Influence of photodegradation on mutagenic activity of aquatic solution of chlorophenols

The paper presents the effect of UV radiation of 254 nmwavelength on the mutagenic activity of aquatic solutions of 3-chlorophenol (3-CP), 2,4-dichlorophenol (PCP), 2,4,6-trichlorophenol (2,4,6-TCP) and pentachlorophenol (PCP). Mutagenicity studies were carried out on two Salmonella strains: TA98 and TA100, with and without metabolic activation by S9 mix. Stable intermediate products of photodegradation have been identified using the GC/MS method. Chlorophenols in doses below 0.1µg were strongly mutagenic towards both strains: TA98 and TA100. The mutagenic properties of aquatic solutions of chlorophenols for both strains decreased as follows: PCP>=2,4-DCP>2,4,6-TCP>3-CP. Aquatic solutions of three chlorophenols after their photodegradation were non-mutagenic for both bacterial strains, except for 2,4-dichlorophenol. In the case of this compound, the products of its decomposition were more mutagenic for the TA100 strain. No significant metabolic activation by S9 mix was observed which suggests a direct mutagenic impact of chlorophenols on the tested Salmonella strains.

Wprowadzenie do pracy przedstawia problemy związane z pojawieniem się w środowisku naturalnym obcych związków chemicznych, ze szczególnym uwzględnieniem niebezpieczeństwa związków mutagennych dla organizmów żywych. Przedstawiono różne rodzaje mutagenów, ich wpływ na organizmy żywe oraz metody badania aktywności mutagennej i kancerogennej związków chemicznych. Charakterystyka Krakowskich Zakładów Farmaceutycznych 'Polfa' obejmowała naturę chemiczną ścieków oraz punkty ich zrzutów. Celem badań była ocena aktywności mutagennej tych ścieków oraz opracowanie prostego, skutecznego i taniego bakteryjnego testu przydatnego do wykrywania substancji mutagennych w ściekach. W pracy dokonano izolacji mutantów auksotroficznych z kolekcji 69 szczepów bakterii z gatunku Escherichia coli, dokonano ich kalibracji z użyciem mutagenów wzorcowych, a następnie użyto do oznaczania aktywności mutagennej związków chemicznych obecnych w ściekach. Krakowskie Zakłady Farmaceutyczne 'Polfa' emitowały do środowiska związki mutagenne. Uzyskane mutanty bakterii Escherichia coli nadają się do badania mutagennych skażeń ścieków.

This article reviews problems related to chemical pollutants present in the natural environment and focuses on the effect of mutagenic compounds on living organisms. Various types of mutagens are shortly presented and the methods of mutagenic and carcionogenic activity testing are breefly listed. The chemical characteristics of the waste waters and the locations of their releasing from industry were showed for pharmaceutical company 'Polfa' in Kraków, Poland. The aim of the study was evaluation of mutagenic activity of the 'Polfa' waste waters and a proposal of effective, useful and cheap bacterial test to detect mutagenic compounds in waste waters. Auksotrophic mutants were isolated from the collection of 69 Escherichia coli strains and then the calibration of obtained mutants using standard mutagens was performed. After the calibration the strains were used to look for mutagenic activity of chemical compounds present in waste waters. Mutagenic compounds were produced by pharmaceutical company 'Polfa' and released within waste waters to environment. The bacterial mutants of Escherichia coli constructed in our laboratory were useful to evaluate mutagenic compounds in waste waters.
