Obraz otaczającego świata staje się coraz bardziej złożony wraz z upływem lat, postępami nauki i rozwojem technologii. W procesie zrozumienia zjawisk, które zachodzą w naszym środowisku, niezwykle istotną rolę odgrywa chemia analityczna. Jej rola i możliwości rosną dzięki postępowi technologicznemu i rozwojowi instrumentalnemu, a laboratoria przemysłowe i kontrolne zostają wyposażane w aparaturę, która jeszcze do niedawna byłaby obiektem zazdrości wielu ośrodków naukowych. Coraz szybszy rozwój technologii, a wraz z nim technik analitycznych ułatwia uświadamianie społeczeństwa o kosztach, jakie niesie ze sobą rozwój cywilizacyjny. Dzięki najnowszej aparaturze analitycznej o nieosiągalnej do niedawna czułości i selektywności, zdaliśmy sobie sprawę z tego, jak wiele antropogennych substancji przechodzi do niemal wszystkich elementów środowiska. Wykrywa się je w powietrzu, wodzie, glebie i w pożywieniu. Mimo że występują zwykle w stężeniach na poziomie ultraśladów, to wieloletnia ekspozycja na pozostałości hormonów, antybiotyków, leków przeciwdrgawkowych, uspokajających, dioksyn, pochodnych metaloorganicznych i związków z grupy WWA i PCB budzi zrozumiałe zaniepokojenie. Czy coraz wcześniejsze dojrzewanie dziewcząt, hirsutyzm u kobiet, plaga alergii wśród dzieci, zwiększający się odsetek kobiet i mężczyzn cierpiących na bezpłodność mogą mieć związek z wieloletnim narażeniem populacji na działanie wspomnianych wcześniej substancji? Nanogramowe dawki farmaceutyków wchłaniane jednorazowo wraz z wodą i pożywieniem są wielokrotnie mniejsze od dawek terapeutycznych, ale regularne i trwające przez długi okres narażenie ludzi na ich działanie zwiększa zagrożenie. Ponadto konsekwencje tego zjawiska mogą się ujawnić nawet w kolejnych pokoleniach. Nasuwa się mnóstwo pytań na temat skutków dla zdrowia ludzkiego, jakie stwarza obecność pozostałości leków w środowisku. Jak liczna jest populacja ludzi narażonych na nieświadome ich przyjmowanie, w związku z korzystaniem z zasobów środowiska? Jakie dawki leków podawane codziennie stają się niebezpieczne na tyle, żeby w dłuższym okresie czasu uszkadzać komórki mózgu, zaburzać gospodarkę hormonalną i wpływać na zachowanie człowieka? Wszechobecność antybiotyków jest istotną przyczyną oporności wielolekowej. Szkody, jakie powodować mogą związki biologicznie czynne, wchłaniane w czasie liczonym w latach, jest tym trudniej ocenić, że ich negatywne skutki występują ze znacznym opóźnieniem. Wciąż odczuwany jest brak rzetelnych informacji na temat faktycznego stanu zanieczyszczenia środowiska pozostałościami farmaceutycznymi, gdyż nawet szacunkowe określenie zagrożenia z tego powodu było dotychczas trudnym do sprostania wyzwaniem dla analityki. Ponadto skala problemu jest jeszcze nieznana. Tak więc przed analitykami stoi nowe i ambitne wyzwanie: znalezienie wiarygodnej odpowiedzi na pytanie: czy, gdzie i na jakim poziomie występują pozostałości niepożądanych substancji. Zadanie to jest trudne ze względu na różnorodność próbek charakteryzujących się złożonym składem matrycy, dużą liczbą potencjalnych analitów oraz bardzo małym poziomem ich stężeń. Autorka niniejszej rozprawy podjęła próbę znalezienia odpowiedzi na pytanie, jakie farmaceutyki i na jakim poziomie występują w środowisku. Ocena, jakie stwarzają one zagrożenia, jest zadaniem dla specjalistów z innych dziedzin. W pracy przedstawione zostały opracowania metodyczne w zakresie przygotowania próbek i chromatograficznego oznaczania organicznych analitów, ze specjalnym uwzględnieniem związków pochodzenia farmaceutycznego oraz możliwości identyfikacji tych substancji za pomocą techniki HPLC-DAD-MS. Szczególna uwaga została zwrócona na nowe rozwiązania metodyczne dotyczące organicznych związków z próbek ciekłych i stałych. Zaprezentowane rozwiązania dotyczą: - izolacji związków organicznych z wody z użyciem dozymetru pasywnego własnej konstrukcji, - adaptacji techniki ekstrakcji do fazy stałej, do jednoczesnego wzbogacania i oznaczania pozostałości leków, - miniaturyzacji procesu ekstrakcji za pomocą rozpuszczalnika i zastosowania tego rozwiązania do badań zawartości węglowodorów aromatycznych w olejach mineralnych, - izolacji organicznych związków z próbek stałych (gleba oraz pasza zwierzęca), z użyciem klasycznych i nowoczesnych technik ekstrakcyjnych. Opracowane rozwiązania zostały omówione na tle współcześnie dostępnych technik i porównane z alternatywnymi oznaczeniami. Wykazane zostały podstawowe zalety i korzyści stosowania proponowanych rozwiązań analitycznych. Efektem końcowym badań umożliwiających adaptację technik ekstrakcyjnych do izolacji organicznych zanieczyszczeń z próbek stałych i ciekłych był monitoring prowadzony w ramach realizacji grantów badawczych oraz krajowych i międzynarodowych projektów, w których autorka uczestniczyła. Ponadto w prezentowanym opracowaniu opisano możliwości i ograniczenia techniki wysokosprawnej chromatografii cieczowej połączonej z detekcją DAD oraz spektrometrem mas. Nadrzędnym celem drugiej części tego opracowania monograficznego było wykazanie przydatności techniki HPLC-DAD-MS do identyfikacji związków, co zaprezentowano na przykładzie analizy jakościowej substancji obecnych w materiałach roślinnych. W ostatniej części pracy omówiono produkty degradacji wybranych związków organicznych.

The picture of surrounding world becomes increasingly complex, together with years which go by as well as the progress of science and technology development. In the process of understanding the phenomena which occur in our environment, an extremely important role is played by analytical chemistry. Its role and capacity are growing thanks to technological progress and instrumental development. With the help of latest analytical instrumentation of unreachable, until recently, sensitivity and selectivity, we have just realized, how many compounds of anthropogenic origin are able to pollute nearly almost all the elements of the environment. Nowadays analysts detect them in the air, water, soil and food. Despite the fact that their concentrations are usually at ultra trace level, long exposure to residues of hormones, antibiotics, anticonvulsants, sedatives, dioxins, organometallic compounds, PAHs and PCBs raises understandable concern. Is it that ever earlier maturing girls, hirsutism in women, the plague of allergies among children, increased number of women and men suffering from fertility problems may be related to the multi-exposure of the population to the aforementioned substances? Nanogram doses of pharmaceuticals absorbed with water and food are much smaller than therapeutic doses, but prolonged of exposure of humans to their action can have unpredictable effects. In addition, the consequences of the exposure may be observed even in subsequent generations. This begs a lot of questions about the implications for human health posed by the presence of those substances in the environment. How big population of people is exposed, in connection with the use of environmental resources? What dose of drugs administered daily become so dangerous that in the long term damage brain cells, disrupt hormonal balance and affect human behavior? The omnipresence of antibiotics is a significant cause of multidrug resistance. It is hard to estimate the damage that biologically active compounds absorbed during the time counted in years may have, since the negative effects can occur with considerable delay. Still there is no reliable information on the actual state of environmental pollution remnants of drugs, since even an estimate of the risk was difficult to meet challenge for analytics. In addition, the scale of the problem is still unknown. Thus, the analysts are facing new and ambitious challenge: to find a credible answer to the question whether, where and at what level, there are residues of anthropogenic substances. The task is difficult because of the diversity of the samples with a complex composition of the matrix, the large number of potential analytes and a very low level of concentrations. The author of this thesis has attempted to find answers for the questions: what pharmaceuticals and at what levels occur in the environment. The assessment of the threat they pose is a task for professionals from other fields. The paper has been focused on sample preparation and chromatographic determination of organic compounds with especial emphasis on pharmaceuticals and the possibility of identifying those substance with HPLC-DAD-MS technique. Particular attention was paid to the new developments on compounds isolated from liquid and solid samples. Presented achievements concern mainly: - the isolation of organic compounds from water using an in-house constructed passive sampler, - adaptation of solid phase extraction technique for the simultaneous extraction and determination of non-steroidal anti-inflammatory drug, - miniaturization of solvent extraction and its application for screening of mineral oils samples, - extraction of organic compounds from solid samples (soil and animal feed) performed with classical and modern techniques of extraction. The developed solutions have been discussed in the viewpoint of contemporary techniques, and compared with alternative ones. The results showed advantages and benefits of proposed solutions. As a final step, extraction techniques were applied during environmental monitoring performed within national and international projects, in which the author participated. Furthermore, the capabilities and limitations of high-performance liquid chromatography combined with DAD detection and mass spectrometry has been discussed. The predominant objective of the second part of this study was to demonstrate the usefulness of HPLC-DAD-MS to identify unknown compounds. Qualitative analysis of substances present in plant material was given as an example. In the last part of the work degradation of selected organic compounds has been discussed.
