A simple, rapid, and sensitive method based on UPLC-MS/MS was developed to determine spiraeoside in mouse blood, and was applied to the pharmacokinetics and bioavailability of spiraeoside after mice after intravenous (a dose of 5 mg kg⁻¹) and oral (a dose of 20 mg kg⁻¹) administration. On HSS T3 column set at 40 °C, chromatographic separation was obtained with the mobile phase of acetonitrile and 0.1% formic acid using the gradient elution. Spiraeoside and internal standard (IS) were quantitatively analyzed using multiple reaction monitoring (MRM) mode in electrospray (ESI) positive interface. The MRM mode was monitoring the fragmentation of m/z 465.4→303.1 and m/z 451.3→ 289.2 for spironoside and IS, respectively. The results showed a good linear relationship was in the concentration range of 1–200 ng mL⁻¹ (r > 0.998) and the lower limit of quantification (LLOQ) was 1.0 ng mL⁻¹. The intra- and the inter-day precision (RSD%) of the method was within 14.0%, and the accuracy ranged from 90.0% to 115.0%. The extraction recovery of spriaeoside was better than 63.0%, and the matrix effects were in the range of 86%–98%. It also showed the half-life was short, and the absolute bioavailability was 4.0% in mice. Therefore, the established UPLC-MS/MS method was suitable for the pharmacokinetic and bioavailability study of spiraeoside in mice.

The hemostasis system is designed to ensure the integrity of the body’s internal environment, stop bleeding, and maintain a liquid state of blood in the vascular channel. Modern biological and veterinary science presents highly fragmented and scarce data containing clinical and diagnostic clotting characteristics in different fish species. An essential point emphasising the practical component of such studies is spontaneous thrombus formation in fish farming described in the literature. The present research is devoted to the study of the functional state of plasma hemostasis in some ray-finned commercial fishes: phylogenetically more ancient cartilaginous ganoids - sturgeon Acipenser baerii and hybrid of sterlet A. ruthenus and starred sturgeon A. stellatus, as well as bony fishes - carp Cyprinus carpio and tilapia Oreochromis niloticus. It should be noted that the current study was performed at the aquaculture development center “AquaBioCenter” of VSDFA from 2015 to 2020. Species-specific features of clotting were revealed: activation by common and extrinsic pathways, characterised by thrombin time (TT), prothrombin time (PT), and fibrinogen concentration, is several times faster in cartilaginous ganoids than in both bony fish species; hemostasis with activation of the intrinsic pathway, characterised by activated partial thromboplastin time (APTT), is faster in hybrids and tilapias, in contrast to carps and sturgeons. Content of soluble fibrin monomer complexes (SFMC) in all fish was higher than in dogs and humans but lower than in cattle. The highest amount of SFMC was detected in carps, the lowest - in cartilaginous ganoids.

The aim of the study was to determine the influence of selected nanoparticles, namely diesel exhaust particles, Arizona test dust, silver and gold on the rheology of human blood. The rheological properties of human blood were determined with the use of a modular rheometer, at two various temperatures, namely 36.6°C and 40°C. Experimental results were used to calculate the constants in blood constitutive equations. The considered models were power-law, Casson and Cross ones. The obtained results demonstrate that the presence of different nanoparticles in the blood may have different effect on its apparent viscosity depending on the type of particles and shear rate.

Arc-discharge atomic-emission spectrometry was used for determining trace amounts of metals in human blood and urine. Radiation spectra were recorded on an atomic-emission multichannel spectrometer AEMS. An optical multichannel analyzer (OMA) based on a photodiode array was used as the detector. The spectral resolution was characterized by the range 0.011 nm/diode and a spectral-line half-width no more than 0.033-0.044 nm. The samples were prepared for the measurements by the method of dry mineralization without acids. The obtained ash was mixed with a graphite powder in 3:7 mass ratio. For urine samples, the second variant of buffering a mixture of a graphite powder with potassium chloride (9.2 % of K) was also conducted. For determining the concentration of Pb, Cd, Cu and Zn in blood and the concentration of Co, Mn, Ni and Cu in urine three types of the calibrating mixtures containing, respectively, equal amounts of graphite powder and having predetermined concentrations of matrix elements (K, Na, Mg, Ca, Fe) were prepared. Lyphochek Urine Metals Control, Level 1 and Seronorm Trace Elements Urine was used as a reference material in determining trace amounts of elements in urine. Limits of detection were 0.13; 0.7 and 5.5 μg l-1 for Cd, Zn and Pb in blood and 1.2; 1.4; 1.5 and 1.9 μg l-1 for Ni, Mn, Co and Cu in urine respectively. The results obtained and the simplicity of preparation of samples for analysis, allow the conclusion that arc-discharge atomic-emission spectrometry can be used for determination of trace elements in human biological samples, forensic and clinical toxicology, screening investigations in ecologically adverse regions and workers in unhealthy industries.

This paper presents an analysis of use of ultrasonic standing wave in cell separation from bodily fluids based on the example of erythrocyte separation from plasma. It describes movement of red blood cells in plasma under the influence of the acoustic field (whose forces result from interaction of red blood cells with plasma as the vibrating medium) and under the influence of resistance forces in Stokes’ and Oseen’s approximation. The general properties of solutions of the motion equation are given. The solutions for the parameters of the ultrasonic wave and blood cells which are interesting in terms of practical applications in medical diagnostics are discussed. Time constants of the cell transportation to the regions of stable equilibrium in the field of ultrasonic standing wave are estimated. The formulas which determine the time needed to obtain the assumed concentration increase in plasma in nodes and/or anti-nodes of the standing wave are derived.

Diabetes self-management relies on the blood glucose prediction as it allows taking suitable actions to prevent low or high blood glucose level. In this paper, we propose a deep learning neural network model for blood glucose prediction. The model is a sequential one using a Long- Short-Term Memory (LSTM) layer with two fully connected layers. Several experiments were carried out over data of 10 diabetic patients to decide on the model's parameters in order to identify the best variant of the model. The performance of the proposed model measured in terms of root mean square error (RMSE) was compared with the ones of an existing LSTM model and an autoregressive (AR) model. The results show that our model is significantly more accurate; in fact, our LSTM model outperforms the existing LSTM model for all patients and outperforms the AR model in 9 over 10 patients, besides, the performance differences were assessed by thWilcoxon statistical test. Furthermore, the mean of the RMSE of our model was 12.38 mg/dl while it was 28.84 mg/dl and 50.69 mg/dl for AR and the existing LSTM respectively.

Ze względu na warunki przechowywania poszczególnych składników krwi, ocenia się, że ryzyko przeniesienia bakterii jest znacznie wyższe w przypadku przetaczania KKP niż w przypadku przetaczania KKCz. Obecnie w krwiodawstwie stosuje się szereg procedur, które minimalizują ryzyko zanieczyszczenia bakteriami krwi lub jej składników, zwiększając tym samym bezpieczeństwo biorcy.

Blood and blood components have never been as safe as they are now though bacterial contamination is still a problem, especially as regards the transfusion of platelet concentrates (PCs). Storage conditions specific for each blood component are responsible for the significantly higher risk of bacterial contamination of transfused PCs as compared to red cells. Currently a number of procedures have been developed to minimize the risk of bacterial contamination of blood or blood components, with the ultimate aim of increasing a recipient’s safety.

Streaming velocity depends on intensity and absorption of ultrasound in the media. In some cases, such as ultrasound scattered on blood cells at high frequencies, or the presence of ultrasound contrast agents, scattering affects the streaming speed. The velocities of acoustic streaming in a blood-mimicking starch suspension in water and Bracco BR14 contrast agent were measured. The source of the streaming was a plane 20MHz ultrasonic transducer. Velocity was estimated from the averaged Doppler spectrum. The single particle driving force was calculated as the integral of the momentum density tensor components. For different starch concentrations, the streaming velocity increased from 8.9 to 12.5mm/s. This corresponds to a constant 14% velocity increase for a 1 g/l increase in starch concentration. For BR14, the streaming velocity remained constant at 7.2mm/s and was independent of the microbubbles concentration. The velocity was less than in reference, within 0.5mm/s measurement error. Theoretical calculations showed a 16% increase in streaming velocity for 1 g/l starch concentration rise, very similar to the experimental results. The theory has also shown the ability to reduce the streaming velocity by low-density scatterers, as was experimentally proved using the BR14 contrast agent.

The aim of this work was to use the streaming phenomena to assist clot dissolution in blood vessel. Such treatment is called sonothrombolysis. Acoustic streaming is a steady flow in a fluid driven by the acoustic wave propagating in a lossy medium. It is a non-linear effect and it depends on ultrasound intensity, and sound absorption in the media. The source of ultrasound was a flat piezoceramic disc generating long pulses at 1 MHz frequency and 0.2 W/cm2 ITA acoustical intensity. The streaming was generated in a vessel simulating free space, and next repeated in a multi-well cell culture plate, and in the limited space inside the 8 mm diameter silicone tube positioned perpendicular to the ultrasonic beam. The tube was filled with a mixture of water, glycerol, and starch, so with acoustic properties similar to blood. The streaming velocity was recorded either by the Siemens Acuson Antares ultrasonic scanner operating in the color Doppler mode at 8.9 MHz, or by the custom built 20 MHz pulsed Doppler flowmeter. The results obtained using both systems were very similar. The recorded streaming velocities were 3.2 cm/s, 6.1 cm/s and 0.3 cm/s, respectively. They were an order of magnitude smaller than that calculated theoretically. However, the results obtained confirm existence of streaming, even very close to the source, in the limited space. This effect will be explored in in-vitro experiments of blood clot dissolution within the tube simulating a blood vessel.

Dzięki czynnikom wirulencji takim jak adhezyna YadA czy białka Yops jest zdolna do przeżycia w komórkach eukariotycznych. Posocznice będące następstwem przeniesienia pałeczek Y. enterocolitica podczas przetaczania krwi i/lub jej składników nie są zjawiskiem powszechnym. Wysoka śmiertelność wśród pacjentów z jersiniozą poprzetoczeniową wskazuje na ryzyko zakażenia biorców krwi lub jej składników pobranych od dawcy znajdującego się w okresie bezobjawowej bakteriemii przejściowej.

Yersinia enterocolitica is a facultative intracellular pathogen. Due to virulence factors such as adhesin YadA or proteins Yop Y. enterocolitica is capable to survive inside the eucaryotic cells. Septicemia after contaminated by Y. enterocolitica blood products trasfusion is an uncommon phenomenon, however high mortality rate among the cases is the reason why attention should be paid to the risk of TTI.

Wolne rodniki i antyoksydanty odgrywają istotną rolę w prawie każdej jednostce chorobowej. Dlatego tak istotne jest ich oznaczanie. W literaturze opisanych jest wiele metod oznaczania zarówno wolnych rodników jak i antyoksydantów. Często jednak sumaryczne wielkości, stres oksydacyjny i całkowity potencjał antyoksydacyjny (CPA), dostarczają więcej informacji o badanym układzie niż stężenia poszczególnych antyoksydantów i/lub rodników. Obie te wielkości są ze sobą odwrotnie skorelowane. W pracy dokonano przeglądu zmian CPA krwi osób w różnych stanach chorobowych. Opisano też jakie udziały CPA przypadają na białka, antyoksydanty niskocząsteczkowe na wartości CPA, a także udziału różnych związków w całkowitym CPA.

In this paper the possibility of measuring of the total antioxidant potential (TAP) to examination of the antioxidative properties of blood serum and plasma is discussed. Particular attention was paid to the possibility of an independent TAP examination of the protein and small molecules’ parts of the blood serum. The participation of various compounds in the total TAP of serum is also described.

ICP-MS należy do grupy metod analitycznych umożliwiających szybką analizę wielopierwiastkową nawet niewielkiej ilości próbki, charakteryzuje się niską granicą wykrywalności i dużą liniowością w szerokim zakresie stężeń. Analizy metali w płynach ustrojowych takich jak krew, osocze, mocz są prowadzone od wielu lat, aby dostarczyć informacji na temat toksyczności, narażenia na ryzyko ekspozycji, wpływu odżywiania na stan organizmu, a także jako narzędzie w diagnostyce wielu schorzeń. W pracy przedstawiono kilka przykładów zastosowania tej techniki w próbkach pochodzenia biologicznego.

Inductively coupled plasma mass spectrometry often helps doctors in solving many medical problems. ICP-MS belongs to the group of the analytical methods allowing rapid multielement analysis of a small amount of sample, a sufficiently low limit of detection, the ability to overcome interference associated with a complex matrix, a high linearity to the greatest possible range of measurements. Analysis of metals in body fluids such as blood, plasma, urine are conducted for many years to provide information on the toxicity, the risk of exposure, the impact of nutrition on body condition and as a tool in the diagnosis of many diseases. The paper presents several examples of this technique in samples of biological origin.

In this work, we have tried to contribute to the analysis of phenobarbital in the blood. To do this, we used different analytical techniques such as liquid chromatography (HPLC). The results obtained in the course of our work, we can conclude that: The HPLC method was separate phenobarbital endogenous blood components, and optimizing the conditions of chromatographic separation as the composition of the mobile phase consisting of 20 % acetonitrile + 20 % methanol + 60 % ammonium acetate buffer. The extraction with diethyl ether to pH = 4; The chromatographic column, microbondapack ZORBAX C18. A system of diode-array UV detection at 254 nm.

The studies considered graphene-based biomaterials dedicated for cardiovascular therapy. Reduced graphene oxide flakes were introduced into the porous structure of the polyelectrolyte based coatings. TEM analysis showed the presence of graphene flakes arranged parallel to the substrate surface, firmly connected to the porous coating. Biomaterials were subjected to a comprehensive diagnosis of the biological and material properties. The material behavior was simulated using finite element method. The coatings were deposited using layer by layer method. Mechanical analysis was done using Berkovich indenter geometry. They confirmed theoretical FEA based calculations, it was observed the coating stiffness incensement under the influence of introduced particles of graphene. The endanger of the bacteriology film formation was verified based on the E-coli and Staphylo coccus bacteria. Blood–material interaction was examined in the dynamic flow conditions. Bacteriological analysis showed reduced presence of bacteria after contact with the surface with introduced graphene flakes. Dynamic analyzes on blood showed high activation of the coagulation, strong platelets consumption and a strong immune response. It is caused by sharp edge of the single plane of the graphene flake.

This study presents total mercury concentrations (HgT) in selected tissues and organs (blood, muscles, liver, kidneys, feathers and claws) of the herring gull (Larus argentatus) in order to assess the environmental pollution in the coastal zone of the southern Baltic. Dead, frozen birds were collected in the vicinity of Wladysławowo between December 2009 and March 2010, an exceptionally severe winter — a total of 13 specimens; 6 adults (>4 years old) and 7 immature birds (≤4 years old). Mercury concentrations in all analyzed samples exceeded the detection limit. The lowest average concentration, at a level of 0.4 μg Hg g−1 (d.w.) was observed in the muscles of adult and immature gulls while the highest, at a level of 3.3 μg Hg g−1 (d.w), was determined in the contour of immature gulls. A significant linear correlation was found between mercury concentration in the blood and the concentration in the kidneys and liver, with the coefficients of determination at R2 = 0.87 and 0.86, respectively. This indicates that blood of birds, as a mercury carrier, may reflects the concentration of mercury in tissues and internal organs of birds and, in the case of kidneys and liver, may express long-term exposure to mercury in nourishment.
