RPTLC and RPHPLC analysis of aliphatic alcohols as the DANS-Cl derivatives

A homologous series of C4-C12 aliphatic alcohols has been deriva-tized with DANS-Cl, to introduce a fluorophoric group to the molecule, to enable TLC and/or HPLC analysis (with UV detection) of the compounds. The progress of the reaction under different sets of conditions was monitored by RPTLC. The chromatographic system was selected after analysis of the retention of the compounds obtained, in terms of the relationship between RM and the concentration, C (%), of organic modifier in the aqueous mobile phase. HPLC on C18 with isocratic elution was performed with 60% acetonitrile in water as mobile phase. The analysis time was approximately halved by use of gradient elution (linear gradient, 60-100% acetonitrile in water in 20 min).
