Zastosowanie lipaz Mucor do syntezy estrów sacharydów

Produced in micro technical scale preparations of mycelium-bound Mucor circinelloides and M. racemosus lipases were used for synthesis of saccharide esters. The immobilized in situ Mucor lipases were additionally entrapped in a composite carrier composed of a mixture of poly(vinyl alcohol), gelatin, sodium alginate and chitozan, which was cross-linked with glutardialdehyde and polyphosphate. pH of lipase micro-environment was regulated in a range from 4 to 8 using buffer solutions. Synthesis of esters of glucose or sucrose and fatty acids was carried out at 50°C in a microreactor equipped with a stirrer and the water activity digital sensor. Either di-n-pentyl ether or its mixtures with petroleum ether were applied as reaction media. It was found that the natural antioxidant, carotene, activated these lipases in organic solvents what could be beneficial for the synthesis of esters of polyunsaturated fatty acids.
Otrzymywane w skali mikrotechnicznej związane z mycelium lipazy Mucor circinelloides i Mucor racemosus zastosowano do syntezy estrów sacharydów. Immobilizowane in situ lipazy dodatkowo unieruchamiano w nośniku kompozytowym będącym mieszaniną PVA, żelatyny, alginianu sodu i chitozanu usieciowanego aldehydem glutarowym i polifosforanem. pH mikrootoczenia lipazy regulowano za pomocą buforu w zakresie pH 4-8. Syntezy estrów glukozy i sacharozy oraz kwasów tłuszczowych prowadzono w temp. 50°C w mikroreaktorze wyposażonym w mieszadło i czujnik aktywności wody. Jako środowisko reakcji stosowano eter di-n-pentylowy oraz jego mieszaniny z eterem naftowym. Stwierdzono, że naturalny antyoksydant karoten aktywuje stosowane lipazy w środowisku organicznym, co może być korzystne podczas syntezy estrów wielonienasyconych kwasów tłuszczowych.
