W pracy przedstawiono wpływ obróbki plazmowej pod ciśnieniem atmosferycznym na wino bez dodatków siarczynów z użyciem powietrza i azotu jako gazu roboczego. Użycie miniaturowego reaktora ze ślizgającym się łukiem elektrycznym pozwoliło na znaczne ograniczenie tlenowych mikroorganizmów mezofilnych, bez zmniejszenia pożądanej ilości antocyjanów.

The paper presents the effect of plasma treatment at atmospheric pressure on wine without sulphite additions using air and nitrogen as the working gas. The use of a miniature sliding arc reactor allowed for a significant reduction in aerobic mesophilic microorganisms, without reducing the desired amount of anthocyanins.

W pracy opisany został problem związany z wysokimi nakładami koniecznymi do prowadzenia badań medyczno- -biologicznych. Zaproponowano możliwość zastosowania niestandardowych rozwiązań poprawiających efektywność kosztową prowadzenia eksperymentów in vitro metodą pomiaru impedancji elektrycznej. Opisano opracowane matryce z elektrodami wykonanych z różnych materiałów do pomiaru impedancji elektrycznej komórek in vitro. Pomiary przeprowadzono wykorzystując komórki mysich fibroblastów L929 z użyciem płytek z elektrodami z Cu, Ti, NiCr i Ni. Posłużyły one do oceny wpływu danego materiału na hodowlę komórek i próbę określenia możliwości ich zastosowania w eksperymentach medyczno-biologicznych.

The paper describes the problem related to the high costs necessary to conduct medical and biological research. The possibility of using non-standard solutions improving the cost-effectiveness of conducting in vitro experiments using the electrical impedance measurement method was proposed. The developed matrices with electrodes made of various materials for the in vitro measurement of cell electrical impedance are described. Measurements were performed using mouse L929 fibroblast cells using plates with Cu, Ti, NiCr and Ni electrodes. They were used to assess the effect of a given material on cell culture and to attempt to determine the possibility of their use in medical and biological experiments.

There is a growing trend in the engineering of biomaterials, focusing on surface modifications of biomaterials to improve their mechanical strength, corrosion resistance, and biological properties. Cold plasma treatment may improve biological properties of biomaterials for biomedical applications by enhancing their integration with host tissue. This study investigated the influence of different cold plasma treatments on the surface properties of a polysaccharides- -based biomaterial to improve cell adhesion to its surface. The samples were subjected to plasma treatment using three different reactors operating at atmospheric pressure: gliding arc discharge (GAD) reactor, dielectric barrier discharge (DBD) plasma jet, and DBD surface reactor. Next, surface chemistry of the biomaterial after plasma treatment was determined by ATR-FTIR analysis. Furthermore, a cell adhesion assay on the samples was carried out using normal human skin fibroblasts (BJ cell line). The attenuated total reflection Fourier transform infrared analysis (ATR- -FTIR) showed that new potential functional groups could be formed on the material surface after plasma treatment. However, plasma treatment of the samples did not enhance cell adhesion to the surface of the polysaccharides-based biomaterial. Thus, the obtained results indicate that plasma treatment using GAD reactor, DBD plasma jet, and DBD surface reactor was not effective for surface modification and cell responses.

Currently, various methods are used to assess the biocompatibility of materials. After an in-depth and detailed review of the literature, the method used in the research was selected. As part of the experiments, a method based on the analysis of the values of electrical parameters of cell cultures measured in the presence of electrodes was used. The electrode is a structure made of a thin layer of metallization. It measures the change in resistance, impedance and capacity of a mixture of cells and the substance in which they are grown. The plate containing the electrode assembly is called the measurement matrix. Currently, commercially used test matrices are made of gold or platinum. However, their high price means that large-scale research is significantly limited. In order to increase the access to the widespread use of this method, it was decided that it was necessary to use cheaper materials, reducing the necessary costs of conducting experiments. Considering this, an attempt was made to use a different conductive material to build matrices compatible with the ECIS® Z-Theta measurement system. Their use would enable in vitro research on living cells. In the presented work, titanium was used as a material that may turn out to be an alternative to the materials currently used. Its application to the production of matrices will allow to study the influence of this metal on the behavior of cells.

Obecnie, do oceny biokompatybilności materiałów wykorzystywane są różne metody. Po dogłębnym i szczegółowym przeglądzie literatury wybrano metodę, którą wykorzystano podczas prac badawczych. W ramach przeprowadzonych eksperymentów wykorzystano metodę bazującą na analizie wartości parametrów elektrycznych kultur komórkowych, zmierzonych w obecności elektrod. Elektroda jest strukturą wykonaną w cienkiej warstwie metalizacji. Służy do pomiaru zmiany wartości rezystancji, impedancji oraz pojemności mieszaniny złożonej z komórek i substancji, w której są one hodowane. Płytka zawierająca zestaw elektrod nazywana jest matrycą pomiarową. Aktualnie, komercyjnie stosowane matryce testowe wykonane są ze złota lub platyny. Ich wysoka cena powoduje jednak, że prowadzenie badań na szeroką skalę jest znacząco ograniczone. Aby zwiększyć dostęp do powszechnego korzystania z tej metody zdecydowano, że koniecznym jest stosowanie tańszych materiałów, redukując niezbędne koszty prowadzenia eksperymentów. Zważywszy na to, podjęto próbę zastosowania innego materiału przewodzącego do budowy matryc kompatybilnych z systemem pomiarowym ECIS® Z-Theta. Ich użycie umożliwiłoby prowadzenie badań nad komórkami żywymi in vitro. W przedstawionej pracy jako materiał, który może okazać się alternatywą dla materiałów wykorzystywanych obecnie wykorzystano tytan. Jego zastosowanie do wytworzenia matryc pozwoli zbadać wpływ tego metalu na zachowanie komórek

The work is devoted to the technology of biocompatible substrates with nickel electrodes for in vitro impedance cell culture studies. The legitimacy of this subject was tested by conducting measurements using a system based on the Electric Cell-Substrate Impedance Sensor method. A device for cell bioimpedance testing, made in thin-film technology, has been described. Parameters and applications of the material used for construction, which is commonly used nickel, are discussed. The results of preliminary studies on melanoma cancer cells from the A375 cell line were presented, during which the already used measurement matrices were used. An analysis of the observed changes and obtained results was carried out.

Praca poświęcona jest technologii biokompatybilnych podłoży z niklowymi elektrodami do badań impedancji hodowli komórek in vitro. Zasadność podjęcia tej tematyki przetestowano przeprowadzając pomiary przy użyciu systemu opartego na metodzie ECIS (ang. Electric CellSubstrate Impedance Sensor). Opisano przyrząd służący do badań bioimpedancji komórek, wykonany w technologii cienkowarstwowej. Omówiono parametry i zastosowania wykorzystanego do budowy urządzania materiału, którym jest powszechnie stosowany nikiel. Przedstawiono wyniki wstępnych badań nad komórkami nowotworowymi czerniaka z linii komórkowej A 375, do których użyto wykonane matryce pomiarowe. Dokonano analizy zaobserwowanych zmian i otrzymanych rezultatów.

The aim of the study was to present a method for assessing the condition of cell culture by measuring the impedance of cells cultured in the presence of nickel. For this purpose, an impedance measurement technique using nickel comb capacitors was used. The capacitor electrodes were made using a thin film magnetron sputtering. In the experimental part, the culture of cells of mouse fibroblasts on the prepared substrate was performed. The cell culture lasted 43 hours and showed that the presented technique allows it to be used to analyze the effect of nickel on cells.

Celem pracy było przedstawienie metody oceny stanu hodowli komórkowej poprzez pomiar impedancji komórek hodowanych w obecności niklu. W tym celu zastosowano technikę pomiaru impedancji z wykorzystaniem niklowych kondensatorów grzebieniowych. Cienkowarstwowe elektrody kondensatora wykonano metodą rozpylania magnetronowego. W części eksperymentalnej przeprowadzono hodowlę komórek mysich fibroblastów na przygotowanym podłożu. Hodowla komórkowa trwała 43 godziny i wykazała, że przedstawiona technika mogłaby być zastosowana do analizy wpływu niklu na komórki.

This article deals in the constantly developing branch of microelectronic devices used in various fields of medicine, i.e. diagnostics and treatment of previously incurable human diseases. A method for assessing and monitoring the vital functions of living cells by measuring cellular impedance in real-time using the ECIS® system and a commercial culture substrate is presented. The goal was to develop a substrate significantly less expensive than a commercial substrate that would be suitable for multiple uses and compatible with the ECIS® measurement station. Moreover, thanks to the use of a material with electrochemical properties other than the biocompatible material (gold or platinum) it is possible to observe the cells behavior with regard to the toxic agent. For this purpose, a culture substrate with nickel comb capacitors was used. To make the electrodes, a thin metal layer was sputtered on polycarbonate plates in the magnetron sputtering process. Prior to the next stages, technological masks were designed so as to fit in the ECIS® measuring station. Subsequently, the microelectronic processes of photolithography and etching the metal layer were performed. Finally, the wells were glued onto the culture medium with a biocompatible adhesive. The completed substrates were transferred to the Department of Human Physiology, Medical University of Lublin, for the culture test on A-375 human melanoma cells. The results of the experiment determined the usefulness of the device for monitoring cell culture vital functions by means of impedance measurement.
