Spectrophotometric and chromatographic characterization of Salmonella typhi polysaccharide - correlation between two chromatographic methods

Salmonella typhi capsular polysaccharide (Polysaccharide Vi) has been screened chromatographically and the feasibility of using UV absorption for detection of the polysaccharide has been investigated. Correlation was obtained between the classical World Health Organization (WHO) chromatographic method, on a low-pressure column, and a high-performance chromatographic method. The results showed the possibility of using UV for quantification of the polysaccharide. Tests conducted on two columns showed the molecular weight to be 2230000 Da. The correlation coefficient for the methods was 97.4% and there were no significant differences between them (P = 0.4789).
