The Role of Hepatotoxins Produced by Cyanobacteria in Brackish Water and Lakes

The toxins of cyanobacteria include among neurotoxins the group of patent hepatotoxins. These cyclic, hepatotoxic peptides cause poisonings with an established syndrom of human and animaI illnesses. Deaths after exposure by ingestion of cyanobacterial cells and toxins from freshwater sources are reported. A new sample preparation method for identification and determination of the hepatotoxic microcystins and nodularin produced by cyanobacteria was developed. The clean-up of toxin containing raw extracts based on high performance size exclusion chromatography (gel permeation chromatography, SEC). The determination of the cyclic peptides was carried out by high performance liquid chromatography (HPLC) with ultraviolet (UV) and diode array detector (DAD) or mass spectrometric (MS) detection. For structure elucidation hydrolysis of the microcystins and nodularin by a fast and effective microwave assisted hydrolysis was applied. After formation of diastereomers by pre-column derivatisation with Marfey's reagent [2,4-Dinitro-5fluorophenyl L-valineamide] the enantioselective determination of the released free amino acids by HPLC using C 18 column with UV- and MS detection is possible.

Toksyny wytwarzane przez cyjanobakterie należą do neurotoksyn z grupy silnych hepatotoksyn. Te cykliczne, hepatotoksyczne peptydy powodują zatruwanie organizmu, wywołując symptomy choroby u ludzi i zwierząt. Odnotowano przypadki śmierci po kontakcie z komórkami cyjanobakterii i wytwarzanych przez nie toksynami, znajdującymi się w wodach słodkich. Rozpoczęto badania nad znalezieniem nowej metody przygotowywania próbek w celu identyfikacji i oznaczania mikrocystyn i nodularyn wytwarzanych przez cyjanobakterie. Toksyny zawarte w surowym ekstrakcie są izolowane za pomocą wysokosprawnej chromatografii żelowej (SEC - Size Exclusion Chromatography). Oznaczenie tych cyklicznych peptydów wykonano za pomocą wysokosprawnej chromatografii cieczowej (HPLC) ze światłem UV i detektorem w postaci panelu diod (DAD- Diode Array Detector) lub spektrometru masowego (MS). Do określenia struktury mikrocystyn i nodularyn stosowano szybką i efektywną hydrolizę wspomaganą mikrofalami. Możliwe jest również użycie kolumny C 18 z detekcją MS w celu enancjoselektywnego oznaczania wolnych aminokwasów opuszczających kolumnę (HPLC), po wytworzeniu diastereomerów zderywatyzowanym odczynnikiem Marfey'a (2,4-dinitro-5-fluorofenylo/L-wanilinoamid).
