Pharmaceuticals and personal care products (PPCPs) discharged into environment has several adverse impacts. PPCPs are widely utilised for veterinary as well as cosmetic and personal health reasons. These are members of the expanding class of substances known as Contaminants of Emerging Concern (CECs). Antibiotic resistance in the environment and garbage generated by PPCP endanger life. The World Health Organisation (WHO) now recognises antibiotic resistance as a significant global health problem due to the expected increase in mortality caused by it. In the past ten years, mounting data has led experts to believe that the environment has a significant impact on the development of resistance. For human diseases, the external environment serves as a source of resistance genes. It also serves as a major pathway for the spread of resistant bacteria among various habitats and human populations. Large-scale DNA sequencing methods are employed in this thesis to better comprehend the dangers posed by environmental antibiotic resistance. The quantification of the number is an important step in this process. Metagenomic measurement of the number of antibiotic resistance genes in various contexts is a crucial step in this process. However, it’s also crucial to put this data into a broader context by integrating things like taxonomic information, antibiotic concentrations, and the genomic locations of found resistance genes.

This study aimed to perform a qualitative and a quantitative assessment of the prevalence of genes encoding resistance to beta-lactam, tetracycline, and chloramphenicol antibiotics in samples of DNA isolated from air in a municipal wastewater treatment plant (WWTP) and a municipal waste management plant (WMP). Air samples were collected in the mechanical (MP) and biological (BP) processing units of WWTP and WMP in winter and spring. The samples of air were collected by impingement into PBS solution and subsequently, DNA was isolated. The prevalence of the 16S rRNA gene and ARGs was determined by PCR, and the most abundant ARGs were quantified by qPCR. The highest diversity of the analyzed ARGs was noted in air samples collected in the mechanical processing units of the WWTP (winter) and the WMP (spring). The copy of ARGs varied between treatment units and seasons. ARGs were most abundant in air samples collected in spring in the MP units of both the WWTP and the WMP. The study demonstrated that ARGs are ubiquitous in the air in both WWTPs and WMPs. The presence of ARGs in the air can exert a negative impact on the health of plant employees.

Bacterial infections of newly hatched chicks are the most common cause of their death in the initial period of rearing. These infections are always treated with antibiotics. The aim of the study was to investigate the antimicrobial activity of herbal extracts of chestnut (Castanea sativa) and clove (Syzygium aromaticum) against bacterial infections i.e. Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, Klebsiella pneumonice in comparison to antibiotics. The results of the microbiological analyses showed that the Castanea sativa and Syzygium aromaticum extracts had a slighter antibacterial activity in comparison to antibiotics. The diameter of zone inhibition of the culture’s growth of gram-negative bacteria (i.e. Escherichia coli and Klebsiella pneumoniae) and gram-positive bacteria (i.e. Staphylococcus aureus and Enterococcus faecalis) was 6–13 mm for these extracts in comparison to 15–30 mm for antibiotics. However, some bacterial strains presented full resistance to the selected antibiotics, e.g., wild strains of Enterobacteriaceae to amoxicillin or Staphylococcus aureus and Enterococcus faecalis to florfenicol, colistin, and doxycycline. In the second experiment, the effect of the herbal extract mixture added into drinking water on the growth and mortality of chicken broiler during the first rearing week was investigated. There was found that the use of herbal extracts improved the chickens’ body weight (157.4 g; P ≤ 0.008) and decreased mortality rate (2.4%) compared to the control group (144.1 g and 3.9%, respectively) but not to the group treated with antibiotic (161.5 and 0.6% respectively; P ≤ 0.009). In summary, the use of herbal extracts as a nutritional supplement for poultry seems to have a positive effect on weight gain of young birds, and to some extent reduce mortality in the first week of rearing.

The environment contaminated by antibiotics and heavy metals as a consequence of human activities is of great concern nowadays. Many pieces of research proved that the environment could act as a reservoir of antibiotic resistance determinants allowing them to spread among different bacterial species via the process called horizontal gene transfer. The result is antibiotic resistance even in pathogen microorganisms. Heavy metals act as important factors in this process because of their potential to select antibiotic resistant bacteria thanks to linkage among antibiotic resistance genes and heavy metals resistance genes. Thus, this experiment was conducted to screen the antibiotic tolerance profile of bacteria obtained from heavy metal contaminated environment of mine, dump and the contaminated soil near the entry of mine. Several samples were collected from the only active gold mine in Slovakia in Hodruša – Hámre. The presence of cultivable bacteria was proved via cultivation approaches with subsequent MALDI – TOF MS (Matrix – Assisted Laser Desorption/Ionisation Time of Flight Mass Spectrometry) identification of selected isolates. Representative bacterial isolates were screened for their antibiotic tolerance against chosen antibiotics (ampicillin (AMP), chloramphenicol (CHLOR), tetracycline (TET) and kanamycine (KAN)) with the aim to define their minimal inhibitory concentration (MIC). The cultivable bacteria from studied environments were dominated by Gram-negative protebacteria of Pseudomonas and Rhizobium genera. Among more than 150 isolates the resistance to ampicillin (MIC>100µg/ml – 49% isolates), kanamycine (MIC>100µg/ml - 18% isolates), and chloramphenicol (MIC>20µg/ml – 16% isolates) dominated. The resistance to tetracycline (MIC>20µg/ml) was detected in less than 1% of isolates. Overall counts of antibiotic resistance and multi-resistance were alarmingly high taking in account that industrial environments with no known antibiotic exposure were analysed. Our data indicate that heavy metals contaminated environment could influence the occurrence and the spread of antibiotic resistance. Possibly, metal contaminated environment act as a reservoir of antibiotic resistant bacteria.

Środowisko zanieczyszczone przez antybiotyki i metale ciężkie jako konsekwencja działalności ludzkiej jest obecnie przedmiotem wielu zmartwień. Wiele badań udowodniło, że środowisko może stanowić swoisty zbiornik odporności bakterii na działanie antybiotyków, pozwalając im na swobodne rozprzestrzenianie się wśród różnych bakterii poprzez proces zwany poziomym transferem genów. Wynikiem jest obecność odporności na antybiotyki nawet u mikroorganizmów patogenicznych. Metale ciężkie działają jako ważne czynniki w tym procesie ze względu na swój potencjał wyboru bakterii, która opiera się antybiotykowi ze względu na swego rodzaju połączenie pomiędzy genami opierającymi się antybiotykowi oraz genami opierającymi się metalom ciężkim. Zatem, wykonano eksperyment aby zbadać tolerancję na antybiotyk dla bakterii uzyskanych ze środowiska kopalni, składowiska oraz gleby z pobliża kopalni będących zanieczyszczonymi metalami ciężkimi. Pobrano próbki z jedynej aktywnej kopalni złota w Słowacji, zlokalizowanej w Hodruša – Hámre. Obecność bakterii kultywacyjnych został udowodniony za pomocą badań kultywacji a następnie techniki identyfikacyjnej MALDI – TOF MS (Matrix – Assisted Laser Desorption/Ionisation Time of Flight Spectrometry). Reprezentatywne izolaty bakteryjne zostały zbadane ze względu na ich tolerancję na wybrane antybiotyki (ampicylina (AMP), chloramfenikol (CHLOR), tetracyklina (TET) oraz kanamycyna (KAN) w celu zdefiniowania ich minimalnego stężenia inhibicyjnego (MIC). Bakterie kultywacyjne z badanych środowisk były zdominowane przez Gram-ujemne proteobakterie rodzaju Pseudomonas oraz Rhizobium. Spośród więcej niż 150 izolatów, odporność na ampicylinę (MIC>100 µg/ml- 49% izolatów), kanamycynę (MIC>100 µg/ml – 18% izolatów) oraz chloramfenikol (MIC> 20 µg/ml – 16% izolatów) dominowała. Odporność na tetracyklinę (MIC> 20 µg/ml) został stwierdzony w mniej niż 1% przypadku izolatów. Ogólna liczba odporności na antybiotyki oraz multi-odporności była alarmująco duża, biorąc pod uwagę, że środowiska przemysłowe z nieznanym stopniem wystawienia na antybiotyki była analizowana. Nasze dane wskazały, że środowisko zanieczyszczone metalami ciężkimi może wpływać na obecność i rozwój odporności na antybiotyki. Możliwym jest, że środowisko zanieczyszczone metalami zachowuje się jak zbiornik dla bakterii odpornych na działanie antybiotyków.

Salmonelloses are among the most common animal-borne infections. The most frequent causes of their occurrence are contaminated chicken eggs. For this reason, the control of bacteria from the Salmonella genus living in the cloaca of laying hens can contribute to the reduction of spreading levels of infections with these bacteria. The most popular methods of rearing laying hens in small agricultural farms comprise run and cage keeping. The aim of the performed studies was to determine the impact of rearing and nutritional systems on the occurrence of egg infections with Salmonella spp. bacteria. Detection by PCR method corroborated presence of bacteria from the Salmonella genus on eggs surface from hens kept in litter system. Latex serotyping test confirmed the presence of Salmonella Enteritidis. Salmonella spp. was not detected on eggs surface from hens kept in cage system. Salmonella spp. isolates from the eggs surface of hens reared on litter were characterized by drug resistance to tetracycline. Addition of EM probiotic failed to show reduction in incidence of Salmonella spp. infection.

Antibiotic resistance of bacteria was observed in various aquatic environments including seas, rivers, lakes, coastal areas, surface water and sediments. The increased implementation of antibiotics into these environments through medical therapy, agriculture and animal husbandry has resulted in new selective pressures on natural bacterial aquatic populations. Antibiotic resistance of heterotrophic bacteria isolated from the surface microlayer and subsurface water of freshwater coastal polymictic and low-productive lake was studied. Antibiotic resistance was determined by the single disc diffusion method. The resistance level of bacteria to various antibiotics differed considerably. Bacteria were most resistant to ampicillin, ciprofloxacin, clindamycin, erythromycin and penicillin. Majority of bacterial strains showed resistance to 4.6 out of 18 antibiotics tested. As a rule, neustonic bacteria (antibiotic resistance index, ARI 0.44) were more resistant to the studied antibiotics than planktonic bacteria (ARI 0.32). 70-90% of neustonic bacteria were resistant to ampicillin, clindamycin and erythromycin, 60-70% of planktonic bacteria were resistant to ampicillin, ciprofloxacin and penicillin. Differences between pigmented and non-pigmented bacteria in their resistance to the tested antibiotics were observed. Above 40% of achromogenic bacterial strains were resistant to ampicillin, clindamycin and penicillin. Among bacterial strains characterised by their ability to synthesize carotenoids, more than 30% was resistant to ampicillin and ciprofloxacin. Bacterial resistance level to antibiotics depended on their chemical structure. Bacteria isolated from study lake were most resistant to quinolones and lincosamides while they were most susceptible to tetracyclines and aminoglycosides. Results presented in this paper indicate that antibiotics are a significant selection factor and probably play an important role in regulating the composition of bacterial communities in aquatic ecosystems. Adaptive responses of bacterial communities to several antibiotics observed in the present study may have possible implications for the public health.
