primed seeds, seasonal fluctuations of endogenous melatonin concentration were noted and significant increase in this indoleamine in the winter month was observed. This suggests that in seeds endogenous melatonin could play a crucial role in seasonal rhythms independently of environmental conditions.

The influence of a high copper sulphate concentration on growth, Cu accumulation, lipid peroxidation as well as on the contents of total phenolic compounds (PhC) and UV-absorbing compounds (UVAC) in roots of lentil (Lens culinars Medic.) cvs. Krak and Tina was investigated. The plants were subjected to 0.5 mM Cu²⁺ for 3 and 5 days in darkness. Growth inhibition and increased lipid peroxidation in the roots of both cultivars, especially in cv. Tina which accumulated more Cu, were observed. Cu²⁺ treatment caused greater PhC and UVAC accumulation in cv. Krak; however, constitutive levels of these compounds were higher in cv. Tina. The maximum absorption peak of UVAC was determined at 270 nm. HPLC analyses of these compounds revealed the presence of two main derivatives of the soluble (aglycone and esterbound) fraction of the hydroxycinnamic acids, ferulic (FA) and p-coumaric (p-CA) acids and the flavonol, kaempferol (Kam). Greater changes in the content of phenolic acids than of Kam may suggest that the former play a more important role in protecting lentil roots against high Cu²⁺ concentration. Thus, while the lower PhC levels at a higher Cu content in the roots of cv. Tina were probably due to stress, their higher levels in cv. Krak could have been a response to ROS signaling. However, though the high concentration of Cu²⁺ stimulated PhC in cv. Krak, it was not sufficient to counteract the amount of ROS generated by metal presence. These observations may suggest that ROS can serve as a common signal for acclimation to Cu²⁺ stress and cause PhC accumulation in dark-grown roots. The role of PhC in lentil tolerance to Cu²⁺ stress is discussed.
