Biological analysis using a luminometer of surfaces intended for preparing meals

The aim of the study was the microbiological analysis using a luminometer of surfaces intended for food preparation in mass catering establishments. Four production stations were selected for the research (cutting and portioning cold meats, processing vegetables, processing raw meat, and the serving line). In addition, samples were taken from the hands of the meal prep staff at those workstations. The biological contamination was measured using a 3MTM Clean-TraceTM NGi mobile Luminometer with a set of 3M reagents. The hygiene status (RLU) of working surfaces and hands at different kitchen stations was compared. The values of RLU measurements taken from the surface of the hands working on the cured meat cutting and portioning station differed significantly from the values obtained from the surface of the hands working on the food serving station (p < 0.05). A similar relationship was found when comparing the RLU values for the working area of both of these stations.

Celem pracy był instrumentalny pomiar ATP przy użyciu luminometru z powierzchni rąk pracowników zakładu mleczarskiego. Wymazy pobierano z powierzchni obu dłoni przed i po procesie ich mycia i dezynfekcji. Ustalono wysoki stopień korelacji wyników (r=0,97) otrzymanych metodą luminometryczną i metodą tradycyjnych posiewów mikrobiologicznych. Na powierzchni rąk brudnych zmierzono średnio 883 umownych jednostek luminescencyjnych RLU (x/max=3082 RLU) i oznaczono 1,2x10/3 jtk ogólnej liczby drobnoustrojów. Po procesie mycia wartości wyników zmierzonych przy użyciu luminometru zmniejszyły się o 24,2% a oznaczonych tradycyjną metodą mikrobiologiczną o 25,1%. Wyniki wymazu rąk czystych mieściły się w granicach 14-637 RLU.

The aim of the work was an instrumental measurement of ATP from the surface of dairy workers' hands using a luminometer. The swabs were obtained from the surface of both palms before and after the process of their washing and disinfection. A high (r=0.97) degree of correlation was observed between results obtained with the luminometric method and the traditional microbial inoculation method. On the surface of dirty hands, an average of 883 RLU (x/max =3082 RLU) was measured and an overall microbes number of 1,2x10/3 cfu was marked. After the washing process, the result values measured with a luminometer decreased by 24.2% and those marked with the traditional microbiological method decreased by 25.1%. The results of clean hands swab did not exceed 14-637 RLU.
