Antioxidant activity: what do we measure?

Inhibition of oxidation of 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) by free radicals generated by decomposition of 2,2'-azobis(2-amidopropane) (ABAP) by antioxidants and biological material was studied. A correlation was found between the ability of various substances to delay the onset of ABTS oxidation and their rapid reduction of the ABTS+* cation radical, and between the ability to reduce the maximal rate of ABTS oxidation and slow reduction of ABTS+*. The length of the lag period of ABTS oxidation was found to be independent of ABTS concentration. Similar decrease of peroxynitrite-induced ABTS+* formation by antioxidants was observed when the antioxidants were added before and after peroxynitrite. All these findings indicate that the main effect of antioxidants in this system is reduction of ABTS+* and not prevention of its formation. Reduction of oxidation products rather than inhibition of their formation may be the predominant mode of action of antioxidants in various assays of antioxidant activity.

In the article are characterized two network methods (critical path method – CPM and program evaluation and review technique – PERT). On the example of an international furniture company’s product, it presented the exemplification of methods to transport cargos (furniture elements). Moreover, the study showed diagrams for transportation of cargos from individual components’ producers to the final destination – the showroom. Calculations were based on the trans- portation of furniture elements via small commercial vehicles.

Scharakteryzowano dwie metody sieciowe (metoda ścieżki krytycznej – CPM; metoda programowania etapów i rewizja terminów – PERT). Na przykładzie produktu międzynarodowej firmy meblowej dokonano egzemplifikacji przedstawionych metod do transportu ładunków (elementów meblowych). Przedstawiono schematy transportu ładunków od poszczególnych wytwórców podzespołów do miejsca końcowego - salonu sprzedaży. W obliczeniach założono transport elementów meblowych małymi samochodami dostawczymi.

Doxorubicin is co-transported with glutathione by several multidrug resistance proteins (MRPs). In order to check whether weak non-covalent aggregates between doxorubicin and glutathione can be formed, which might be substrates for the transporter, the effect of glutathione on the partition coefficient of doxorubicin was studied. No evidence of an effect of glutathione (at levels up to 20 mM) on the partition coefficient of doxorubicin was found in the pH range of 4.0-7.4. These results indicate that non-covalent doxorubicin-glutathione complexes do not form.

We studied the effect of ischemia and reperfusion on the total antioxidant capacity (TAC) of blood plasma during cardiopulmonary bypass surgery employing the modified St. Thomas Hospital cardioplegic solution. TAC was determined using the FRAP method. TAC decreased during surgery, but no further decrease in TAC was observed during reperfusion, indicating that it is a relatively stable parameter of the antioxidative barrier of the body.
