The aim of this work was the production and characterisation of polyclonal antibodies against different regions of the membrane protein ApoER2 as tools for immunohistochemical localisation of ApoER2 during brain development in mice. To this end, we immunised different rabbits with an MBP-fusion protein of ApoER2 and four different keyhole limpet hemocyanin (KLH) coupled peptides. As the antibodies should detect the native receptor protein, we tested the confirmation of the recombinant protein with an ELISA assay prior to immunisation. The obtained antibodies were characterised by ELISA immunocytochemistry of transfered HeLa cells, and finally used for the immuno-histochemical detection of ApoER2 in brain and testis of mice. We conclude that all antibodies except the one against the intracellular NPXY motif, which had a low affinity, are suitable for ELISA and immunohistochemistry applications.
