An automatic computer system to assist assessment of a human semen quality is described. The hardware of the system consists of PC with frame grabber and CCD camera connected to an optical microscope with bright field optic. The software of the system consists of four modules: (1) acquisition of real time sequences of semen, (2) assessment of mobility o sperm cells, (3) calculation of density, and (4) morphology examination of sperm cells. There are few commercial CASA (Computer Assisted Semen Analysis) systems that measure a quantity of semen. Some of them measure only one factor of semen whereas others recognize sperm cells based on user-defined parameters. The described system calculates all factors required for semen quality assessment and recognizes sperm cells by its shape, structure and size.

The article describes the first module of a computer system for human sperm assessment. Different image acquisition methods were described and optical microscope with bright optic was selected as the most appropriate one. Image enhancement and processing methods were selected and customized to achieve effective sperm cells recognition and tracking. Trajectories of sperm cells and their movement parameters were found, calculated and classified in compliance with WHO standards.

Algorithms for examination of the density and selected parameters of movement of sperm cells have been elaborated and implemented. The conducted research is a part of work on a computer system for semen analysis. The system will allow for an increase in the precision of examination thanks to an exact specification of numerical values of the chosen parameters. An additional advantage of the system is an increased time efficiency of examination. Nowadays, the basic type of examination is an estimated analysis of semen parameters by visual observation of a sample, based on a physician's subjective assessment of an image. Registration of images is impossible in visual analysis.

The system will allow for an increase of the preciseness of examination thanks to the exact specification of numerical values of chosen parameters. Additional advantage of the system is a shorter time of the examination. Nowadays, the basic type of examination is an estimated analysis of parameters of semen done by visual observation of a sample. This type of examination is based on a subjective assessment of an image by a physician. Moreover the registration of images in visual analysis is not possible.

The worked out algorithm determines the density of the semen in the specific volume of ejaculate. The algorithm constitutes the first stage in the computer analysis of the picture. The system will allow for the automatic and objective measurement of the chosen parameters of a sample of semen. On the received results a doctor will be able to estimate the quality of the examined ejaculate. Presently, the basic type of examination is the visual analysis. It requires from a doctor a big experience and there is a risk of serious mistakes: • big subjectivism of the received results (doctor estimates visually the number of sperm cells in the sample) • lack of possibility of repetition of the examination on the same sample by more persons because of lack of possibility of any recording of the picture • lack of standardization of examination – in the cause the patient changes hospital it is necessary to conduct again the investigation. The application of the computer technique will increase the precision of the examination. The advantage of the system will be the possibility of reaching accurate results instead of result based only on the visual method. It will be also possible to record the chosen sequences of pictures what will allow for farther analysis and investigations. Moreover, the computer examination will be performed in shorter time. In the first stage of work on the system we elaborated an application evaluating the density of semen in the given picture.
