Suction caissons are often used for the caissons of both offshore oil platforms and offshore wind power projects because of their advantages of simple construction, economical cost, and reusability. In this study, model tests were conducted in sand in order to investigate the effects of the caisson installation method on the penetration depth and the critical suction. Results of the test program showed that the method of changing the frequency of suction during different stages of the process can increase the penetration depth of the caisson. Combining with the deformation of the soil body inside and outside the caisson, the existing method for calculating the critical suction is modified, and the critical suction calculation equation of the discontinuous penetration test is proposed. Based on the test results, the calculation equation of the soil heave height can be more accurate predicted. The analysis results verify that the calculation method and the actual results are in good agreement.

In order to assess the contribution of adenosine triphosphate and its metabolites to the cellular metabolism process in Saccharomyces cerevisiae, it is very important to simultaneously determine the relative concentrations of ATP and its metabolites. In this study, a fast, simple reversed-phase high-performance liquid chromatography with high selectivity was developed to simultaneously measure adenosine triphosphate and its metabolites (adenosine diphosphate, adenosine monophosphate, and cyclic adenosine monophosphate) in yeast. The method was performed under the gradient grogram, and the detection was monitored at 254 nm. Analysis was achieved within 25 min. The four components can be detected with linear response over the concentration range from 1 to 100 mg L−1 with excellent correlation coefficients (r2) > 0.999. The recovery of the four analytes was 92.9%, 90.4%, 99.1%, and 105.1%, respectively. To demonstrate the good analysis of yeast samples, changes in the four adenine nucleotides levels caused by caloric restriction in yeast were determined. It is expected that the current method may contribute to further metabolomics and system biology investigations of yeast.

Based on a Takagi-Sugeno (T-S) fuzzy model and an inverse system method, this paper deals with the problem of actuator fault estimation for a class of nonlinear dynamic systems. Two different estimation strategies are developed. Firstly, T-S fuzzy models are used to describe nonlinear dynamic systems with an actuator fault. Then, a robust sliding mode observer is designed based on a T-S fuzzy model, and an inverse system method is used to estimate the actuator fault. Next, the second fault estimation strategy is developed. Compared with some existing techniques, such as adaptive and sliding mode methods, the one presented in this paper is easier to be implemented in practice. Finally, two numerical examples are given to demonstrate the efficiency of the proposed techniques.
