First Report on Fusarium brachygibbosum Isolate FIR 16_ITS Isolated from Iraqi Wheat Plant

In this study, the Fusarium Brachygibbosum fungus isolate FIR 16_ITS was isolated for the first time from the infected wheat plants in Iraq, during the winter of 2018, in the AL-Tewatha region – 20 km south east of Baghdad. In order to verify the identity of the species of the F. brachygibbosum isolate FIR 16_ITS with the growth symptoms on the wheat plants, the wheat seeds and tissue plants were obtained and used as a source for the re-isolation of the infection agent. F. brachygibbosum isolate FIR 16_ITS was hardly identified using visual approach. However, a molecular technique is important in verifying the species of F. brachygibbosum isolate FIR 16_ITS. The isolate obtained was used for the extraction of DNA and later used for the molecular identification and phylogenetic analyses based on rDNA-internal transcribed region (ITS) primer sets. The authors successfully obtained the amplified products of the ITS-rDNA region, and a sequence analysis indicated that the Fusarium sp. isolate FIR 16_ITS species screened in this study belonged to the F. brachygibbosum species. Isolation and identification of a mercury resistant strain

A mercury resistant strain 2 was isolated. It was identified as Pseudomonas putida and designated as Pseudomonas putida 2 on the basis of morphological and biochemical analysis in combination with phylogenetic analysis. Strain 2 could tolerate and aerobically grow in the medium containing up to of 50 mgdm 3 Hg(II). The highest Hg(II) removal of strain 2 was 85.2% at an initial concentration of 15 mg-dm3 Hg(II). The highest Hg(II) removal of strain 2 was obseved at 30 °C and pH 7.

Celem obecnej pracy była charakterystyka genetyczna pięciu niemieckich szczepów wirusa RHD (Eisenhuttenstadt, Erfurt, Frankfurt, Rossi, Wika), w oparciu o wyniki analiz molekularnych fragmentu o długości 510 nukleotydów, kodującego N-końcową część białka strukturalnego VP60. Analiza genetyczna na podstawie wygenerowanych drzew filogenetycznego i homologii wskazuje na stosunkowo niewielką zmienność szczepów w obrębię badanej sekwencji, która wynosiła 1-8%. Zaznaczyć jednak należy, że pomiędzy grupą I tworzoną przez szczepy Rossi i Wika), a grupą drugą, w której znajdują się pozostałe analizowane szczepy niemieckie (Eisenhuttenstadt, Erfurt, Frankfurt) widać dystans genetyczny.

The aim of the study was genetic characteristic of five German strains of RHD virus (Eisenhuttenstadt, Erfurt, Frankfurt, Rossi, Wika) on the basis on molecular analysis of a 510 bp fragment, coding N-terminal part of structural VP60. The analysis was conducted to lead to determine the phylogenetic dependence of chosen RHD strains. The analysis of five German strains of RHD virus showed that the changeability of those strains is Low, ranging from l to 8%. Nevertheless there is a need to emphasize, that between the first group consisting of Rossi and Wika and the second group with all others analyzed strains from German (Eisenhuttenstadt, Erfurt, Frankfurt) the genetic distance is visible.

Ostatnim etapem analizy filogenetycznej jest ocena wiarygodności powstałego drzewa. Nie jest to łatwy problem, gdyż nie znamy pełnego modelu probabilistycznego ewolucji. Zaproponowano kilka metod, ale w literaturze jest szeroka dyskusja, co do ich stosowalności oraz interpretacji.

The final step of phylogenetic analysis is the test of the generated tree. This is not a easy task for which there is an obvious methodology because we do not know the full probabilistic model of evolution. A number of methods have been proposed by they is a wide debate concerning the interpretations of the results they produce.
