Nitrite content in potable water samples collected from the Upper Silesia and neighbouring regions was determined by the modified Griess method. Higher nitrite levels were found rather in well water than in pipe water. Nitrite concentrations in water from intermittently and continously operated installations were compared. Nitrite levels may seriously increase in intermittently operated water supply installations; it is not advisable to drink water when the supply installation has been inoperative for some time. However, the nitrite levels determined in our study do not seem dangerous to human health.

Ions of irons, especially ferrous ions may be harmful for living organisms, because they generate reactive oxygen species like O2 •- or •OH. Probability of the risk rises especially in pathological conditions, in which high level of iron is observed. For this reason scientists try to establish new methods that can support organism in eliminating reactive ferrous ions. Nowadays, attention focuses on substances present in plants, especially polyphenols, whose administration prevents oxidative damages in iron overloading. This new approach requires some research on behavior of plant-derived compounds in human organism, within a system containing other biomolecules involved in iron metabolism. The aim of this study has been to investigate the influence of black currant (Ribes nigrum) seed extract, a source of polyphenols, on the activity of ceruloplasmin, an enzyme participating in Fe(II) elimination from blood plasma in human organism. Depletion of Fe(II) caused by ceruloplasmin isolated from healthy blood donors was compared to its decrease in a system containing both ceruloplasmin and the extract. The results have shown that addition of a particular amount of the extract elevates the effectiveness of ceruloplasmin in eliminating Fe(II) from the sample but only under physiological condition (pH 7.4; T 37°C). In a weak acidic solution, addition of the extract does not lead to any change in Fe(II) concentration.

Jony żelaza, szczególnie jony żelazawe Fe(II), ze względu na wysoki potencjał oksydoredukcyjny mają zdolność generowania wolnych rodników, takich jak O2 •- i •OH. W pewnych stanach patologicznych, którym towarzyszy przeładowanie organizmu żelazem, ryzyko pojawienia się wolnych jonów Fe(II) inicjujących wiele procesów wolnorodnikowych jest bardzo prawdopodobne. W tym celu dąży się do opracowania terapii, które w chorobach żelazozależnych wspomogą naturalne mechanizmy usuwania reaktywnych jonów żelaza z organizmu. Ostatnio dużą uwagę poświęca się aktywnym substancjom obecnym w roślinach, szczególnie związkom polifenolowym, ze względu na ich silne właściwości chelatujące. Ten nowy kierunek badań wymaga jednak wnikliwej analizy, która wyjaśni mechanizm współdziałania substancji roślinnych z innymi biomolekułami zaangażowanymi w prawidłowy metabolizm żelaza w organizmie oraz rozstrzygnie o bezpieczeństwie ich stosowania. Celem badań było ustalenie wpływu ekstraktu z pestek porzeczki czarnej (Ribus nigrum), bogatego w związki polifenolowe, na aktywność ludzkiej ceruloplazminy, enzymu uczestniczącego w usuwaniu Fe(II) w organizmie ludzkim. Ubytek Fe(II) obserwowany w obecności ceruloplazminy porównywano z jego ubytkiem w mieszaninie ceruloplazmina-ekstrakt. Stwierdzono, że dodatek ekstraktu podnosi skuteczność ceruloplazminy w eliminowaniu Fe(II) ze środowiska. Istnieje więc przypuszczenie, że związki pochodzenia roślinnego takie jak polifenole mogą wspomagać naturalne mechanizmy eliminowania Fe(II) w organizmie.

The aim of the present study was to estimate the ability of individual phenolic acids to eliminate Fe(II) ions from the solution. Moreover, the influence of phenolic acids on the ferroxidase activity of ceruloplasmin (Cp) isolated from blood plasma of healthy volunteers (CpC) and patients with atherosclerosis obliterans (CpAO) was established in vitro. Phenolic acids demonstrated a ferroxidase-like activity, i.e. the ability to eliminate Fe(II) ions, within the studied concentration range of 2.0-17.0 mol 10-5 dm-3, in the following order of decreasing effectiveness: caffeic acid (CA)>ellagic acid (EA)>chlorogenic acid (ChA)>ferulic acid (FA) ≈ p-coumaric acid (PcA) = sinapic acid (SA). The study showed that the ability of phenolic acids to eliminate Fe(II) ions by oxidation or chelation was related to the structure of the former, to the presence of orto -OH groups, especially. Furthermore, the effect of the molar ratio of phenolic acid to Fe(II) ion was observed. EA and ChA, both containing two orto-OH groups and the highest number of –OH groups (4 and 5, respectively), demonstrated the greatest ability to eliminate Fe(II) ions, especially at the Fe(II) to phenolic acid molar ratio of 6:1. Phenolic acids added to samples with a constant amount of Cp caused decrease in the concentration of Fe(II) ions. Therefore, it may be assumed that the addition of phenolic acids to CpC and CpAO, even in low concentrations, caused a significant decrease in the concentration of Fe(II) ions.

We present the case of a 9-year-old boy with DOOR syndrome recognized in the first year of his life because of a delayed development of speech. The diagnosis was based on characteristic abnormalities, including congenital deafness, nail and bone abnormalities, and mild mental retardation.

Ceruloplasmin (Cp) is the major antioxidant in plasma, a protein that carries about 95% of the total copper. Multiple biochemical activities of ceruloplasmin have been described, including copper transport or oxidation of Fe(II) to Fe(III) for subsequent uptake by transferrin and ferritin. Reduction of the pool of free Fe(II) ions by Cp prevents the generation of free radicals and reactive oxygen species by oxidation of Fe(II), thus inhibiting the Haber-Weiss reaction. The removal of both free Fe(II) and Fe(III) ions from blood plasma by polyphenols enhances the antioxidant system of the living organism. However, the mechanism of interactions between exo- and endogenous antioxidants is still under consideration. The effect of raspberry seed extract (RSE) on the ferroxidase activity of ceruloplasmin isolated from plasma of patients with chronic arterial occlusion of the lower limbs due to atherosclerosis (CpAO) was investigated. Moreover, the effect of RSE on the ferroxidase activity of Cp isolated from healthy volunteers (CpC) was also estimated. The ferroxidase activity of Cp, expressed as ΔFe(II), was determined by spectrophotometry with the use of the Fe(II) – histidine complex and ferrozine as a chromogenic reagent. The addition of RSE to samples with the same amount of both CpAO or CpC in each caused an increase in ΔFe(II). The polyphenol-rich RSE may assist Cp in the fight against free radicals and reactive oxygen species when the disease occurs due to an excessive use or reduced production of endogenous antioxidants.

Oxidative stress is one of several factors which contribute to the development of colorectal carcinogenesis. The aim of the study was an assessment of the activity of antioxidant enzymes in tumour and corresponding normal distal mucosa in a group of patients with colorectal adenocarcinoma. Samples of tumour and corresponding normal mucosa were obtained during a resection of colorectal cancer from 47 patients aged between 26 and 82 years. The average distance of corresponding normal distal mucosa from the tumour was 4.49 cm. Activities of antioxidant enzymes: superoxide dismutase (SOD), glutathione reductase (GR), glutathione peroxidase (GPx), glutathione S-transferase (GST), and catalase (CAT) were measured in tissue homogenates. The patients were grouped according to the tumour stage (Duke's staging), grading, localization, and size of tumour, as well as age and sex. Statistical analysis was performed. The activity of SOD and GPx was considerably increased, while the activity of GST decreased significantly in tumour as compared with normal mucosa. GR activity in colorectal cancer was evidently higher in tumours of proximal location compared with the distal ones. The distance of corresponding normal distal mucosa from the tumour was analyzed and related to all assayed parameters. A decreased GST activity was observed in corresponding normal mucosa more than 5 cm distant from the tumour in patients with CD Duke's stage. The higher activity of superoxide dismutase and glutathione peroxidase in tumour compared to corresponding normal mucosa could indicate higher oxidative stress in colorectal adenocarcinoma cells.
